Proteomic profiling of the parasitic nematode Ascaridia galli

ABSTRACT: Ascaridia galli (A. galli) is one of the most common intestinal nematodes of domesticated chickens and the cause of avian ascaridiasis, a globally important disease in poultry. Its genome is sparsely sequenced and its proteome is uncharacterized. We her investigate the proteome composition in larvae and in female and male worms of A. Galli, querying the LC-MS/MS data against a hybrid database of two related nematodes; Ascaris suum and Anisakis simplex. We compare the proteome of larvae, female and male worms.

TMT-based quantitative proteomics to study CHN-1/HSP-1 complex substrate turnover in C. elegans

Project description:The wild type (WT) and HSP-1 EEYD (worms generating endogenously mutated HSP-1) C. elegans were cultured under normal conditions (20 degrees) or exposed to 30 degrees for 16 hours as L4 larvae (referred to as heat stress/HS). In WT worms undergoing HS, the activity of CHN-1 ubiquitin ligase is anticipated to be inhibited by its interaction with wild-type HSP-1. In contrast, HSP-1 EEYD is expected not to inhibit CHN-1 in mutant worms, consequently leading to the degradation of a specific pool of proteins. This dataset unveils the identities of these proteins.

Characterization of Heterorhabditis bacteriophora Response to Insect-Derived and Non-Biological Stimuli: Insights into Nematode Recovery and Released Proteins

Project description:Entomopathogenic nematodes (EPNs) are gaining significant popularity as biocontrol agents due to their ability to rapidly kill insect hosts. These nematodes produce bioactive molecules known as excreted/secreted products (ESPs) that help them overcome host immune defenses. Understanding the mechanisms that trigger ESP release and their composition is crucial for unraveling the factors underlying nematode pathogenicity and enhancing their effectiveness as biocontrol agents. Leveraging the insect parasitic nematode, Heterorhabditis bacteriophora, we describe an in vitro study focused on the recovery process of infective juveniles upon contact with selected activation materials. We evaluated three host-derived materials from Galleria mellonella larvae: live larvae, frozen larvae, and heat-inactivated homogenates; and two non-insect-derived materials: filtered water and phosphate-buffered saline. While none of the tested materials resulted in 100% IJ recovery, all induced ESP release. The ESPs were collected and analyzed using liquid chromatography-tandem mass spectrometry, resulting in the identification of 372 proteins. The highest number of proteins, approximately 200, were detected in response to heat-inactivated homogenate and phosphate-buffered saline treatment. Among the identified ESPs were proteins involved in immunomodulation and protease inhibition, both crucial for overcoming host defenses. Our findings highlight the complexity of nematode-host interactions and suggest avenues for improving biocontrol strategies through a deeper understanding of nematode activation and the released spectrum of ESPs.

Brugia malayi glycophosphatidylinositol-anchored proteome

Project description:A proteomics strategy was used to identify putative GPI-APs from adult B. malayi. Three different sample types were prepared for analysis. Firstly, intact adult worms were treated with PI-PLC to enzymatically release and solubilize the protein away from the lipid moiety. A mock-treatment with no PI-PLC was performed as a negative control. The samples derived from treatment of the intact worms have been named “Surface” but it should be noted that the proteins could originate from any exposed surface like the mouth, vagina, or rectum of the worm. Secondly, a membrane fraction of B. malayi adult female worms was prepared by ultracentrifugation of a total lysate in a sucrose buffer to separate membrane proteins from soluble proteins. This membrane fraction was also treated with PI-PLC or mock-treated without PI-PLC as a negative control. Lastly, a GPI-AP enriched sample was prepared by performing a series of organic solvent partitions to extract GPI-APs from a membrane fraction. This sample was not treated with PI-PLC. Proteins in all three samples types were digested with trypsin and the resulting peptides analyzed by LC-MS/MS.

Comparative proteome analysis of the tegument of male and female adult Schistosoma mansoni

Project description:We present a comparative analysis of sex-specific tegument proteins of paired or virgin Schistosoma mansoni. By apply a new and highly sensitive workflow, detection of even low abundance proteins from the worm was achieved Therefore, a streptavidin-biotin affinity purification technique in combination with single pot solid-phase enhanced sample preparation was established for subsequent LC-MS/MS analysis. We were able to identify 1519 tegument proteins for male and female as to virgin and paired worms. Sex-specific proteins were screened, present in both virgin and paired adult schistosomes to reduce the impact of mating. Bioinformatic analysis revealed an involvement of female-specific tegument proteins in signaling pathways of cellular processes and antioxidant mechanisms. Male-specific proteins were found to be enriched in processes linked to phosphorylation and signal transduction. This suggests a task sharing between the sexes that might be necessary for survival in the host. Our datasets provide a basis for further studies to understand and ultimately decipher the strategies of the two worm sexes to evade the immune system.

