Project description:Traumatic muscle injury associated with volumetric muscle loss results in reversible muscle regeneration and function. Bottlenecks to conventional tissue engineering strategies to augment muscle regeneration are limited by low cell transplantation capacity by conventional injection of dissociated cells, the lack of customization of engineered tissues based on muscle defect geometry, as well as the feasibility of intra-operative use. Here, we developed a facile approach to engineer modular units of scaffold-free high-density muscle tissues in customizable geometric shapes and sizes with integration potential. For the first time, we describe aspiration-assisted intra-operative bioprinting of scaffold-free modular units for preclinical treatment of volumetric muscle loss. We demonstrated the ability of 3D modular muscle units to be bioprinted to the site of the muscle defect, leading to improved muscle, vascular, and neuromuscular regeneration in a mouse model of volumetric muscle loss. Transcriptional profiling revealed that the scaffold-free muscle units enable pre-formed cell-cell interactions that support myogenesis, unlike dissociated cells that lack cell-cell interactions. This technology of intraoperative modular muscle unit implantation is promising for clinical use for precise bioprinting of scalable tissues from geometrically tunable modular units.

Project description:The human skin has a highly developed extracellular matrix (ECM) that is vital for proper skin functioning, its 3D architecture playing a pivotal role in support and guidance of resident and invading cells. To establish relevant in vitro models mimicking the complex design observed in vivo, scaffold-based and scaffold-free 3D cell culture systems have been developed. Here we show that scaffold-free systems are well suited for the analysis of ECM protein regulation. Using quantitative mass spectrometry-based proteomics in combination with magnetic 3D bioprinting we characterize changes in the proteome of skin fibroblasts and squamous cell carcinoma cells. Transferring cells from 2D to 3D without any additional scaffold induces a profound upregulation of matrisome proteins indicating the generation of a complex, tissue-like ECM.

Project description:Glioma is a highly aggressive form of brain cancer, with some subtypes having 5-year survival rates of less than 5%. Tumour cell invasion into the surrounding parenchyma seems to be the primary driver of these poor outcomes, as most gliomas recur within 2 cm of the original surgically-resected tumour. Many current approaches to the development of anticancer therapy attempt to target genetic weaknesses in a particular cancer, but may not take into account the microenvironment experienced by a tumour and the patient-specific genetic differences in susceptibility to treatment. Here we demonstrate the use of complementary approaches, 3D bioprinting and scaffold-free 3D tissue culture, to examine the invasion of glioma cells into neural-like tissue with 3D confocal microscopy. We found that, while both approaches were successful, the use of 3D tissue culture for organoid development offers the advantage of broad accessibility. As a proof-of-concept of our approach, we developed a system in which we could model the invasion of human glioma cells into mouse neural progenitor cell-derived spheroids. We show that we can follow invasion of human tumour cells using cell-tracking dyes and 3D laser scanning confocal microscopy, both in real time and in fixed samples. We validated these results using conventional cryosectioning. Our scaffold-free 3D approach has broad applicability, as we were easily able to examine invasion using different neural progenitor cell lines, thus mimicking differences that might be observed in patient brain tissue. These results, once applied to iPSC-derived cerebral organoids that incorporate the somatic genetic variability of patients, offer the promise of truly personalized treatments for brain cancer.

Project description:The study of muscle dystrophies, including Duchenne and Myotonic dystrophies, has faced significant challenges due to the lack of physiologically relevant in vitro models. This research introduces a novel, scaffold- and biomaterial-free platform based on cell sheet engineering to create in vitro models using patient-specific cells. This innovative approach, known as anchored cell sheet engineering, offers a comprehensive biofabrication method that replicates mature cell phenotypes and tissue-specific extracellular matrix (ECM) deposited by the cells themselves. Primary cells from both healthy individuals and patients with Duchenne and Myotonic dystrophy type 1 were used to develop robust anchored 3D muscle fibers. The models were extensively characterized using proteomics analysis revealing that they form mature constructs closely resembling in vivo conditions, surpassing traditional 2D cultures in translational potential. Disease-specific models effectively captured key phenotypic features of the respective dystrophies. Additionally, therapeutic treatments led to significant proteomic profile changes, documented using custom Python scripts. This scaffold-free 3D modeling technique offers a promising platform for advancing muscle dystrophy research and improving drug screening methodologies for developing new treatments for complex diseases with better translatability for clinical applications.

Project description:Development of systems allowing the maintenance of native properties of mesenchymal stromal cells (MSC) is a critical challenge for studying physiological functions of skeletal progenitors, as well as towards cellular therapy and regenerative medicine applications. Conventional stem cell culture in monolayer on plastic dishes (2D) is associated with progressive loss of functionality, likely due to the absence of a biomimetic microenvironment and the selection of adherent populations. Here we demonstrate that 2D MSC expansion can be entirely bypassed by culturing freshly isolated bone marrow cells within the pores of 3D scaffolds in a perfusion-based bioreactor system, followed by enzymatic digestion for cell retrieval. The 3D-perfusion system supported MSC growth while maintaining cells of the hematopoietic lineage, and thus generated a cellular environment mimicking some features of the bone marrow stroma. As compared to 2D-expansion, sorted CD45- cells derived from 3D-perfusion culture after the same time (3 weeks) or a similar extent of proliferation (7-8 doublings) maintained a 4.3-fold higher clonogenicity and exhibited a superior differentiation capacity towards all typical mesenchymal lineages, with similar immunomodulatory function in vitro. Transcriptomic analysis performed on MSC from 5 donors validated the robustness of the process and indicated a reduced inter-donor variability as well as a significant upregulation of multipotency-related gene clusters following 3D-perfusion as compared to 2D expansion. The described system offers a model to study how factors of a 3D engineered niche may regulate MSC function and, by streamlining conventional labor-intensive processes, is prone to automation and scalability within closed bioreactor systems. Nucleated cells were isolated from 5 fresh human bone marrow aspirates by means of red blood cells lyses buffer and then were seeded into a 3D perfusion bioreactor system using a pure hydroxyapatite 3D scaffold and in conventional Petri dishes (2D). After culture for 19 days, cells from both systems were enzymatically retrieved and sorted using anti-CD45-coated magnetic beads. Total RNA was extracted from CD45- cells, QCed and hybridized to Affymetrix microarrays.
