RNA-Seq analysis used to explore the bone-froming mechanism of microbially catalyzed biomaterials

Project description:We developed a microbial catalytic concept and strategy to prepare calcium carbonate with micro/nanostructures on the surface of bioceramics to improve bone-forming bioactivity. It involves three processes: bacterial adhesion on biomaterials, production of carbonate assisted with bacteria, nucleation and growth of calcium carbonate nano-crystals on the surface of bioceramics. The microbially catalyzed biominerals exhibited relatively uniform micro/nanostructures on both 2D and 3D CaSiO3 bioceramics. The descriptive analysis of RNA-sequencing revealed that the topographic and chemical cues presented by microbially catalyzed micro/nanostructures could stimulate the biological processes including adhesion, proliferation and differentiation. The study offers a microbially catalytic concept and strategy of fabricating micro/nanostructured biomaterials for tissue regeneration.

PURPOSE: This randomized clinical trial is studying two different doses of cholecalciferol to compare how well they work when given together with calcium carbonate in treating patients with colon cancer that has been removed by surgery.

Efficacy of Liming Forest Soil in the Context of African Swine Fever Virus.

Project description:Since September 2020, Germany has experienced the first ever outbreak of African swine fever (ASF). The first known cases occurred exclusively in wild boar in forest areas in Brandenburg and Saxony; in July 2021, infected domestic pigs were also confirmed for the first time. As wild boar are considered the main reservoir for the virus in the European region, an effective interruption of this infection chain is essential. In particular, the removal and safe disposal of infected carcasses and the direct disinfection of contaminated, unpaved ground are priorities in this regard. For the disinfection, highly potent as well as environmentally compatible disinfectants must be used, which are neither influenced in their effectiveness by the soil condition nor by increased organic contamination. Thus, in this study, slaked lime, milk of lime and quicklime (1% to 10% solutions) were selected for efficacy testing against the test virus recommended by the German Veterinary Society (DVG), Modified Vaccinia Ankara virus (MVAV), and ASF virus (ASFV) in conjunction with six different forest soils from Saxony in two different soil layers (top soil and mineral soil) each. In summary, 10% of any tested lime type is able to inactivate both MVAV and ASFV under conditions of high organic load and independent of the water content of the soil. At least a 4 log reduction of the virus titer in all tested forest soil types and layers and by all applied lime types was observed. In conclusion, the high efficacy and suitability of all tested lime products against both viruses and in the presence of high organic load in forest soil can be confirmed and will help to control ASF spread.

Characterization and quantification of chicken eggshell proteins during pivotal events of shell calcification

Project description:Gallus gallus avian eggshell is composed of 95% calcium carbonate on calcitic form and of 3.5% extracellular organic matrix (proteins, polysaccharides and proteoglycans). This highly ordered structure with a polycrystalline organization result of the control of mineral deposition by the organic matrix components in the lumen of the uterus. This interaction leads to the eggshell ultrastructure and consequently contributes to its resulting mechanical properties. This study used GeLC MS/MS analyses combined to label free quantitative analysis to identify and quantify matrix proteins at the pivotal step of the calcification process (amorphous calcium carbonate deposit, amorphous calcium carbonate transformation into calcite, large calcite crystal units and rapid growth phase). The study gave new insight on proteins playing crucial role in the biomineralization of the shell.

Effects of liming on forage availability and nutrient content in a forest impacted by acid rain.

Project description:Acidic deposition and subsequent forest soil acidification and nutrient depletion can affect negatively the growth, health and nutrient content of vegetation, potentially limiting the availability and nutrient content of forage for white-tailed deer (Odocoileus virginianus) and other forest herbivores. Liming is a mitigation technique that can be used to restore forest health in acidified areas, but little is known about how it affects the growth or nutrient content of deer forage. We examined the effects of dolomitic limestone application on the growth and chemical composition of understory plants in an acidified forest in central Pennsylvania, with a focus on vegetative groups included as white-tailed deer forage. We used a Before-After-Control-Impact study design with observations 1 year before liming and up to 5 years post-liming on 2 treated and 2 untreated 100-ha sites. Before liming, forage availability and several nutrients were below levels considered optimal for white-tailed deer, and many vegetative characteristics were related to soil chemistry. We observed a positive effect of liming on forb biomass, with a 2.7 fold increase on limed sites, but no biomass response in other vegetation groups. We observed positive effects of liming on calcium and magnesium content and negative effects on aluminum and manganese content of several plant groups. Responses to liming by forbs and plant nutrients show promise for improving vegetation health and forage quality and quantity for deer.
