AAV delivered lysosome-targeting chimeras mediate sustained antibody depletion in vivo (absolute IgG quantification)

ABSTRACT: Genetically-encoded lysosome targeting chimeras (GELYTACs) were developed against circulating IgGs. To achieve long-lasting IgG depletion, GELYTACs were encoded in an AAV gene therapy vector and continuous expression in mice was established. To assess the efficiency of IgG depletion and subclass biases, we absolutely quantified IgGs from serum samples using calibration curves of heavy labeled reference peptides.
