Selective degradation of platelet BTK by the PROTAC NX-5948

ABSTRACT: This study presents a tandem mass tag (TMT)–based quantitative proteomic analysis of human platelets treated with the BTK degraders NX-2127 or NX-5948, or vehicle control (DMSO). Washed platelet lysates from three independent biological replicates per condition were digested with trypsin, labelled with TMT reagents, pooled, fractionated by high-pH reversed-phase chromatography, and analysed by nano-LC–MS/MS using an SPS-MS3 workflow on an Orbitrap Fusion Lumos mass spectrometer. The dataset was generated to assess the selectivity and specificity of BTK degradation in human platelets and to evaluate global proteomic changes associated with targeted BTK degradation.
