Global gene expression analysis of resistant and susceptible apple cultivars challenged with Penicillium expansum

Project description:Blue mold, caused by Penicillium expansum, is responsible for postharvest losses of apple fruit, and threatens human health through production of the potent mycotoxin patulin. No major gene(s) providing resistance have as yet been identified, but recent studies indicate a quantitative control of the disease. An AryANE chip covering 60K apple transcripts was used to identify possible candidate gene(s) that are differentially regulated between resistant and susceptible cultivars upon P. expansum infection. Induction of cell wall related gene (PGIP1), and three genes involved in the â€˜down-streamâ€™ flavonoid biosynthesis pathway (CHS, FLS and LDOX), shows the fundamental role of cell wall as an important barrier, and contents of polyphenolic compounds of fruits as a quantitative components in enhancing disease resistance to blue mold. Moreover, exogenous application of Jasmonic acid hormone enhanced the defense mechanism in fruits. This is the first report linking Jasmonic acid and activation of cell wall and flavonoid pathway genes in apple fruit resistance to blue mold. Results provide an initial categorization of genes that are potentially involved in the resistance mechanism, and should be useful for developing tools for gene marker-assisted breeding of apple cultivars with an improved resistance to blue mold. Comparative transcriptome analysis of a lowly virulent strain of Erwinia amylovora in shoots of two apple cultivars susceptible and resistant to fire blight.

Project description:The aim of our study was to decipher differences in the response of a lowly virulent E. amylovora strain to infection of susceptible and resistant apple cultivars at the transcriptome level. For this purpose, we applied an RNA-seq technique to see the global changes in gene expression of E. amylovora while interacting with two apple cultivars at two time points after inoculation of shoots. Additionally, we compared transcriptomes of E. amylovora growing on a microbiological medium and in planta to elucidate transcriptional changes in bacterial cells induced by the host plant environment.

Identification of allergenomic signatures in allergic and well-tolerated apple genotypes using LC-MS/MS

Project description:The apple fruit (Malus domestica L. Borkh) is one of the most popular fruits worldwide. Beyond their beneficial properties, apples contain proteins that trigger allergic reactions in susceptible consumers. Mal d1 to 4 are allergens present in a variety of different isoforms in apples, with Mal d1 and Mal d3 being the main allergens. In this study, we used proteomics to quantify all four Mal d proteins in 52 apple genotypes with varying allergenic potentials. We revealed the expression of at least 21 Mal d1 isoforms while two Mal d3 and two Mal d4 proteins were found among the genotypes. The allergenic potential of the Mal d isoforms was characterized by comparing isoform abundance with allergenic scores of genotypes (from oral challenge tests). The Mal d peptides detected by comparative analysis presumably have different IgE binding properties and could therefore be used as potential molecular markers to discriminate between hypoallergenic and hyperallergenic cultivars.
