Project description:In this project we have applied palaeoproteomics to study archaeological shell beads dating back to the beginning of the 7th mill. cal BCE and recovered from a prehistoric site in Jordan (Ba'ja). These beads were made of mollusc shells and were of unknown origin, but suspected to be made of larger bivalves such as Tridacna or Spondylus sp. The aim of the study was to analyse ancient proteins extracted from these beads in order to infer their biological origin. Tiny amounts of ancient samples were used and the extracted proteins were analysed by high resolution liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS). In order to identify obtained peptides, we have created an ‘in-house’ molluscan protein database, by gathering molluscan sequences (proteins and transcriptomes), which were downloaded from publicly available databases in NCBI. Searching against this comprehensive database, we were able to identify a number of shell proteins from Tridacna sp., the giant clam shells. Our study reports the oldest molluscan protein sequences ever recovered that come from a very warm environment.

Project description:Nacre, the iridescent material found in pearls and shells of molluscs, is formed through an extraordinary process of matrix-assisted biomineralization. Despite recent advances, many parts of the biomineralization process and its evolutionary origin remain a mystery. The pearl oyster Pinctada fucata martensii is a well-known master of biomineralization, but the molecular mechanisms underlie its production of remarkable shells and pearls is not fully understood. We sequenced the highly polymorphic genome of the pearl oyster and conducted multi-omic and biochemical studies to probe nacre formation. We identified a large set of novel proteins participating in matrix-framework formation, many in expanded families, including components similar to that found in vertebrate bones such as collagen-related VWA-containing proteins (VWAP), chondroitin sulfotransferases and regulatory elements.Considering that there are only collagen-based matrices in vertebrate bones and chitin-based matrices in most invertebrate skeletons, the presence of both chitin and elements of collagen-based matrices in nacre matrices suggests that elements of chitin- and collagen-based matrices are deeply rooted and might be part of an ancient biomineralizing matrix. Our results expand the current shell matrix-framework model and provide new insights into the evolution of diverse biomineralization systems.

Project description:BackgroundThe Mollusca constitute one of the most morphologically and ecologically diverse metazoan phyla, occupying a wide range of marine, terrestrial and freshwater habitats. The evolutionary success of the molluscs can in part be attributed to the evolvability of the external shell. Typically, the shell first forms during embryonic and larval development, changing dramatically in shape, colour and mineralogical composition as development and maturation proceeds. Major developmental transitions in shell morphology often correlate with ecological transitions (e.g. from a planktonic to benthic existence at metamorphosis). While the genes involved in molluscan biomineralisation are beginning to be identified, there is little understanding of how these are developmentally regulated, or if the same genes are operational at different stages of the mollusc's life.ResultsHere we relate the developmental expression of nine genes in the tissue responsible for shell production - the mantle - to ecological transitions that occur during the lifetime of the tropical abalone Haliotis asinina (Vetigastropoda). Four of these genes encode evolutionarily ancient proteins, while four others encode secreted proteins with little or no identity to known proteins. Another gene has been previously described from the mantle of another haliotid vetigastropod. All nine genes display dynamic spatial and temporal expression profiles within the larval shell field and juvenile mantle.ConclusionThese expression data reflect the regulatory complexity that underlies molluscan shell construction from larval stages to adulthood, and serves to highlight the different ecological demands placed on each stage. The use of both ancient and novel genes in all stages of shell construction also suggest that a core set of shell-making genes was provided by a shared metazoan ancestor, which has been elaborated upon to produce the range of molluscan shell types we see today.

Project description:In this study we conducted proteomics analyses of historical and contemporary Bombyx mori cocoon shells to case-study the human-driven introduction and diversification of this species in Europe. We employed LC-MS/MS analyses protocols for 81 cocoon shell samples to identify that the cocoon shell of this species contains on average 98±13 (Mean±SD) proteins, Our proteomic datasets provide a valuable foundation for advancing further exploitations of silkworm cocoon shells in multiple scientific perspectives.

Project description:Molluscan shells are diverse, beautiful, and mechanically robust. They are commonly used during ancient human life as money, decorative objects, or functional tools. To reconstruct their origin is important for us to understand their production process as well as underlying cultural contexts. Luodian, a unique shell art in China since 3000 years ago, has served as a cultural carrier between China and Europe. This study combined proteomics and microstructural analysis to identify the raw materials origin of three Luodian samples from Qing Dynasty with species resolution and potential age levels. Shell proteomics and multiple sequence alignment have identified key shell matrix proteins (N66 and Pif) to be of Pinctada maxima, a pearl oyster distributed in the indo-pacific regions. After establishing that the thickness of nacreous tablets in the shell is inversely proportional to the shell size for the first time, we proposed to infer the age of raw materials in different Luodian samples, which are tailored to different purposes. This study provides a framework and foundation for identifying the origin of various shell arts especially made from pearl oysters.
