A pathological phosphorylation pattern enhances tau cooperativity on microtubules and facilitates tau filament assembly
Ontology highlight
ABSTRACT: The phosphorylation of the microtubule-associated protein tau plays important roles in both physiological and pathological processes such as fetal development and the formation of neurofibrillary tangles (NFTs) in neurodegenerative diseases. Although tau phosphorylation levels are similarly elevated in both processes, the phosphorylation patterns and their respective functions remain unclear. In this project, we identified and analyzed the levels of specific phosphorylation sites in physiological- and pathological-like tau proteins expressed in mammalian and insect cells respectively. This project aims to investigate the effect of different phosphorylation patterns on tau’s microtubule binding, cooperative envelope formation, oligomerization, and filament assembly relevant to tauopathies. The associated research paper is in preparation.
