Project description:Inflammatory periodontal disease (periodontitis) is widespread in dogs. This study aimed to evaluate site-specific changes in the canine gingival crevicular fluid (GCF) proteome during the longitudinal progression from very mild gingivitis to mild periodontitis. Periodontitis diagnosis in dogs requires anaesthesia, our ultimate aim was to develop a periodontitis diagnostic that could be applied to samples taken from conscious dogs. The objective of this work was to identify potential biomarkers of periodontal disease progression in the GCF of dogs.

Project description:Single-cell transcriptomics were used to comprehensively resolve the osteoimmunology microenvironment that are involved in periodontal tissues from human chronic periodontitis before and after nonsurgical periodontal therapy, as well as clinically healthy controls. This allows unbiased assessment of many heterogeneous cells at the single-cell level, hence revealing complexities of the molecular components and differences with counterparts residing in periodontal tissues.

Single-cell RNA sequencing quantitative analysis of PBMC transcriptomes from dogs with atopic dermatitis and healthy dogs

Project description:We used Drop-seq and next generation sequencing to determine gene expression differences in dogs with atopic dermatitis and healthy dogs in peripheral blood mononuclear cells in an unbiased way. Using Seurat, we find 13 discrete immune cells clusters, including a cluster enriched for Gata3 expressing T cells with 95 differentially expressed genes between healthy and allergic dogs.

Proteomic analysis of cerebrospinal fluid in epileptic and healthy dogs

Project description:This study included four dog groups (group A: 10 healthy dogs, group B: 9 dogs with idiopathic epilepsy receiving antiepileptic medication-AEM, group C: 8 dogs with idiopathic epilepsy without AEM administration and group D: 7 dogs with seizures due to structural brain abnormalities). The epileptic dogs were allocated into these 3 groups after a diagnostic investigation that included laboratory testing, thoracic radiographic and abdominal ultrasonographic examination, brain imaging, CSF routine and proteomic analysis. The purpose of the current study was to investigate and compare the proteomic profile among the four groups of dogs.

Project description:The aim of the study was to analyse differences in serum and urinary proteomes between healthy dogs and dogs with renal dysfunction in babesiosis using label-based high-resolution quantitative proteomic approach. In the study 8 healthy control dogs (group H) and 22 dogs with naturally occurring babesiosis were enrolled. Dogs with babesiosis were divided into 3 groups: group A consisted of 6 non-azotaemic dogs (serum creatinine <140 μmol/L) with normal UPC (UPC < 0.5), group B of 10 non-azotaemic dogs with proteinuria (UPC > 0.5) and group C of 6 azotaemic dogs (serum creatinine > 140 μmol/L) and significant proteinuria (UPC > 2).

Circulating microRNA profiling by array in dogs with myxomatous mitral valve degeneration (MMVD) and pulmonic stenosis (PS) compared with healthy dogs

Project description:In this study, we aimed to demonstrate expression profiles of circulating microRNAs (miRNAs) in dogs with eccentric or concentric cardiac hypertrophy, and investigate whether there is a difference in miRNA expression according to the type of cardiac hypertrophy. Dogs with myxomatous mitral valve degeneration (MMVD) or pulmonic stenosis (PS) were included in this study, which are the two representative diseases of eccentric or concentric cardiac hypertrophy in dogs, respectively. Circulating miRNAs were isolated from the serum samples of five dogs with MMVD, five dogs with PS, and five healthy dogs. The circulating miRNA expression levels of dogs with MMVD or PS were compared with those of the healthy dogs by microarray analysis (Affymetrix GeneChip miRNA 4.0), using two independent parameters, a fold change cut-off of > 1.5 (up or down regulation) and p-value of < 0.05.

Expression data from uninephrectomized dogs compared to sham operated dogs

Project description:Renal insufficiency is a risk factor for development of cardiovascular disease. In this study our aim was to investigate functional and structural changes in the heart of dogs with mild renal insufficiency induced by uninephrectomy (UNX). In addition to comprehensively assessing functional parameters 12 week after UNX or sham surgery, we harvested tissue for assessment of structural changes. Microarray analyses were performed on snap frosen tissue from the left atrium (LA) and left ventricle (LV) of UNX and sham operated animals. Gene expression in LA was compared between the groups, and gene expression in the LV was compared between the groups. We aimed to assess whether a potentially altered gene expression in the heart of the UNX group also included genes associated with cardiac remodeling. Six dogs were assigned to unineprectomy and 6 dogs were assigned to sham operation. Microarray analyses were performed on 5 samples from the left atrium of uninephrectomized animals and on 5 samples from the left atrium of sham operated animals. Further, microarray analyses were performed on 5 samples from the left ventricle of uninephcretomized animals and on 5 samples from the left ventricle of sham operated animals. No replicates were perfomed.

Dogs that progressed during, or after, treatment and were ultimately euthanized had tissue collected from primary and metastatic tumors to analyze gene expression and assess mechanisms of resistance to this therapy.
