Phosphoproteomic analysis of control and Ste20-like kinase (Slk) knockdown cortical neurons
Ontology highlight
ABSTRACT: The Ste20-like kinase (Slk) is a serine/threonine kinase that regulates cytoskeletal organization, apoptosis, and cell polarity in non-neuronal cells. In neurons, Slk guides dendrite morphogenesis and inhibitory synapse stability, thereby controlling excitation-inhibition balance. We compared the phospho-signaling of control and Slk knockdown cortical neurons using quantitative phosphoproteomics. Tandem mass tagging (TMT), phosphopeptide enrichment, fractionation, and Orbitrap mass spectrometry were used to identify Slk-dependent phosphorylation sites in neurons.
