Project description:Due to climate change and increasing anthropogenic activities, lakes are disturbed frequently, usually by press (e.g., diffused pollution, rising temperatures) or pulse (e.g., storms, rainfall, pollution events) disturbances. Both press and pulse disturbances can affect abiotic and biotic environments, changing the structure of ecosystems and affecting ecosystem services. To confront with the effects of climate change and increasing anthropogenic activities, understanding the different effects of press and pulse disturbances on lake ecosystems is essential. This study assessed the effect of press and pulse disturbances of phosphorus on a microcosmic aquatic ecosystem by measuring the total phosphorus (TP), algae density, and physiological indicators of submerged macrophytes. We found that the microcosmic aquatic ecosystem responded differently to press and pulse disturbances. Our results suggested that it had a lower resistance to pulse phosphorus disturbances than to press phosphorus disturbances. There were significantly higher nutrient concentrations and algal densities in the pulse treatment than in the press treatment. Positive feedback was found between the biomass of submerged macrophytes and the water quality. There was a higher submerged macrophytes biomass at low TP concentration and algal density. In the context of climate change, press and pulse disturbances could have severe impacts on lake ecosystems. Our findings will provide some insight for further research and lake management.

Comparison of DNA and RNA, and cultivation approaches for the recovery of terrestrial and aquatic fungi from environmental samples.

Project description:Estimates of fungal biodiversity from environmental samples are all subject to bias. Major issues are that the commonly adopted cultivation-based approaches are suitable for taxa which grow readily under laboratory conditions, while the DNA-based approaches provide more reliable estimates, but do not indicate whether taxa are metabolically active. In this study, we have evaluated these approaches to estimate the fungal diversity in soil and freshwater samples from a subtropical forest, and compared these to RNA-based culture-independent approach intended to indicate the metabolically active fungal assemblage. In both soil and freshwater samples, the dominant taxon recovered by all three approaches was the same (Anguillospora furtiva). This taxon was cultivable from all samples and comprised 85-86 % DNA libraries and 90-91 % RNA libraries. The remaining taxa were phylogenetically diverse and spanned the Ascomycota, Basidiomycota, and Fungi incertae sedis. Their recovery was not consistent among the three approaches used and suggests that less abundant members of the assemblage may be subjected to greater bias when diversity estimates employ a single approach.

Microcosm experiment combined with process-based modeling reveals differential response and adaptation of aquatic primary producers to warming and agricultural run-off.

Project description:Fertilizers, pesticides and global warming are threatening freshwater aquatic ecosystems. Most of these are shallow ponds or slow-flowing streams or ditches dominated by submerged macrophytes, periphyton or phytoplankton. Regime shifts between the dominance of these primary producers can occur along a gradient of nutrient loading, possibly triggered by specific disturbances influencing their competitive interactions. However, phytoplankton dominance is less desirable due to lower biodiversity and poorer ecosystem function and services. In this study, we combined a microcosm experiment with a process-based model to test three hypotheses: 1) agricultural run-off (ARO), consisting of nitrate and a mixture of organic pesticides and copper, differentially affects primary producers and enhances the risk of regime shifts, 2) warming increases the risk of an ARO-induced regime shift to phytoplankton dominance and 3) custom-tailored process-based models support mechanistic understanding of experimental results through scenario comparison. Experimentally exposing primary producers to a gradient of nitrate and pesticides at 22°C and 26°C supported the first two hypotheses. ARO had direct negative effects on macrophytes, while phytoplankton gained from warming and indirect effects of ARO like a reduction in the competitive pressure exerted by other groups. We used the process-based model to test eight different scenarios. The best qualitative fit between modeled and observed responses was reached only when taking community adaptation and organism acclimation into account. Our results highlight the importance of considering such processes when attempting to predict the effects of multiple stressors on natural ecosystems.
