Eggs of the mosquito Aedes aegypti survive desiccation by rewiring their polyamine and lipid metabolism

Project description:Upon water loss, some organisms pause their life cycles and escape death. While widespread in microbes, this is less common in animals. Aedes mosquitoes are vectors for viral diseases. Aedes eggs can survive dry environments, but molecular and cellular principles enabling egg survival through desiccation remain unknown. In this report, we find that Aedes aegypti eggs, in contrast to Anopheles stephensi, survive desiccation by acquiring desiccation tolerance at a late developmental stage. We uncover unique proteome and metabolic state changes in Aedes embryos during desiccation that reflect reduced central carbon metabolism, rewiring towards polyamine production, and enhanced lipid utilization for energy and polyamine synthesis. Using inhibitors targeting these processes in blood-fed mosquitoes that lay eggs, we infer a two-step process of desiccation tolerance in Aedes eggs. The metabolic rewiring towards lipid breakdown and dependent polyamine accumulation confers resistance to desiccation. Furthermore, rapid lipid breakdown is required to fuel energetic requirements upon water re-entry to enable larval hatching and survival upon rehydration. This study is fundamental to understanding Aedes embryo survival and in controlling the spread of these mosquitoes.

Gene trapping in the Asian malaria vector, Anopheles stephensi

Project description:A piggyBac transposon-based gene trap element was transformed into the Asian malaria vector, Anopheles stephensi, and remobilized using the jumpstarter approach using genetic crosses. Individuals that displayed a gene trap remobilization event were then photodocumented and their RNA and DNA complements were extracted. The DNA compelement was used to determine the genomic insertion site, while the RNA was used to determine the transcript coverage of the genes into which the transposons inserted. In nearly half of the cases, insertion was identified to fall within introns present in the 5'-UTR of transcripts- which are not indicated in the current ab initio models for Anopheles stephensi. The ability to utilize next generation RNA-Seq elucidated the functionality of the gene trap elements that inserted outside of the ab initio gene models, providing clear evidence that not only was the gene trap element working properly, but that it also had a bias towards 5'-end insertion, in particular, 5'-UTR intronic insertion. RNA from 200 pooled individually-extracted An. stephensi that demonstrated a gene trap remobilizable event.

Salinity adaptation in the principal arboviral vector Aedes aegypti characterized by larval cuticle changes

Project description:Aedes aegypti, the principle global vector of arboviral diseases, has been widely regarded to only lay eggs and undergo preimaginal development in fresh water collections. Recent observations however show that it has adapted to develop in anthropogenic brackish water habitats of up to 50% sea water in coastal areas in different continents. This adaptation is characterised by greater salinity tolerance in adult oviposition preference, larvae and changes in sizes of anal papillae. The physiological basis for salinity tolerance in either Ae. aegypti or any of the known salinity-tolerant species of Anopheles malaria vectors is not established. To address this knowledge gap which is of fundamental biological interest and important for control of major diseases we performed RNAseq analysis of gut, anal papillae, and rest of the carcass of Ae. aegypti collected in the field from brackish water (BW) and fresh water habitats (FW) and then maintained as laboratory colonies in BW and FW respectively. We also examined the cuticle structure of larvae, pupae and adult BW and FW Ae. aegypti by microscopy and performed proteomic analysis of the shed cuticles of fourth instar larvae (L4) when they transformed into pupae. The results show that major changes in cuticle structure and composition characterize, and may be the principal factor that permits, the adaptation of Ae. aegypti to brackish water.

The Hsf1-sHsp cascade has pan-antiviral activity in mosquitoes

Project description:Aedes mosquitoes transmit pathogenic arthropod-borne (arbo) viruses, putting nearly half the world’s population at risk. Blocking virus replication in mosquitoes rather than in humans serves as a promising approach to prevent arbovirus transmission, which requires in-depth knowledge of mosquito immunity. By integrating multi-omics data, we identified that heat shock factor 1 (Hsf1) regulates eight small heat shock protein (sHsp) genes within one topological associated domain. This Hsf1-sHsp cascade acts as an early response against chikungunya virus (CHIKV) infection and shows pan-antiviral activity in three vector mosquitoes, Aedes aegypti, Aedes albopictus, and Anopheles gambiae. We then assessed the baseline expression of sHsp genes in different tissues of female Ae. aegypti using RNA-seq, and we observed a highly dynamic expression pattern of sHsp genes that varied dramatically across different tissues. Interestingly, sHsp genes were expressed at low levels in two main barrier tissues, the midgut and salivary glands, compared to other tissues such as the crop. Importantly, activation of Hsf1 led to a reduced CHIKV infection rate in adult Ae. aegypti mosquitoes, demonstrating Hsf1 as a promising target for the development of novel intervention strategies to limit arbovirus transmission by mosquitoes.