Tetracycline-treated Litomosoides sigmodontis vs. untreated control Litomosoides sigmodontis

Project description:The filarial nematodes Brugia malayi, Wuchereria bancrofti and Onchocerca volvulus cause elephantiasis, dermatitis and blindness, resulting in severe morbidity in developing countries. 1.3 billion people are at risk of infection. Targeting the essential Wolbachia endobacteria of filarial nematodes with doxycycline has proven to be an effective therapy, resulting in a block in embryogenesis and worm development, and macrofilaricidal effects. However, doxycycline is contraindicated for a large portion of the at-risk population. To identify new targets for anti-wolbachial therapy, understanding the molecular basis of the Wolbachia-filaria symbiosis is required. We performed cross-species hybridization by using the Brugia malayi microarray to identify differentially expressed genes in the rodent filaria Litomosoides sigmodontis after depletion of Wolbachia which therefore might have a role in symbiosis. Female adult Litomosoides sigmodontis from patent infections were treated with tetracycline to deplete endosymbiotic Wolbachia bacteria. RNA from tetracycline-treated Litomosoides sigmodontis was compared to untreated age-matched control worms. This experiment was performed for three different timepoints: day 6, 15 and 36 of tetracycline treatment. One biological replicate was performed each with two technical replicates (dye-flip replicates).

Filariasis test strip (FTS) antibody recognition of Brugia malayi glycoproteins

Project description:Rapid and accurate prevalence mapping of lymphatic filariasis (LF) is necessary to eliminate this disfiguring and disabling neglected tropical disease. Unfortunately, rapid tests such as the filariasis test strip (FTS) for Wuchereria bancrofti, the causative agent of LF in Africa, can cross-react with antigens circulating in some persons infected by the African eye worm, Loa loa, rendering the test unreliable in eleven co-endemic nations. The intended target of the FTS is a heavily glycosylated W. bancrofti circulating filarial antigen (Wb-CFA). Previously, we determined that the FTS monoclonal antibody, AD12.1, which detects a carbohydrate epitope on Wb-CFA, also detects multiple L. loa proteins in cross-reactive sera from persons with loiasis. Since the carbohydrate epitope recognized by AD12.1 is present on glycoproteins of other parasitic nematodes, including Brugia species, it is unclear why reactive glycoproteins are not detected in infections with other filarial parasites. To gain a better understanding of the proteins recognized by the FTS diagnostic antibody, we used proteomics and to characterize filarial glycoproteins that are bound by the AD12.1 antibody using Brugia malayi as a model. This included using shotgun proteomics to catalog the AD12-reactive glycoproteins found in adult female somatic (BmSOM) and excretory/secretory (BmES) antigens. As well as in-gel proteomics of two predominant protein bands from BmES.

Deep sequencing of C. elegans microRNAs under starvation conditions

Project description:In nature, animals often face feast or famine conditions. We aimed to identify the miRNAs of Caenorhabditis elegans that changed their expression under starvation conditions in stage L4 larvae. Our results highlight 14 miRNAs that show differential expression in starved versus well-fed larvae. In particular, miRNAs of the miR-35-3p/miR-41-3p family were upregulated 6-20 fold upon starvation. We verified the upregulation of miR-35-3p with qPCR. Additionally, we showed that the expression of gld-1, important in ovogenesis, and a validated target of miR-35-3p, was downregulated when the expression of miR-35-3p was higher. This study represents a starting point for a more comprehensive understanding of the role of miRNAs during starvation in C. elegans. Illumina small RNA sequencing of starved and well-fed L4 worms.

After 24 hours at 25°C, the worms were collected into trizol for further RNA extraction. Controls worms were non infected.

The Drosophila immune response towards entomopathogenic nematodes and their associated bacteria.

Project description:Entomopathogenic nematodes (EPNs) of the genera Heterorhabditis are obligate and lethal insect parasites. In recent years they have been used increasingly as biological control agents. These EPNs are symbiotically associated with bacteria of the genera Photorhabdus. The bacterial symbionts are essential to kill the host (within 24-48 hours) and digest its tissues to provide nutrients for themselves as well for expanding nematodes. Drosophila larvae are suitable insect hosts and part of the tripartite model system we used before to show the importance of haemolymph clotting and eicosanoids during the infection. We used the well-established tripartite model (Drosophila, nematodes, bacteria), DNA chips and bioinformatic tools to compare gene expression in non-infected and infected fly larvae. We focused on the early time point of nematode infection and therefore infected Drosophila larvae using H. bacteriophora harbouring GFP-labelled P. luminescens bacteria. Infected (GFP positive) larvae were collected 6 hours after infection.
