The role of Akkermansia muciniphila sulfatases in colonic mucin utilisation

ABSTRACT: Akkermansia muciniphila proteome analysis by label-free mass spectrometry showed the upregulation of three sulfatases (Amuc0491, Amuc1655 and Amuc1182) when grown on colonic mucins versus gastric mucins.

Nutrient-specific Proteomic Analysis of the Mucin Degrading Bacterium Akkermansia muciniphila

Project description:Akkermansia muciniphila is a prominent mucin-degrading bacterium that acts as a keystone species in regulating the human gut microbiota. Despite recently increasing research into this bacterium and its relevance to human health, a high-resolution database of its functional proteins remains scarce. Here, we provide a proteomic database of A. muciniphila grown in different nutrient conditions ranging from defined to complex. Of 2,318 protein-coding genes in the genome, we identified 841 (40%) that were expressed at the protein level. Overall, proteins involved in energy production and carbohydrate metabolism indicate that A. muciniphila relies mainly on the Embden-Meyerhof-Parnas pathway, and produces short-chain fatty acids through anaerobic fermentation. Moreover, this bacterium possesses a broad repertoire of glycosyl hydrolases, together with putative peptidases and sulfatases, to cleave O-glycosylated mucin. Furthermore, it uses mucin-derived monosaccharides as sources of energy and cell wall biogenesis. Our database provides nutrient-dependent global proteomes of A. mucinphila ATCC BAA-835 to offer insights into its metabolic functions that shape the composition of the human gut microbiota via mucin degradation.

Akkermanisa muciniphila response to oxygen exposure

Project description:The anaerobic gut microbe Akkermansia muciniphila ATCC BAA-835 lives in the mucus layer where it is exposed to oxygen. To investigate how it survives the changing oxygen concentrations, we exposed a exponentially growing culture to oxygen. The experiment was performed in parallel fermentor systems. To one system we added 0.2l/h oxygen after the culture reached an OD of 0.1, while the other remained anaerobic. Samples were taken just before addition of oxygen, about 1h after and when the cell reached stationary phase.

Secreted Akkermansia muciniphila threonyl-tRNA synthetase functions to monitor and modulate immune homeostasis

Project description:Commensal bacteria are critically involved in the establishment of tolerance against inflammatory challenges, the molecular mechanisms of which are just being uncovered. All kingdoms of life produce aminoacyl-tRNA synthetases (ARSs). Thus far, the non-translational roles of ARSs have largely been reported in eukaryotes. Here, we report that the threonyl-tRNA synthetase (AmTARS) of the gut-associated bacterium Akkermansia muciniphila is secreted and functions to monitor and modulate immune homeostasis. Secreted AmTARS triggers M2 macrophage polarization and orchestrates the production of anti-inflammatory IL-10 via its unique, evolutionary-acquired regions, which mediates specific interactions with TLR2. This interaction activates the MAPK and PI3K/AKT signaling pathways, which converge on CREB, leading to an efficient production of IL-10 and suppression of the central inflammatory mediator NF-κB. AmTARS restores IL-10-positive macrophages, increases IL-10 levels in the serum, and attenuates the pathological effects in colitis mice. Thus, commensal tRNA synthetases can act as intrinsic mediators that maintain homeostasis.

Trans-Golgi protein TVP23B regulates host-microbe interactions via Paneth cell homeostasis and Goblet cell glycosylation

