Quantification of subunit abundance of wild-type and tagged Wadjet from inducible expression in an E. coli lab strain
Ontology highlight
ABSTRACT: We observed that a tagged variant of the anti-plasmid defence system Wadjet (JetABCD), carrying an N-terminal HaloTag on the JetA subunit, restricts large plasmids more efficiently than the wild-type system. To test whether this increased activity against large plasmids results from altered expression of the JetABCD subunits in E. coli, we performed proteomic analysis of crude cell lysates from cells expressing either wild-type or HaloTag-JetA Wadjet. Expression was induced after backdilution into fresh LB medium with arabinose (0.02% w/v) for 10 hr at 37 °C. Protein extracts were prepared by lysozyme treatment and sonication.
