This 2-year study employed RNA-sequencing technology to evaluate the pre- and post-vaccination muscle transcriptome of early-weaned Bos indicus-influenced beef calves born from dams offered different supplementation strategies from 57 ± 5 d prepartum until 17 ± 5 d postpartum.

Project description:The study examined changes in the transcriptional profile of skeletal muscle tissue in heifer calves offered varying planes of nutrition during the early life period. From approximately 3 weeks of age Angus X Holstein-Friesian heifer calves were offered either a high or moderate plane of nutrition to achieve target growth rates of 1.2 kg/d and 0.5 kg/d, respectively. After 18 weeks of differential feeding all calves were euthanized and skeletal muscle tissue samples collexted from each calf. RNA was extracted and subsequently subjected to RNAseq analysis. 482 genes were significantly differentially expressed between calves on the High plane of nutrition and those on the Moderate plane of nutrition. Biological pathway analysis revealed an enrichement from these genes for processes and pathways related to energy production and cellular growth and proliferation.

Project description:Fetal programming via maternal nutrition during gestation plays a key role in shaping skeletal muscle development, directly affecting meat quality and production efficiency. This study examines how maternal feed restriction at different gestational stages alters the expression of novel long non-coding RNAs (lncRNAs) in the skeletal muscle of goat offspring. LncRNAs are emerging as important regulators of biological processes, including myogenesis, yet their role in fetal programming remains largely unexplored. Using RNA sequencing (RNA-Seq), we analyzed lncRNA expression in the Longissimus thoracis muscle of kids born to goats exposed to two feeding regimens: early gestation restriction (50% of maintenance from day 8–84 of gestation, then 100% until parturition; RM, n=6) and late gestation restriction (100% from day 8–84, then 50% until parturition; MR, n=8). RNA-Seq data underwent quality control, alignment, transcript assembly, and differential expression analysis using Cufflinks and FEELnc pipelines. Three differentially expressed (DE; FDR ≤ 0.05) lncRNAs were identified and classified based on genomic context and potential regulatory function. The upregulated lncRNA_10996.1 in the RM group was linked to ENSCHIG00000020055 and ENSCHIG00000024132. Downregulated lncRNA_1180.1 and lncRNA_7992.1 were associated with ZNF546, PSMC4, SNORA65, ZNF79, LRSAM1, and ENSCHIG00000023017. These results suggest that maternal nutrition-induced shifts in lncRNA expression may influence muscle development through regulatory interactions with protein-coding genes. Our findings identify candidate lncRNAs as potential biomarkers and contribute novel insights into the molecular mechanisms underlying fetal programming in livestock.

Project description:Compairsion of transcriptional profiles of heart and skeletal muscle tissue of fetal rhesus monkey exposed to maternal Bisphenol A or vehicle during early or late gestaion. Maternal exposure to the endocrine disrupting chemical, bisphenol A (BPA) affects the development of multiple organ systems in rodents and monkeys. However, effects of BPA exposure on cardiac and skeletal muscle development have not been assessed. Given that maternal BPA crosses placenta and reaches developing fetus, examining the physiological consequences of gestational exposure during development is of research significance. Therefore, we evaluate the effects of daily, oral BPA exposure of pregnant rhesus monkeys (Macaca mulatta) on the fetal heart and skeletal muscle transcriptome. Pregnant monkeys were administered daily oral doses (400 M-BM-5g/kg body weight) of BPA during early (50 M-bM-^@M-^S100 M-BM-1 2 days post conception, dpc) or late (100 M-BM-1 2 dpc - term), gestation. At the end of treatment, fetal heart tissues; left ventricle (LV), right ventricle (RV), left atrium (LA), right atrium (RA) and skeletal muscle; biceps femoris (BFM), were collected. Transcriptome expression was assessed using genome-wide microarray in each of the tissues and compared paired-wise between the BPA and matched control fetuses. Our results show that maternal BPA exposure alters transcriptional profile of several coding and non-coding genes in fetal heart and skeletal muscle. A total of 12 samples were analyzed for each tissue; LV, RV, LA, RA and BFM. This includes 6 samples at each time period (early vs. late gestation) and 3 biological replicates for each treatment (BPA, n=3; control, n=3).

