Project description:We found that western diet consumption resulted in decrease in the percentage of normal Paneth cell population in wild type mice, indicating that western diet negatively regulates Paneth cell homeostasis. We performed transcriptional profiling to identify molecular mechanisms associated with Paneth cell defect in wild type mice exposed to western diet. Male mice were used at 4-5 weeks of age, and were fed with either standard or western diet. The whole ileum of the mice were harvested for transcriptomic analysis after completing 8 weeks of diet consumption.

Project description:We found that mainstream cigarette smoking (4 cigarettes/day, 5 days/week for 6 weeks using Kentucky Research Cigarettes 3R4F) resulted in >20% decrease in the percentage of normal Paneth cell population in Atg16l1 T300A mice but showed minimal effect in wildtype littermate control mice, indicating that Atg16l1 T300A polymorphism confers sensitivity to cigarette smoking-induced Paneth cell damage. We performed transcriptional profiling to identify molecular mechanisms associated with Paneth cell defect in Atg16l1 T300A mice exposed to cigarette smoking. Female mice were used at 4-5 weeks of age. Cigarette smoking was performed using smoking chamber with the dosage and schedule as described above. The whole ileum of the mice were harvested for transcriptomic analysis after completing 6 weeks of smoking.

Proteomic Analysis of Full-Thickness Rotator Cuff Tears: Potential Role of Tetranectin in Synovial Fluid

Project description:Rotator cuff tears are one of the most common shoulder disorders and can lead to pain, weakness, and limited mobility. Although the clinical condition has been widely studied, its molecular mechanisms are still not fully understood. In this study, we analyzed the proteomic profile of synovial fluid in patients with full-thickness rotator cuff tears and compared the results to those from a control group. Proteomic analysis was performed on synovial fluid samples from seven patients with full-thickness tears and three patients with partial-thickness tears, who served as the control group. Label-free quantification using the MaxQuant platform identified 284 proteins at 1% FDR. Among them, CLEC3B (tetranectin) showed the most significant difference, with a fold change of approximately 40 and a p-value of 2.6 × 10⁻³. Gene ontology enrichment analysis revealed biological processes related to extracellular matrix remodeling, fibrinolysis, and inflammation. Given its known roles in extracellular matrix remodeling, tetranectin identified as a candidate for full-thickness rotator cuff tears, providing insight into pathological processes such as tendon degeneration and fatty infiltration.