Project description:A key feature in intestinal immunity is the dynamic intestinal barrier, which separates the host from resident and pathogenic microbiota through a mucus gel impregnated with antimicrobial peptides. The mechanisms underlying the maintenance and function of this intestinal barrier are not completely understood. Using a mouse forward genetic screen for defects of intestinal homeostasis, we have found a mutation in Tvp23b, which conferred susceptibility to chemically induced and infectious colitis. Golgi apparatus membrane protein TVP23 homolog B (TVP23B) is a transmembrane protein conserved from yeast to humans. In the intestine, the protein is localized to the epithelium and its deficiency in the hematopoietic extrinsic compartment was essential to the colitis phenotype. We found that TVP23B controls the homeostasis of Paneth cells and function of goblet cells in vivo, leading to a decrease in antimicrobial peptides as well as a more penetrable mucus layer. As a result, Tvp23b-/- mice displayed decreased barrier function and a loss of host-microbe separation. TVP23B-deficient colonocytes have a loss of core-3 O-glycosylation of colonic proteins, which is the major O-glycosylation present on gel forming mucins. TVP23B binds with another Golgi protein, YIPF6, which is similarly critical for intestinal homeostasis. The Golgi proteomes of YIPF6 and TVP23B-deficient colonocytes have a common deficiency of several critical glycosylation enzymes, including those necessary for core-3 glycosylation of mucins. TVP23B is necessary for the formation of the sterile mucin layer of the intestine and its absence disturbs the balance of host and microbe in vivo.

IL-22 promotes postnatal formation of MUC17 glycocalyx barrier that protects host enterocytes

Project description:The apical aspects of absorptive epithelial cells form a highly organized brush border membrane (BBM), shaped by densely packed microvilli that are coated with a glycocalyx. Here, we present evidence showing that the glycocalyx forms an epithelial barrier that prevents exogenous molecules from gaining access to the BBM. We used a multiomics approach to investigate function and regulation of membrane mucins exposed on the enterocytic BBM during postnatal development of the small intestine. Muc17 was identified as a major membrane mucinin the glycocalyx that was specifically upregulated by IL-22 as part of an epithelial defense repertoire during suckling-weaning transition. High levels of IL-22 at the time of weaning primed progenitor enterocyte to express Muc17 as they migrate out of intestinal crypts to replace neonatal enterocytes. Our findings propose a role for membrane mucin Muc17 in epithelial barrier function in the small intestine.

Proteome profiling of Akkermansia muciniphila, Bifidobacterium longum, and Bifidobacterium breve in different nutrient environments

Project description:To understand how glycans influence the growth of probiotic strains in monocultures and communities, we characterized the growth and perform proteome profiling of 3 major probiotics, A. muciniphila (AM) BAA-835, B. breve (BB) JCM1192, and B. longum (BL) JCM1217 in monocultures and co-cultures grown in media with monosaccharides (Glucose & GlcNAc), Mucin, or Arabinoxylan. For the proteomic file annotations, A refers to AM monoculture, B refers to BB monoculture, and L referes to BL monoculture, AL refers to AM+BL co-culture, AB refers to AM+BB co-culture. The letter following the underline specify the media: g refers to Glc+GlcNAc, m refers to mucin, ax refers to arabinoxylan. If there is no underline following the species annotations, the cultures were grown in the base media only. Numbers following the letters specify biological replicates.

COPD lungs show an attached stratified mucus layer resembling the protective colonic mucus

Project description:Mucus accumulation is a key feature of respiratory diseases such as cystic fibrosis (CF) and chronic obstructive pulmonary disease (COPD). This is associated with goblet cell metaplasia and mucin overexpression, which can be induced by the increased activity of neutrophil elastase. The aim of the study was to characterization of mucus and epithelial cell proteomics in a porcine pancreatic elastase (PPE) mouse model of COPD/CF. The major proteins detected in mucus plugs obtained from PPE-treated mice included mucins Muc5ac and Muc5b, mucus-related proteins Clca1, Fcgbp, and Bpifb1. These proteins were upregulated in bronchoalveolar lavage (BAL) fluid and epithelial cells in mice exposed to elastase. Similar changes were found in BAL fluid of COPD patients.

Lung transplant patients BALF proteomics, study of CLAD patients

Project description:In the current study, we use mass spectrometry-based proteomics to analyze bronchoalveolar lavage fluid (BALF) samples collected from a cohort of 43 patients of which 30 patients present a lung function that is stable over 36 months and 13 patients that develop CLAD within the first-year post-transplant. The mass spectrometry method used label free quantification (LFQ) of all the proteins detected as well as heavy labelled peptides for absolute quantification of mucins and related proteins.

Akkermansia muciniphila secretome
