Project description:The homozygous of mutants of Arabidopsis gene (AT1G31870) are lethal. The gene encodes a homolog of yeast BUD13 of RES complex, AtBUD13, which is involved in pre-mRNA splicing of target genes. To assay the introns characters of target genes of AtBUD13 in Arabidopsis thaliana, the siliques at 0, 1 and 2 days after pollination of wild type and heterozygous mutants were used for RNA-Seq.

Project description:This experiment aims at analyzing crossover distribution in male and female meiosis, in the Arabidopsis HEI10oe, zyp1, and zyp1 HEI10oe compared to wild type. Plants double heterozygous of zyp1-1+/- HEI10oe (Col) were crossed with zyp1-6+/- mutant plants (Ler) to generate zyp1-1/zyp1-6 HEI10oe heterozygous, HEI10oe heterozygous, zyp1-1/zyp1-6 and wild type in Col/Ler hybrid background, and then crossed as male or as female with wild-type Col. Backcross populations were grown in the greenhouse (16 h day/8 h night) for 3 weeks and 4 days in the dark, 100-150mg leave samples from plants from the 8 obtained BC1 populations were used for DNA purification and library preparation for Illumina sequencing (HiSeq 3000 2x150 bp), performed by the Max Planck-Genome-centre Cologne, Germany (https://mpgc.mpipz.mpg.de/home/).

Project description:Col-0 floral stem was grafted on the msh1 mutant (Col-0/msh1); on the dcl2,3,4,msh1 quadruple mutant (Col-0/dcl2,3,4,msh1); on Col-0 (Col-0/Col-0). Seeds were collected from the grafted Col-0 scion after grafts were established. Seed coming from the graft then were grown on the peat mix, leaf tissue was collected at the bolting and used for the total RNA sequencing.
