A detailed Proteomic map of the mechanical disease vector Musca domestica adult flies

ABSTRACT: In the present study, we have mapped the global proteome of the adult stage of the mechanical disease vector M. domestica. Our study resulted in identification of 3,615 proteins from both the male and female stages of M. domestica. Our study provides the first large scale proteomic map of the common housefly M. domestica.

Project description:Complement factor H (CFH) is an enzyme responsible for inactivating components of the complement cascade, thus limiting the downstream effector mechanisms which would otherwise occur were the process to continue unregulated. In view of the close structural and functional homology between β2GPI and CFH we sought in this study to determine whether CFH is a substrate for oxidoreductases, and whether distinct redox forms can be found in the plasma or serum of individuals and if so to determine the functional implications of such redox transformations to CFH complement regulatory function. This dataset relates to the detection of reduced disulfide bonds in CFH by TRX-1/TRX-R/NADPH.

Analysis of Plasma Membrane, Endosomal and Cytoplasmic fractions of mouse embryonic fibroblast

Project description:This project aimed to identify novel endosomal proteins and therefore cellular fractionation experiments were performed. Adherent mouse embryonic fibroblast were scraped on ice into hypotonic lysis buffer (10 mM HEPES-KOH pH 7.2, 0.25 M sucrose, 1 mM EDTA, 1 mM MgOAc and protease & phosphatase inhibitors (PhosSTOP and Complete mini tablets from Roche). Cell membranes were fragmented with French Press and nuclei removed with 10 min 1000 x g centrifugation. Plasma membrane fraction was collected with 10 min 10 000 x g centrifugation and endosomal/cytoplasmic fractions with 1h 100,000 x g centrifugation. Membrane fractions were washed at least once with lysis buffer and cytoplasmic fractions were centrifuged twice. All fractionation steps were performed at +4°C or on ice. All fractions were dissolved in sample buffer for immunoblotting

Quantitative phosphoproteomic analysis reveals system-wide signaling pathways regulated by site-specific phosphorylation on Keratin-8 in skin squamous cell carcinoma derived cell-line

Project description:The epithelial specific keratin pair, Keratin 8/18 (K8/18), has been reported to be aberrantly expressed in squamous cell carcinoma (SCC) which is also correlated with increased invasiveness and poor prognosis. A Majority of Keratin 8 (K8) functions are governed by its phosphorylation at Serine73 (head domain) and Serine431 (tail domain) residues. Although deregulation of K8 phosphorylation has been associated with progression of different carcinomas, its role in skin SCC and the underlying mechanism is obscured. To understand the molecular basis of K8 phosphorylation mediated regulation of skin SCC progression, we performed TMT-based quantitative phosphoproteomics by expressing K8 wild type, phosphodead and phosphomimetic mutants in K8 deficient A431 cells. Bioinformatic analysis of our phosphoproteomic data showed differential regulation of phosphoproteins associated with migratory, proliferative and invasive potential in these cells. Further validation of protein phosphorylation levels and phenotypic validation of our phosphoproteomic data suggested potential role of K8 site specific phosphorylation/dephosphorylation in neoplastic progression of A431 cells.

Depletion of Keratin8 affects tumorigenic potential of A431 cells via altering multiple signaling pathways

Project description:Keratins are the largest subgroup of intermediate filament proteins which are expressed in a tissue specific and differentiation state dependent manner. Apart from their mechanical functions, they have been shown to perform various regulatory roles. Most of these functions have been studied on keratin pair of 8/18 (K8/18) which is preferentially expressed in simple epithelia. The aberrant expression of this keratin pair has been correlated with increased invasiveness and poor prognosis of various squamous cell carcinomas (SCCs) including Skin SSCs. However the exact role it’s of aberrant expression in skin SCCs and associated mechanism remains elusive. To understand the role of K8/18 in skin SCC, ShRNA based stable K8 knockdown clones were generated in skin epidermoid carcinoma derived A431 cells which resulted in the decreased tumorigenic potential of these cells. Next, to decipher the molecular basis behind K8 mediated regulation of tumorigenic potential of A431 cells, TMT based total quantitative proteomics was performed for vector control and K8 knockdown clones (K8KD1/K8KD2). There were a total of 2952 proteins detected in the TMT based total quantitative proteomics, out of which, K8KD1 clone showed 140 proteins to be differentially expressed, whereas K8KD2 clone showed 119 proteins to be differentially expressed in comparison to vector control cells. Further, functional analysis of our data showed involvement of cell death and survival together with cellular movement pathway to be affected upon K8 knockdown. Altogether, the present data potentiates the role of K8 in neoplastic progression of skin SCC, together with the possible mechanism underlying the same.

Immunoprecipitation of PTEN coupled Nano-LC−ESI-MS/MS

Project description:Nuclear localization of PTEN is essential for its tumor suppressive role, and loss of nuclear PTEN is more prominent than cytoplasmic PTEN in many kinds of cancers. However, nuclear PTEN-specific regulatory mechanisms were rarely reported. Based on the finding that nuclear PTEN is more unstable than cytoplasmic PTEN. Thus, 293T cells were transiently transfected with plasmid encoding Flag-tagged PTEN along with empty vector, followed by affinity purification using an anti-Flag antibody, and the bound proteins were analyzed by liquid chromatography with tandem mass spectrometry (LC−MS/MS). Here we identify that F-box only protein 22 (FBXO22) induces ubiquitylation of nuclear but not cytoplasmic PTEN at lysine 221, which is responsible for the degradation of nuclear PTEN.

Human melanoma-derived extracellular vesicles reprogram dendritic cells potentially enabling lymph node metastases

Project description:Evolution of melanoma from a primary tumor to widespread metastasis is crucially dependent on lymphatic spread. The mechanisms regulating the initial step in metastatic dissemination via regional lymph nodes remain largely unknown. We have previously described a dysfunctional immune profile that precedes evidence of metastasis in the first node draining from the primary tumor, the sentinel lymph node (SLN). Herein, we explore the role of melanoma-derived extracellular vesicles (EVs) as mediators of this pre-metastatic niche through cargo-specific polarization of dendritic cells (DCs). Utilizing mass cytometry, pre-metastatic SLNs demonstrate compromised co-stimulatory CD80 expression compared to healthy lymph nodes. Similarly, DCs matured in vitro in the presence of melanoma EVs showed impaired co-stimulation and polarization towards a chronic inflammatory cytokine milieu. Profiling of melanoma EV cargo identified shared proteomic and RNA signatures including the signaling axis S100A8, S100A9 and cognate receptor TLR4. Mechanistically, S100A8 and S100A9 compromised DC maturation, a phenotype which was partially recovered following TLR4 blockade. Early evidence demonstrates similar EVs can be isolated from human afferent lymphatic fluid ex vivo. Taken together, we propose synergistic interactions among melanoma EV cargo are responsible for suppressing DC maturation, potentially explaining the survival of malignant melanocytes metastasizing into seemingly “normal” regional lymph nodes.

Xylem sap shotgun proteomics from Mn-excess tomato plants

Project description:The aim of this work was to study the effects of Mn excess on the protein profile of the xylem sap of tomato (Solanum lycopersicum), with the aim of elucidating plant response mechanisms to these nutritional stresses. Tomato was chosen as a model plant because the tomato genome has been published and this plant species has adequate root pressure and turgid stems that permit xylem sap sampling in sufficient amounts by de-topping. The high- throughput shotgun analysis has permitted to identify and quantitate a large number of low abundance proteins in the tomato xylem sap.
