LCMS spermatozoa and seminal plasma

ABSTRACT: This dataset contains raw and processed LC–MS/MS data from a paired proteomic analysis of seminal plasma and spermatozoa fractions obtained from the same ejaculates of two adult stallions. Samples were analyzed using a Vanquish nanoLC system coupled to an Orbitrap Exploris 240 mass spectrometer operated in data-dependent acquisition (DDA) mode. Database searches were performed in Proteome Discoverer (v3.1) using the Sequest HT search engine against the Equus caballus UniProt reference proteome (UP000002281). Peptide and protein identifications were filtered at 1% FDR with a minimum of two unique peptides per protein. The dataset supports abundance-based comparison of functional specialization between seminal plasma and spermatozoa compartments.

Ejaculate samples for male infertility screening

Project description:The problem of the male reproductive system has both medical and social importance. It is highlighted that spermatozoa or seminal plasma are suitable samples for proteomic analysis of the male reproductive system. We suggested that the ejaculate could serve as one of the analytic specimens to determine fertility, which obtainable with relative ease and which contains many proteins. Moreover, the testis-specific proteins could form the basis of protein panel for detecting disorders associated with male infertility. The current study aimed to examine the ejaculate proteome compared to the spermatozoa and seminal plasma proteomes to determine a panel of proteins that can be used to assess sperm quality. Our results showed that the ejaculate represented a productive source for understanding the male reproductive physiology and could serve as a potential origin for the identification of novel protein biomarkers related to spermatozoa function in infertile. We believe that proteomic analysis based on testis-specific proteins of ejaculate along with spermatozoa (or even instead) can be useful for distinguishing the fertile, sub fertile, and infertile men.

Social status and ejaculate investment in the house mouse c4pr_liv

Project description:Sperm competition theory predicts that males should tailor ejaculates according to their social status. Here we test this in a model vertebrate, the house mouse (Mus musculus domesticus), combining experimental data with a quantitative proteomics analysis of seminal fluid composition. Our analyses reveal that both sperm production and the relative production of proteins found in seminal fluid differ according to social dominance. Notably, whereas dominant males produce and ejaculate more sperm, subordinate males produce greater relative amounts of key proteins used in the formation of copulatory plugs. These findings have important implications for understanding the dynamics and outcome of sperm competition.

The transcriptome of ejaculates used for successful/failed in vitro-fertilization

Project description:We investigated the transcriptome obtained from microarray analysis of human ejaculates used in an IVF programme. The gene expression profiles are to be correlated with successful/failed induction of oocyte fertilisation and/or pregnancy. Since human ejaculates are often contaminated with so-called 'round cells' (mostly somatic cells such as leukocytes and macrophages, but also immature germ cells), a considerable part of the gene expression pertains not to spermatozoa. Rigorous statistical analysis of the data is to be used to extract genes that correlate with fertilization success. A set of 25 human ejaculates (12 pregnancy; 3 biochemical pregnancy; 7 oocyte fertilization; 3 no oocyte fertilization) from an IVF procedure were used for RNA extraction and microarray hybridization.

Characterization of the Sperm Proteome and Reproductive Outcomes with in Vitro Fertilization after a Reduction in Male Ejaculatory Abstinence Period

Project description:Semen samples from men after a short ejaculatory abstinence show improved sperm quality and result in increased pregnancy rates, but the underlying mechanisms remain unclear. Herein, we report that ejaculates from short (1–3 hours) compared with long (3–7 days) periods of abstinence showed increases in motile sperm count, sperm vitality, normal sperm morphology, acrosome reaction capacity, total antioxidant capacity, sperm mitochondrial membrane potential, high DNA stainability, and a decrease in the sperm DNA fragmentation index (P < 0.05). Sperm proteomic analysis showed 322 differentially expressed proteins (minimal fold change of ±1.5 or greater and P < 0.05), with 224 up-regulated and 98 down-regulated. These differentially expressed proteins are profoundly involved in specific cellular processes, such as motility and capacitation, oxidative stress, and metabolism. Interestingly, protein trimethyllysine modification was increased, and butyryllysine, propionyllysine, and malonyllysine modifications were decreased in ejaculates from a short versus long abstinence (P < 0.05). Finally, the rates of implantation, clinical pregnancy, and live births from in vitro fertilization treatments were significantly increased in semen samples after a short abstinence. Seminal plasma and transforming growth factor-β regulate gene expression in human Ect1 ectocervical epithelial cells

