DIA proteomic profiling of conditioned media from human umbilical cord mesenchymal stem cells with different telomere lengths
Ontology highlight
ABSTRACT: Human umbilical cord mesenchymal stem cells (hUMSCs) were isolated from the Wharton’s jelly of 50 neonatal umbilical cords. Telomere length was determined in passage 1 hUMSCs using terminal restriction fragment analysis (TRF). Five hUMSC samples with the shortest telomeres (bottom 10%) and five samples with the longest telomeres (top 10%) were selected as the short-telomere and long-telomere groups, respectively. Equal numbers of passage 5 hUMSCs were seeded into T75 culture flasks at a density of 1 × 10^4 cells/cm². After 72 h of culture, conditioned media were collected and subjected to DIA-based quantitative proteomic analysis. A total of 3,339 protein groups were quantified, and 204 differentially expressed proteins were identified between the short- and long-telomere groups.
