BioID screen for Uhrf2 interactors in AML12 cells

Project description:We identified the ubiquitin ligase Uhrf2 as an important regulator of liver regeneration. To gain insight into the mechanisms of action of Uhrf2 in hepatocytes, we searched for Uhrf2 interactors in the AML12 hepatocyte cell line using BioID interaction screening. This resulted in the identification of major chromatin remodelling proteins as Uhrf2 interactors.

Transcription profiling of mouse AML12 hepatocytes transfected with v-erbA

Project description:The v-erbA oncogene belongs to a superfamily of transcription factors called nuclear receptors, which includes the retinoic acid receptors (RARs) responsible for mediating the effects of retinoic acid (RA). Nuclear receptors bind to specific DNA sequences in the promoter region of target genes and v-erbA is known to exert a dominant negative effect on the activity of the RARs. The repressor activity of v-erbA has been linked to the development of hepatocellular carcinoma (HCC) in a mouse model. We have used microarray analysis to identify genes differentially expressed in hepatocytes in culture (AML12 cells) stably transfected with v-erbA and exposed to RA. We have found that v-erbA can affect expression of RA-responsive genes. We have also identified a number of v-erbA-responsive genes that are known to be involved in carcinogenesis and which may play a role in the development of HCC. Experiment Overall Design: AML12 control cells and v-erbA-transfected AML12 cells were exposed to 1 ÂµM RA for 3h or 24h. Using microarray analysis, we compared gene expression in the presence and absence of v-erbA and identified RA-regulated genes differentially expressed in the presence of v-erbA.

Mouse hepatocyte conditioned media LC-MS/MS.

Project description:Hepatocyte caspase-8 is elevated in MASH and promotes fibrosis independently of apoptosis changes. To investigate how hepatocyte caspase-8 might drive liver fibrosis progression in MASH, we focused on activated hepatic stellate cells (HSCs), which are the primary source of collagen-producing myofibroblasts and central players in MASH fibrosis. To assess a potential link between hepatocyte caspase-8 and HSC activation, we used an ex vivo model where primary murine HSCs were cultured with conditioned media from siCasp8-treated or control primary hepatocytes. Supporting our hypothesis, HSC activation markers were reduced in HSCs exposed to conditioned media from Casp8-silenced AML12 hepatocytes compared to controls. To pinpoint caspase-8-dependent secretory proteins that could activate HSCs, we conducted LC-MS/MS on conditioned media from these cells, identifying secretory proteins reduced in Casp8-silenced AML12 hepatocytes.

Genes expression profile in BMDM challenging with S1P or DAMPs

Project description:To explore the differently expressed genes in BMDM after the treatment with S1P or DAMPs (Damage associated molecular patterns). BMDM cells were incubated with apoptotic cell culture medium (ACM) or S1P (500nM) for 24 hours before harvest for RNA extraction. ACM generated from palmitic acid treated AML12 cells was used as DAMPs. We also want to see whether these genes regulated by S1P or DAMPs are Trem2 dependent or independent.
