Mass spectrometry characterization of O-GlcNAc modification of GFP-tagged G3BP1

ABSTRACT: This dataset contains mass spectrometry data generated to characterize O-GlcNAc modification of human G3BP1 expressed as an N-terminal EGFP fusion in H9c2 cardiac cells. EGFP-G3BP1 was overexpressed by adenoviral transduction, affinity purified using GFP-Trap, resolved by SDS-PAGE, and the band corresponding to EGFP-G3BP1 was excised for mass spectrometric analysis. HCD-triggered EThcD acquisition was used to identify HexNAc-modified peptides and localize modification sites. The dataset provides peptide sequence coverage and site-level information for G3BP1 O-GlcNAcylation, including localization of a HexNAc modification to T268 as reported in the associated study.
