The aim was to identify differentially expressed matrix proteins between FB and FT or OV which may promote the neoplastic transformation of serous tubal intraepithelial lesions (STICs) into high-grade serous ovarian cancer, HGSOC, and metastasis from the FB to the OV.

Investigating macrophage multinucleation by LC-MS/MS

Project description:Macrophage multinucleation (MM) is essential for various biological processes such as osteoclast-mediated bone resorption and multinucleated giant cell-associated inflammatory reactions. Here we study the molecular pathways underlying multinucleation in the rat through an integrative approach combining MS-based quantitative phosphoproteomics and transcriptome (high throughput RNA-sequencing) to identify new regulators of MM. We show that a strong metabolic shift towards HIF1-mediated glycolysis occurs at transcriptomic level during MM, together with modifications in phosphorylation of over 50 proteins including several ARF GTPase activators and polyphosphate inositol phosphatases. These results will provide a new framework for the combined analysis of transcriptional and post-translational changes during macrophage multinucleation, prioritizing essential genes and revealing the sequential events leading to the multinucleation of macrophages.

Effect of the Src inhibitor peptide TAT-Cx43266-283 in lung cancer brain metastasis models.

Project description:Background: TAT-Cx43266-283 is a novel inhibitor of Src, which has shown important antitumor effects in preclinical models of glioblastoma. Because Src plays a pivotal role in several tumors, including lung cancer and brain metastasis, in this study we investigated the effect of TAT-Cx43266-283 in lung cancer brain metastasis, an unmet clinical need. Methods: Cancer Stem Cells (CSCs) from the murine lung cancer cell line LLC, named LLC-CSCs, were isolated and used to study the effect of TAT-Cx43266-283 in vitro and to develop an in vivo model of lung cancer brain metastasis. Thus, in vivo models were carried out performing an intracranial implantation of LLC-CSCs in immunocompetent mice and the impact of TAT-Cx43266-283 in these models was assessed. A phosphoproteomic analysis was employed to identify signaling pathways affected by TAT-Cx43266-283, and the most prominent targets were validated by immunofluorescence or Western Blot. Results: We found that TAT-Cx43266-283 significantly reduced LLC-CSCs viability and increased the survival of immunocompetent mice bearing lung cancer derived-brain metastatic tumors. Phosphoproteomic analysis identified ERK as a key effector of TAT-Cx43266-283 treatment in the LLC-CSC-derived brain metastasis model. TAT-Cx43266-283 also induced apoptosis of tumoral cells and disrupted the cytoskeleton and vascularization of the tumor tissue in this preclinical lung cancer brain metastasis model. Conclusions: Our results suggest that TAT-Cx43266-283 is a promising antitumoral drug for lung cancer brain metastasis treatment. We identified the inhibition of ERK as an important effector, together with the inhibition of Src, in the antitumoral effect of TAT-Cx43266-283 in lung cancer brain metastasis. All this together, places TAT-Cx43266-283 one step forward into its journey to its therapeutic implementation.

Renal gene expression in uninephrectomized diabetic OVE26 mice

Project description:OVE26 (OVE) mice provide a useful model of advanced diabetic nephropathy (DN) with respect to albuminuria and pathologies. We showed that albuminuria, reduced GFR and interstitial fibrosis, which normally take 8-9 months to develop, are more advanced in uninephrectomized OVE mice within 10 weeks of surgery, at 4.5 months of age. The accelerated progression of renal damage, especially renal fibrosis in OVE-uni mice, was also identified at the gene expression level. The hepatic fibrosis/hepatic stellate cell activation pathway was by far the most significant Ingenuity canonical pathway identified by gene array in OVE-uni mice. Many inflammatory- and immune-related pathways were found among the top pathways up-regulated in OVE-uni kidneys, including acute-phase response signaling, leukocyte extravasation, IL6, IL10, IL12 signaling, TREM1 signaling, dendritic cell maturation and the complement system. These pathways were also dramatically up-regulated in 8-month-old OVE mice (GSE20636). Nephrectomized OVE mice are a much faster alternative model for studying advanced renal disease in diabetes. Extracellular matrix educates a tumor macrophage phenotype found in ovarian cancer metastasis

Project description:Recent studies have shown the tumor extracellular matrix (ECM) associates with immunosuppression, and that targeting the ECM can improve immune infiltration and immunotherapy response. A question that remains is whether the ECM is directly educating the immune phenotypes seen in cancer. We identified a tumor-associated macrophage (TAM) population correlated with poor prognosis, interruption of the cancer immunity cycle, and tumor ECM composition. To investigate whether ECM was capable of generating the TAM phenotype seen, we developed a decellularized tissue model that retains the native ECM architecture and composition. Macrophages cultured on decellularized ovarian metastasis shared transcriptional profiles with the TAMs found in human tissues. ECM educated macrophages have a tissue remodeling and immunoregulatory phenotype, inducing altered T cell function. We conclude that the tumor ECM is directly educating this macrophage population found in cancer tissues. Therefore, current and emerging cancer therapies that target the tumor ECM may be tailored to improve macrophage phenotype and their downstream regulation of immunity.