Project description:We used an RNA-Seq transcriptomic approach to investigate Longissimus dorsi muscle gene expression profiles of calves from cows receiving weekly Se-yeast boluses at supranutritional concentrations during different trimesters of gestation. Pregnant beef cows received, except the control group (CTR), weekly supranutritional selenium-yeast boluses (105 mg Se/wk) during the first (TR1), second (TR2), or third (TR3) trimester of gestation. Within 12 to 48 h of birth, muscle samples were collected from the Longissimus dorsi using a Bergstrom biopsy needle (inner diameter: 5 mm). Muscle biopsies were placed into sterile vials, stored on ice and frozen at – 80 ºC until total RNA isolation was performed. After sequencing and read quality control, we identified 3,048 unique differentially expressed genes (DEGs) across all group comparisons (FDR < 0.05 and |log2FC| > 1.5). Furthermore, we predicted 237 unique transcription factors that putatively regulate the DEGs. Our findings suggest a beneficial effect of supranutritional maternal organic Se supplementation during late gestation on Se-status and muscle development and function of newborn calves.

Project description:We evaluated if a higher plane of maternal nutrition during late gestation and weaning age alters the offspring’s Longissimus muscle (LM) transcriptome. A microarray analysis was performed in LM samples of early (EW) and normal weaned (NW) Angus × Simmental calves born from cows that were grazing endophyte-infected tall fescue/red clover pastures with no supplement (low plane of nutrition (LPN)), or supplemented with 2.3 kg of dried distiller’s grains with solubles and soyhulls (70% DDGS/30% soyhulls) (medium plane of nutrition (MPN)) during the last 90 days of gestation. Biopsies were harvested at 78, 187 and 354 days of age. Bioinformatics analysis highlighted that offspring transcriptome did not respond markedly to cow plane of nutrition, resulting in only 13 differentially expressed genes. However, weaning age and a high-starch diet strongly impacted the transcriptome, especially the immediate activation of the lipogenic program in EW steers. In addition, between 78 and 187 days of age, these animals had an activation of the innate immune system due presumably to macrophage infiltration in intramuscular fat. Between 187 and 354 days of age (i.e. the fattening phase), NW steers had an activation of the lipogenic transcriptome machinery, while EW steers had a clear gene transcription inhibition. The latter appears to have occurred through the epigenetic control of histone acetylases, which were down-regulated. Higher cow plane of nutrition alone affected 35 genes in the LM of steers that underscore the presence of a mechanism of macrophage infiltration likely originating from localized oxidative stress as a result of increased levels of hypoplasia and hypertrophy in LM.

Project description:This study evaluated how the rate of maternal body weight gain in F0 dams during early gestation affects gene expression in the skeletal muscle of F1 offspring and their F2 fetuses. During the first 84 days of gestation, we applied a nutritional plan to the F0 dams to promote two different rates of body weight gain (0.28 kg/day and 0.79 kg/day). We tested the hypothesis that maternal nutrition aiming at two rates of body weight gain (low [LG] or moderate [MG] gain) during early gestation would affect the gene expression of skeletal muscle in both the F1 offspring and the subsequent F2 generation. Thus, our aim was to measure the gene expression profiles and identify differentially expressed genes (DEG), metabolic pathways (KEGG), and biological processes (GO BP) in the skeletal muscle of F1 heifers at birth, as well as during the early gestation phase, applying the same approach to F2 fetuses.

Project description:We hypothesized that poor maternal nutrition during gestation would reduce the growth and development of offspring muscle prenatally, reduce the number of myogenic progenitor cells, and result in changes in the global expression of genes involved in prenatal muscle development and function. Ewes were fed a control (100% NRC)-, restricted (60% NRC),- or over-fed (140% NRC) diet beginning at day 30 of gestation until day 45, 90, and 135 of gestation or until parturition. In offspring, we report altered secondary: primary muscle fiber ratios and percent PAX7 positive cells during fetal development. In addition, changes in muscle mRNA expression were observed between maternal dietary treatments and over timepoints during gestation. In conclusion, poor maternal nutrition during gestation contributes to altered offspring muscle growth during early fetal development which persists throughout the fetal stage.
