LC-MS/MS label-free quantification of Mal d 1 isoforms in apple cultivars hypoallergenic for birch pollen-allergic individuals before and after long-term storage

ABSTRACT: Birch pollen-related apple allergy is the most frequent IgE-mediated food allergy in Central-Northern Europe, with Mal d 1 as the major allergen. Its concentration varies with the cultivar and storage time. In this study, Mal d 1 isoforms were profiled by LC-MS/MS in fruits of three well-storable cultivars bred at the Bavarian Centre of Pomology and Fruit Breeding ('Bay 4069' (tradename 'Graefin Goldach'), 'Bay 4152' and 'Bay 4210' (tradename 'Sonnenglanz')) and in the reference cultivars 'Golden Delicious' (high allergenicity) and 'Santana' (low allergenicity). Fruits were harvested in September 2018, stored at 1.5 C and analysed after 7 weeks (November 2018) and 29 weeks (April 2019) of storage (raw files of the April 2019 sampling are labelled 'May' in their file names); 'Santana' was analysed in November 2018 only because of its limited storability. Three individual fruits per cultivar and time point were measured (27 LC-MS/MS runs, LTQ Orbitrap Elite, data-dependent CID). Mal d 1 isoforms were identified and quantified label-free with MaxQuant against a dedicated Mal d 1 isoform sequence database. The data accompany Strobl et al., Clin Transl Allergy 2022, doi:10.1002/clt2.12186, in which the MS results were combined with immunoblotting, ELISA, basophil activation tests and clinical challenges.