Project description:In this study we examined the influence of seminal plasma on gene expression in human Ect1 ectocervical epithelial cells, and the extent to which recombinant TGFβ3 elicits comparable changes. Ect1 cells were incubated with recombinant human TGFβ3 (5 ng/ml), 10% pooled human seminal plasma (v/v), or medium alone for 10h. RNA was reverse transcribed into cDNA and hybridized to Affymetrix GeneChip® Human Genome U133 plus 2.0 microarrays (Affymetrix, Santa Clara, CA). Exposure of Ect1 cells to seminal plasma resulted in differential expression of a total of 3955 probe sets, identified using high stringency criteria with MAS 5.0 analysis. These corresponded to 1338 genes up-regulated and 1343 genes down-regulated by seminal plasma. TGFβ3 treatment of Ect1 cells resulted in differential expression of 884 probe sets, corresponding to 346 up-regulated genes and 229 down-regulated genes. The genes differentially regulated by seminal plasma included several genes associated with cytokine–cytokine receptor interaction, TGFβ signalling, JAK/STAT signalling or VEGF signalling pathways, as specified by the KEGG database. Of 47 genes in these families, 17 (36.1%) were similarly regulated by both seminal plasma and TGFβ3. These data, together with additional experiments showing all three TGFβ isoforms can regulate inflammatory cytokine expression in Ect1 cells, identify TGFβ isoforms as key agents in seminal plasma that signal induction of pro-inflammatory cytokine synthesis in cervical cells. RNA from each of four biological replicates, each comprising pooled material from separate sets of 4 replicate wells, was analysed for each treatment. Total RNA was reverse transcribed into cDNA and sent to the Australian Genome Research Facility (AGRF; Melbourne, Australia) for single-cycle labeling and hybridization to 12 Affymetrix GeneChip® Human Genome U133 plus 2.0 microarrays (Affymetrix, Santa Clara, CA).

Seminal fluid components alone stimulate post-mating changes in honey bee queens (Apis mellifera)

Project description:Honey bee queens undergo dramatic behavioral (e.g., reduced sexual receptivity), physiological (e.g., ovary activation, ovulation, and modulation of pheromone production) and molecular changes after they complete mating. To elucidate how queen post-mating changes are influenced by seminal fluid, a non-spermatozoa-containing component of semen, we injected queens with semen or seminal fluid alone. We assessed queen sexual receptivity, ovary development, worker retinue response (which is influenced by queen pheromone production), and transcriptional changes in queen abdominal fat body and brain tissues. Injection with either seminal fluid or semen resulted in decreased sexual receptivity, increased attractiveness of queens to workers, and altered expression of several genes that are also regulated in naturally mated queens. The post-mating and transcriptional changes of queens receiving seminal fluid were not significantly different from queens treated with seminal fluid, suggesting that components in seminal fluid, such as seminal fluid proteins, are largely responsible for stimulating post-mating changes in queens.

Project description:Epididymal Protease Inhibitor (EPPIN) is a sperm-associated protein that serves as a docking site for protein protein interactions. Its binding to seminal plasma proteins plays a key role in modulating sperm function. However, the molecular mechanisms underlying these roles remain unclear. This dataset contains proteomic profiles of proteins from human ejaculated spermatozoa that were immunoprecipitated using an anti EPPIN antibody and analyzed by mass spectrometry. This study aimed to characterize proteins associated with EPPIN in human spermatozoa, with a focus on identifying potential partners related to sperm function within intracellular compartments.
