Development of a laser injury model using human cardiac spheroids
Ontology highlight
ABSTRACT: This study establishes a laser injury model using 3D cardiac spheroids composed of human induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) and cardiac fibroblasts (iPSC-CFs). Following the in vitro differentiation of both iPSC-CMs and iPSC-CFs, the two cell types are co-cultured to allow self-aggregation into cardiac spheroids. Focal injury is induced via targeted laser ablation, after which the injured spheroids are cultured for 7 days post-ablation to observe biological responses. Spheroids are subsequently harvested and snap-frozen in liquid nitrogen for downstream molecular and histological analysis.
