Screening and functional study of genes related to tuberculous ulcers

Project description:Transcriptome analysis was performed to systematically compare human tuberculous ulcers with non-tuberculous skin ulcers. Focusing on the former, this study aimed to identify key associated genes, elucidate their expression profiles and functional attributes, and preliminarily dissect the molecular mechanisms governing their pathological progression. By bridging the current gap in the genetic characterization of tuberculous ulcers, this research not only provides novel targets for investigating disease pathogenesis but also establishes an experimental foundation for early clinical diagnosis and targeted therapeutic strategies, thereby holding significant implications for advancing disease prevention and management.（This study used Agilent Human lncRNA V6 microarray. Host cellular and immune responses in models of inflammatory skin conditions

Project description:Skin colonisation of varied communities of commensal microorganisms, such as Staphylococcus aureus (SA), Staphylococcus epidermidis (SE) and Staphylococcus capitis (SC) form the microbiome; a necessity for healthy skin. The skin changes characteristic of atopic dermatitis, a common inflammatory skin disease, have been shown to provide a favourable niche for SA colonisation. We utilised a reconstructed human epidermal (RHE) model recapitulating the stratified anatomy of the epidermis on which to test host responses to bacterial colonisation. SA proliferation was significantly inhibited in contrast to that seen with SE at both high and low colonisation loads after 24 hours. These data strongly suggest species specific regulation of staphylococcal growth, which is partially mediated by interaction with the epidermis.

A Microneedle Device for Rapid Dermal Interstitial Fluid Sampling

Project description:Dermal interstitial fluid (ISF) shows promise for novel skin diagnostics and as an alternative to invasive blood tests. This dataset includes ISF sampled via a microneedle device from two ex vivo human abdominal skin specimens and the forearms of ten healthy volunteers. Paired blood samples via finger prick were also collected from five volunteers. We studied the ISF proteome in relation to collection time and pressure ex vivo. We also created a reference ISF proteome from the healthy volunteers, compared it to the plasma proteome, and quantified errors from blood contamination.

Project description:Skin commensal bacteria (Staphylococcus epidermidis) can help defend against skin infections, and they are increasingly being recognized for their role in benefiting skin health. This study aims to demonstrate the activities that Myristica fragrans Houtt. seed extracts, crude extract (CE) and essential oil (EO), have in terms of promoting the growth of the skin commensal bacterium S. epidermidis and providing metabolites under culture conditions to disrupt the biofilm formation of the common pathogen Staphylococcus aureus.

Transcriptional programming of human epidermis during wound repair and in pressure ulcer at single-cell resolution

Project description:Pressure ulcer (PU) is a chronic non-healing wound caused by continuous pressure of the bodyweight to the skin, which is often seen in spinal cord injury patients and in the bedridden elderly population. In spite of high mortality, the pathophysiology of PU remains poorly understood. We performed single-cell transcriptomic analysis of epidermal cells from PU wound-edges, and compared them with epidermal cells from the intact skin and normal acute wounds (AW) of healthy donors. We identified four keratinocyte clusters, one melanocyte cluster, and one immune cell cluster, and the cellular heterogeneity and gene expression were altered in PU. Our findings provided a high-resolution map of human PU and AW, which are likely to yield new areas for exploration of the pathophysiology of chronic wounds and development of wound therapy.
