Pollutants bioavailability and toxicological risk from microplastics to marine mussels

Project description:Microplastics represent a growing environmental concern for the oceans due to their potential capability to adsorb different classes of pollutants, thus representing a still unexplored source of exposure for aquatic organisms. In this study polystyrene (PS) microplastics were characterized for their capability to adsorb pyrene (PYR) as model compound for polycyclic aromatic hydrocarbons, and transfer this chemical to filter feeding mussels Mytilus galloprovincialis. Gene expression analyses of Mytilus galloprovincialis exposed to polystyrene (PS) microplastics and to polystyrene contaminated with pyrene (PS-PYR) have been performed trough a DNA microarray platform.

Polyethylene Microplastics Breach Chorion Protection, Triggering Cardiotoxicity Across Aquatic Models

Project description:Polyethylene microplastics are pervasive environmental pollutants that pose potential risks to aquatic organisms. This study investigates the effects of polyethylene microplastics on zebrafish (Danio rerio) embryo development, with a focus on whether the chorion, a protective barrier surrounding the embryo, is effective in blocking polyethylene penetration. Contrary to previous findings that suggested the chorion could prevent larger microplastics (>0.7 µm) from entering, our study demonstrates that polyethylene particles sized 1-4 µm can still negatively impact embryo development without dechorionation. Embryos were exposed to polyethylene at concentrations of 0.01, 0.1, and 1 mg/L, followed by RNA sequencing to assess gene expression changes. Despite no significant differences in survival, hatching, or body length between control and treated groups, a significant reduction in heart rate was observed at higher concentrations, indicating potential sub-lethal cardiotoxicity. Further, qRT-PCR validation confirmed significant downregulation of key heart development-related genes, particularly fbln1 and fn1b, in polyethylene-exposed embryos. Additionally, polyethylene exposure also impacted Daphnia magna, reducing heart rate and delaying growth. These findings highlight the ability of polyethylene microplastics to penetrate natural barriers such as the chorion and induce physiological and developmental changes. Our results emphasize the need for further research into the long-term effects of microplastic exposure on aquatic ecosystems.

We included 4 experimental groups of mice; chow diet, chow diet and microplastics, high-fat/fructose/cholesterol diet, high-fat/fructose/cholesterol diet and microplastics. Following tissue collection after treatment, bulk RNA sequencing was carried out to profile transcriptomic activity of each group.

Nitrosomonas europaea expression analysis of biofilm growth and biofilms exposed to phenol and toluene

Project description:The physiological and transcriptional response of Nitrosomonas europaea biofilms to phenol and toluene was examined and compared to suspended cells. Biofilms were grown in Drip Flow Biofilm Reactors under continuous flow conditions of growth medium containing ammonia as growth substrate. The responses of N. europaea biofilms to the aromatic hydrocarbons phenol and toluene were determined during short-term (3 h) additions of each compound to the biofilms. Ammonia oxidation in the biofilms was inhibited 50% by 60 uM phenol and 100 uM toluene. These concentrations were chosen for microarray analysis of phenol- and toluene-exposed N. europaea biofilms. Liquid batch cultures of exponentially growing N. europaea cells were harvested alongside the biofilms to determine differential gene expression between attached and suspended growth of N. europaea.

Low-dose exposure to microplastics retards meiotic maturation via HDACs insufficiency

Project description:Microplastics (MPs) are considered as one of the main reasons for male and female infertility. However, the reproductive toxicity and its related mechanisms are understood by animal models with acute exposure to MPs at present. In the study, we show the low-dose polystyrene microplastics (PSMPs) exposure results in severely abnormal reproduction in female, but not male in mouse model, exhibiting failed oocyte meiotic maturation. Mechanistically, the PSMPs exposure induces the over-activation of cell metabolism pathways, insufficient HDACs and H4K16 hyperacetylation in oocytes in vivo and in vitro. By addition of HDAC3 inhibitor, the failed oocyte maturation, over-activation of cell metabolism pathways and H4K16 hyperacetylation are recapitulated, and the overexpression of HDAC3 can rescue the defects of meiotic maturation induced by PSMPs. Our observations suggest a direct link of the maturation defects induced by PSMPs to HDAC3 insufficiency. Thus, we propose the potential treatments for therapy of the failed meiotic maturation of oocyte from women highly exposed to MPs by activating or supplying HDAC3.

Effect of the PET microplastics on pancreas - in vivo study on pig

Project description:Microplastics are a relatively newly discovered environmental hazard that can contribute to the disruption of many physiological processes in the organism. There is evidence that they affect the physiology of the pancreas, but research is still very limited. Therefore, the aim of the study was to determine the effects of PET microplastics on the global proteomic profile of the porcine pancreas using LC-MS/MS analysis. The pigs were treated with a low (0.1 g/day) or a high dose (1 g/day) of PET microplastics for 4 weeks. The analysis revealed that PET microplastics affected protein expression in a dose-dependent manner - the low dose affected the abundance of 7 proteins, while the high dose of 17.

RNA sequencing of CT26 colorectal cancer xenograft tumors following intratumoral exposure to polystyrene microplastics

Project description:Microplastics are emerging environmental pollutants with potential biological effects on human health. However, the molecular responses of colorectal cancer tissues following microplastic exposure remain poorly characterized. In this study, we performed bulk RNA sequencing to investigate transcriptomic alterations associated with polystyrene microplastics (PS-MPs) exposure in a murine colorectal cancer model. CT26 colorectal cancer cells were subcutaneously implanted into BALB/c mice to establish xenograft tumors. After tumor formation, mice received daily intratumoral injections of 1 μm PS-MPs suspension (10 μg/mL, 50 μL/day), while control mice received no PS-MPs treatment. Tumor tissues were collected for RNA extraction and high-throughput sequencing. Differential gene expression analysis and downstream bioinformatic analyses were performed to characterize molecular changes associated with PS-MPs exposure. This dataset provides transcriptomic profiles of CT26 colorectal cancer xenograft tumors following PS-MPs exposure and may facilitate further investigation of the biological responses associated with environmental microplastic exposure.
