Project description:The goal of this study was to understand the link between maternal oral dysbiosis and the gut health of offspring. We demonstrate that maternal oral dysbiosis can have lasting health impacts on offspring. Ligature-induced periodontitis in mothers promotes the expansion of oral pathobionts in the mouth, which are transmitted to the infant gut, rendering offspring more susceptible to enteritis. Notably, although these maternal oral pathobionts are eradicated as the microbiota matures, the imprinted susceptibility to enteritis persists into adulthood.

Transgenerational inheritance of diabetes susceptibility in male offspring with maternal hyperandrogenism [RNA-Seq]

Project description:Maternal hyperandrogenism is closely related to metabolic disorders, yet its transgenerational effects on male descendants remain unclear. Here we demonstrate that hyperandrogenism in women predisposes their sons to islet β-cell dysfunction. Male offspring mice with prenatal androgen exposure exhibited hyperglycemia and glucose intolerance due to compromised insulin secretion. Notably, these metabolic disturbances were transmitted across three generations, and exacerbated by aging and a high-fat diet. Altered DNA methylations in F1 sperm and F2 islet contribute to suppressed β-cell functional genes. We identified shared differentially methylated signatures in F1 sperm, type 2 diabetes patients, and sons with maternal hyperandrogenism. Furthermore, caloric restriction and metformin treatments in F1 males corrected hyperglycemic defects and prevented their transmission to offspring. Our findings highlight the transgenerational inheritance of hyperglycemia and β-cell dysfunction in male offspring from maternal hyperandrogenism via DNA methylation changes, providing methylation biomarkers and therapeutic strategies to safeguard future generations’ metabolic health.

Multi-omic brain and behavioral correlates of cell-free fetal DNA methylation in macaque maternal obesity models

Project description:Maternal obesity during pregnancy is associated with neurodevelopmental disorder (NDD) risk. We utilized integrative multi-omics to examine maternal obesity effects on offspring neurodevelopment in rhesus macaques by comparison to lean controls and two interventions. Differentially methylated regions (DMRs) from longitudinal maternal blood-derived cell-free fetal DNA (cffDNA) significantly overlapped with DMRs from infant brain. The DMRs were enriched for neurodevelopmental functions, methylation-sensitive developmental transcription factor motifs, and human NDD DMRs identified from brain and placenta. Brain and cffDNA methylation levels from a large region overlapping mir-663 correlated with maternal obesity, metabolic and immune markers, and infant behavior. A DUX4 hippocampal co-methylation network correlated with maternal obesity, infant behavior, infant hippocampal lipidomic and metabolomic profiles, and maternal blood measurements of DUX4 cffDNA methylation, cytokines, and metabolites. We conclude that in this model, maternal obesity was associated with changes in the infant brain and behavior, and these differences were detectable in pregnancy through integrative analyses of cffDNA methylation with immune and metabolic factors.

Effect of lactational high fat diet and metformin on mid-lactation murine mammary gland gene expression

Project description:Maternal health and diet can have important consequences for offspring nutrition and metabolic health. Signals are communicated from the mother to the infant during lactation through milk via macronutrients, hormones and bioactive molecules. In this study we designed experiments to probe the mother-milk-infant triad in the condition of normal maternal health and upon exposure to high fat diet (HFD) with or without concurrent metformin exposure. We examined maternal characteristics, milk composition and offspring metabolic parameters on postnatal day 16, prior to offspring beginning to wean. We found that lactational HFD increased maternal adipose tissue, mammary gland adipocytes, and altered milk lipid composition causing a higher amount of n-6 long chain fatty acids and lower n-3. Offspring of HFD dams were heavier with more body fat during suckling. Metformin exposure decreased maternal glucose and several amino acids. Offspring of met dams were smaller during suckling. Gene expression in the lactating mammary glands was impacted to a greater extent by metformin but both metformin and HFD altered genes related to muscle contraction, indicating that these genes may be more susceptible to lactational stressors. Our study demonstrates the impact of common maternal exposures during lactation on milk composition, mammary gland function and offspring growth with metformin having little capacity to recuse from the effects of a maternal HFD during lactation.
