Project description:The experiment aims to identify the response of the mature nodules and denodulated roots of Medicago truncatula /Sinorhizobium medicae md4 to the systemic signaling of plant water deficit. The root systems of nodulated symbiotic plants were divided into two compartments. The N source of N2 fixing plants was exclusively the air. To apply the water stress, we treated half root system with PEG while the other was kept on control conditions. To characterize the systemic responses of the transcriptome to the PEG treatment , nodules and roots of the non-treated half root system were harvested and total RNA isolated for RNAseq analysis. We performed a time-course (6 hours, 1-, 3- and 5-days treatment) .

Project description:The experiment aims to identify the response of mature nodules and denodulated roots of Medicago truncatula /Sinorhizobium medicae md4 to the systemic signaling of plant N deficit. The root systems of nodulated symbiotic plants were divided into two compartments. The N source of N2 fixing plants was exclusively the air. To reduce the plant nitrogen (N) supply (N-deficient conditions), we flushed one half root system with Ar/O2 (80/20 v/v) while the other half root system was kept on control conditions (Air). To characterize the systemic responses of the transcriptome to plant N deficit , nodules and roots of the non-treated half root system were harvested and total RNA isolated for RNAseq analysis. We performed a time-course (6 hours, 1-, 3- and 5-days treatment).

Time-course transcriptome analysis of Sinorhizobium medicae (MD4 strain) in mature nodules of Medicago truncatula grown in split-root under different nitrogen (N) supply

Project description:The experiment aim is the identification of the transcriptional response of mature nodules of Medicago truncatula inoculated with the strain MD4 of Sinorhizobium medicae in response to different nitrogen (N) supply (N-satiety and N-deficient conditions). We performed a time-course (1 and 3 days treatment) split-root experiment to identify plant systemic responses: the root systems of nodulated symbiotic plants were divided into two compartments, one of them treated and the other kept on control conditions. Nodules were isolated from the non-treated side of the split root and total RNA isolated for RNAseq analysis. To analyse Sinorhizobium medicae MD4 expression, a ribosomal RNA depletion protocol was performed before library preparation.

Project description:affy_med_2013_05 - affy_med_2013_05 - The experiment is designed to detect transcriptome differences between wild type and two fix minus (NF737 and MS240) mutant lines.-Medicago truncatula WT (R108) and two mutant lines (NF737 and MS240/dnf2) were nodulated using WSM419 Sinorhizobium medicae symbiotic bacteria. Plants were grown in vitro on BN medium and the nodules harvested 14 days post inoculation (dpi). RNA was extracted using the plant RNA extraction kit from Thermo Scientific followed by a DNAse I treatment (TurbO DNAse I Ambion).

Project description:affy_popsec_nancy_pophydro_roots_poplar - This project aims to identify genes of interest for control of root growth in response to water deficit in a tree species: poplar. We look for genes and gene expression networks related to drought stress. We intend to analyse the transcriptome in root apices of cuttings grown in hydroponics and osmotically stressed with PEG. Root apex is the location of root elongation and these analyses intend to identify genes involved in the control of cell expansion and thus of root elongation. Indeed, root growth maintenance in response to water shortage contributes to plant tolerance to water deficit.-Poplar cuttings (cv Soligo) were grown in hydroponics. A moderate water deficit was applied by adding PEG to the nutrient solution (200g/l). After 3 days, the apex (1 cm-long) of the roots of each cutting were collected. For control and stressed treatments, RNAs were extracted from two pools of 5 to 9 roots (issued from 2 to 5 cuttings). A pool is considered as one biological replicate and corresponds to one Affymetrix slide. Keywords: treated vs untreated comparison 4 arrays - poplar
