ERdj5 protects goblet cells from endoplasmic reticulum stress-mediated apoptosis under inflammatory conditions

Project description:Endoplasmic reticulum stress is closely associated with the onset and progression of inflammatory bowel disease. ERdj5 is an endoplasmic reticulum-resident protein disulfide reductase that mediates the cleavage and degradation of misfolded proteins. Although ERdj5 expression is significantly higher in the colonic tissues of patients with inflammatory bowel disease than in healthy controls, its role in inflammatory bowel disease has not yet been reported. Hence, in the current study, we utilized ERdj5-knockout mice to investigate the potential roles of ERdj5 in inflammatory bowel disease. ERdj5 deficiency causes severe inflammation in mouse colitis models and weakens gut barrier function by increasing NF-κB-mediated inflammation. ERdj5 may not be indispensable for goblet cell function under steady-state conditions, while its deficiency induces goblet cell apoptosis under inflammatory stimuli.

IL-10 inhibition of miR-155 in inflammation

Project description:Inflammatory disorders including arthritis, inflammatory bowel disease, psoriasis and multiple sclerosis are characterised by the excessive production of pro-inflammatory cytokines resulting in inflammation, tissue damage and chronic pain. In a healthy immune system, the host has developed mechanisms to protect itself from excessive inflammation. One such mechanism is mediated by the anti-inflammatory cytokine IL-10, which primarily acts to limit the production of pro-inflammatory cytokines and antigen presenting capabilities of cells in the innate immune system. Since the discovery of IL-10, much emphasis has been placed on extrapolating its exact mechanism of inhibition with the aim of potentiating its use therapeutically. Significantly, the systemic administration of IL-10 has had success in many animal models of inflammation as well as clinical success in human disorders such as psoriasis and Crohn’s disease. Despite this, major gaps in our knowledge remain which have weakened the expected therapeutic effect for this anti-inflammatory cytokine. Our early studies discovered that in response to innate immune stimulation, IL-10 can potently inhibit miR-155 (McCoy et al., 2010), a miRNA characterised as a promoter of inflammation and found over-expressed in autoimmune pathologies. This finding suggests that the IL-10/miR-155 axis is a novel mechanism utilised by IL-10 to administer its function. In this study, an Affymetrix array was employed to characterise genes regulated by the IL-10/miR-155 axis in bone-marrow derived macrophages. This project proposal aims to characterise the mechanism of IL-10 inhibition on miR-155, as well as understand the functional outcome and significance of this effect by characterising genes regulated by the IL-10/miR-155 axis.

Whole blood gene expression from infliximab treated Crohn's disease patients at three time points: pre-treatment, two weeks and fourteen weeks post first treatment

Project description:Personalized treatment of complex diseases is an unmet medical need pushing towards drug biomarker identification of one drug-disease combination at a time. Here, we used a novel computational approach for modeling cell-centered individual-level network dynamics from high-dimensional blood data to predict infliximab response and uncover individual variation of non-response. We identified and validated that the RAC1-PAK1 axis is predictive of infliximab response in inflammatory bowel disease. Intermediate monocytes, which closely correlated with inflammation state, play a key role in the RAC1-PAK1 responses, supporting their modulation as a therapeutic target. This axis also predicts response in Rheumatoid arthritis, validated in three public cohorts. Our findings support pan-disease drug response diagnostics from blood, implicating common mechanisms of drug response or failure across diseases.
