Research on the secretome of Arabidopsis PHR1 in Pi-starvation response

ABSTRACT: Through hydroponic culture under phosphate-sufficient (+Pi) and phosphate-deficient (−Pi) conditions, a total of 2,078 high-confidence secreted proteins were quantitatively identified from WT and phr1 plants. These proteins were functionally categorized into phosphatases, signal transduction proteins, pathogenesis-related (PR) proteins, cell wall remodeling proteins, and reactive oxygen species (ROS) metabolism-related proteins. Under phosphate deficiency, PHR1 appears to be involved in regulating the expression of phosphatases and PR proteins, which participate in organic phosphorus utilization, pathogen resistance, and rhizosphere microbial colonization. Meanwhile, under Pi-starved conditions, the expression levels of ROS-associated proteins and cell wall remodeling proteins were significantly upregulated, and these proteins are implicated in the modification of cell wall architecture and root system architecture. Furthermore, by analyzing acid phosphatase (APase) activity and ROS content, we further elucidated the functional significance of the differentially expressed secreted proteins between WT and phr1 mutant plants under both +Pi and −Pi conditions.
