Project description:Genus Halimeda is a worldwide distributed reef-building organism facilitated by fast growth and a rapid turnover rate in tropical and sub-tropical oceans. It is an excellent candidate for calcareous algae research because its calcification occurs extracellular, which deposits aragonite in the semi-enclosed inter-utricle space. In this study, we sampled the thalli of Halimeda macroloba in Sanya Bay of the South China Sea. The mean winter and summer water temperatures in Sanya Bay were 25.05℃ and 28.72℃, respectively. Additionally, the mean surface and bottom layer water temperatures in summer were 29.61℃ and 27.23℃. In order to simulate the natural temperature range, a factorial experimental design with three treatments (25°C, 28°C, and 31°C) was set. The goal of this study was to decipher the underlying traits that may facilitate H. opuntia to maintain high productivity and active physiology under different temperatures. Here, we employed iTRAQ-based quantitative proteomics and untargeted metabolomics approaches to investigate the protein and metabolite profiles of H. macroloba.

Project description:Chloropicrin (CP) can effectively combat soil-borne diseases but has significant side effects on nontarget microorganisms. The rhizosphere microflora play a crucial role in promoting plant growth and protecting plants from infection by soil-borne pathogens. We conducted a laboratory pot experiment to evaluate the effect of CP on the rhizosphere soil bacterial flora and the effect of biochar amendments on the reconstruction of microbial communities. Our results show that CP fumigation and biochar additions promoted the growth of cucumber plants in the later stage of the pot experiment. CP significantly inhibited the rhizobacterial diversity and changed the community composition. Biochar amendments after CP fumigation shortened the time for the rhizobacterial diversity to recover to unfumigated levels. Biochar amendments promoted the transplantation of new populations to empty microbiome niches that were caused by CP and, in particular, stimulated many beneficial microorganisms to become the predominant flora. The relative abundances of many functional taxa related to plant-disease suppressiveness and pollutant bioremediation increased, including Pseudomonas, Stenotrophomonas, Bacillus, Massilia, Acinetobacter, Delftia, Micromonospora, Cytophagaceae, and Flavisolibacter. These changes stimulated by biochar amendments would promote multifunctionality in the soil rhizosphere and benefit plant growth and disease resistance.

Project description:Lead (Pb)-polluted soils pose a serious threat to human health, particularly by transmitting this heavy metal to the food chain via the crops grown on them. The application of novel amendments in Pb-polluted soils can significantly reduce this problem. In this research, we report the effects of various organic and inorganic amendments i.e., bentonite (BN), biochar (BR), lignin (LN), magnesium potassium phosphate cement (CM) and iron hydroxyl phosphate (FeHP), on the Pb bioavailability in Pb-polluted soil, upon Pb distribution in shoots, roots, grain, the translocation factor (TF) and the bioconcentration factor (BCF) of Pb in pea (Pisum sativum L.) grain. Furthermore, effects of the said amendments on the plant parameters, as well as grain biochemistry and nutritional quality, were also assessed. Lead pollution significantly elevated Pb concentrations in roots, shoots and grain, as well as the grain TF and BCF of Pb, while reducing the nutritional quality and biochemistry of grain, plant height, relative water content (RWC), chlorophyll contents (chl a and chl b) and the dry weight (DW) of shoot, root and grain. The lowest Pb distribution in shoots, roots and grain were found with BN, FeHP and CM, compared to our control. Likewise, the BN, FeHP and CM significantly lowered the TF and BCF values of Pb in the order FeHP > CM > BN. Similarly, the highest increase in plant height, shoot, root and grain DW, RWC, chl a and chl b contents, grain biochemistry and the micronutrient concentrations, were recorded with BR amendment. Biochar also reduced grain polyphenols as well as plant oxidative stress. Given that the BR and BN amendments gave the best results, we propose to explore their potential synergistic effect to reduce Pb toxicity by using them together in future research.

Project description:Eggshell is a cheap and environmentally friendly calcium source. In this study, Ca-modified biochar adsorbents (CEA) were prepared by 1:10, 1:2, and 1:1 mass ratio of the eggshell and Eupatorium adenophorum. The CEA-2 sample prepared with a 1:2 mass ratio showed a maximum Pb adsorption capacity (97.74 mg·g-1) at the conditions of an initial pH of 7.0, an adsorbent dosage of 0.5 g·L-1, and a contact time of 8.0 h. The kinetic and isotherm studies indicated that the adsorption process of the CEA-2 sample had monolayer adsorption characteristics, which was controlled together by intraparticle and interface diffusion. Thermodynamic studies indicated that the adsorption process of CEA-2 was spontaneous (ΔG 0 <0) and endothermic (ΔH 0 > 0). X-ray diffraction and scanning electron microscopy analyses showed a uniform distribution of Ca-Pb precipitation on the CEA-2 surface, which proved that chemical precipitation was the main adsorption mechanism. Fourier transform infrared spectra found that CEA-2 had abundant active groups, especially nitrogen-containing functional groups, which could adsorb Pb through a surface complexation reaction. The Brunauer-Emmett-Teller surface area of CEA-2 was found to be 621 m2·g-1, and such developed pores could ensure the smooth diffusion of Pb. Finally, the effect of coexisting cation and anion experiment and the cyclic regeneration experiment indicated that CEA-2 had prominent stability and reusability for Pb adsorption.

Project description:A series of the magnetic CuFe2O4-loaded corncob biochar (CuFe2O4@CCBC) materials was obtained by combining the two-step impregnation of the corncob biochar with the pyrolysis of oxalate. CuFe2O4@CCBC and the pristine corncob biochar (CCBC) were characterized using XRD, SEM, VSM, BET, as well as pHZPC measurements. The results revealed that CuFe2O4 had a face-centered cubic crystalline phase and was homogeneously coated on the surface of CCBC. The as-prepared CuFe2O4@CCBC(5%) demonstrated a specific surface area of 74.98 m2·g-1, saturation magnetization of 5.75 emu·g-1 and pHZPC of 7.0. The adsorption dynamics and thermodynamic behavior of Pb(II) on CuFe2O4@CCBC and CCBC were investigated. The findings indicated that the pseudo-second kinetic and Langmuir equations suitably fitted the Pb(II) adsorption by CuFe2O4@CCBC or CCBC. At 30 °C and pH = 5.0, CuFe2O4@CCBC(5%) displayed an excellent performance in terms of the process rate and adsorption capacity towards Pb(II), for which the theoretical rate constant (k2) and maximum adsorption capacity (qm) were 7.68 × 10-3 g·mg-1··min-1 and 132.10 mg·g-1 separately, which were obviously higher than those of CCBC (4.38 × 10-3 g·mg-1·min-1 and 15.66 mg·g-1). The thermodynamic analyses exhibited that the adsorption reaction of the materials was endothermic and entropy-driven. The XPS and FTIR results revealed that the removal mechanism could be mainly attributed to the replacement of Pb2+ for H+ in Fe/Cu-OH and -COOH to form the inner surface complexes. Overall, the magnetic CuFe2O4-loaded biochar presents a high potential for use as an eco-friendly adsorbent to eliminate the heavy metals from the wastewater streams.
