Project description:The chemical characteristics (proximate composition, amino acids, and fatty acids) and sensory quality of the longissimus lumborum (LL) muscle of 45 farmed male fallow deer were investigated. The animals were divided into three separate groups (n = 15 per treatment): pasture-fed (P), pasture-fed and supplemented with barley (B), and pasture-fed and supplemented with barley and lysine (BL). Differences were observed in LL moisture and the intramuscular fat contents, the latter being almost two-fold greater in the meat of B and BL groups compared to P. The concentrations of histidine, leucine, alanine, glutamic acid and glycine in the raw meat were higher in the BL group compared to the P group. Higher contents of n-3 polyunsaturated fatty acids (PUFAs), and consequently lower n-3 ratios, were found in the P group, compared to the BL group. The grilled meat samples from the P group scored higher than the other groups for grassy flavour, and lower for liver flavour.

Project description:Purpose: The goal of this study is to compare (RNA-seq) transcriptomes of in vitro cultured human bone marrow-derived mesenchymal stem cells (hMSCs) and fallow deer antler-derived skeletal progenitors (FD RM Cells) under multiple conditions to identify candidate proliferation and mineralization genes responsible for fast antler regeneration Methods: hMSCs and FD RM Cells were cultured in vitro under 1) serum-free (0% serum) or serum (10% serum) conditions for 2.5 days or 2) Control (0 ng/mL BMP-2 and 0 nM dexamethasone) and osteogenic (100 ng/mL BMP-2 and 100 nM dexamethasone) media for 24 days. mRNA profiles were generated by deep sequencing, in duplicate, using Illumina HiSeq 2000. The sequence reads were analyzed at the transcript isoform level STARS followed by Cufflinks. Validation for genes of interest was performed using immunofluorescence staining. Results: Comparison of human and fallow deer skeletal progenitor datasets yielded proliferation and mineralization gene candidates Conclusions: Our study represents the first detailed analysis of human and fallow deer transcriptomes of skeletal progenitor cells under proliferation and mineralization conditions, with biologic replicates, generated by RNA-seq technology. Our in vitro comparative approach circumvent some of the logistical and technical challenges in identifying candidate proliferation and mineralization genes responsible for rapid deer antler regeneration. We conclude that in vitro comparison of RNA-seq based transcriptomes identified candidate proliferaiton and mineralization genes to advance bone biology and holds promise to rapidly regenerate large bone volumes for regenerative medicine. The comparative approach utilized here can be adapted for almost any tissue to study a specific phenomenon of interest.

Project description:PurposeSarcocystis spp. are protozoan parasites of livestock which also infect birds, lower vertebrates and mammals, including man. Wild and domestic ruminants such as red deer, roe deer, fallow deer, cattle, sheep and goats may act as intermediate hosts for many Sarcocystis species, some of which are significant pathogens causing sarcocystosis in livestock and humans. The purpose of the present study was to determine the prevalence of Sarcocystis species in fallow deer farmed in an open pasture system.MethodsSamples of heart and oesophagus tissue taken from five fallow deer were examined by light microscope for the presence of sarcocysts. Genomic DNA was extracted from individual sarcocysts. ssu rRNA was successfully amplified using their DNA as templates.ResultsAnalysis of the ssu rRNA identified the presence of two S. morae sarcocysts in the heart tissue; similarly, S. gracilis sarcocysts were identified in the heart and oesophagus, and Sarcocystis sp. most closely related to S. linearis and S. taeniata were detected in oseophagus.ConclusionsThese findings confirm the presence of Sarcocystis spp. in farmed fallow deer in Poland; however, more molecular studies are needed.

Project description:BackgroundAssortative mating can help explain how genetic variation for male quality is maintained even in highly polygynous species. Here, we present a longitudinal study examining how female and male ages, as well as male social dominance, affect assortative mating in fallow deer (Dama dama) over 10 years. Assortative mating could help explain the substantial proportion of females that do not mate with prime-aged, high ranking males, despite very high mating skew. We investigated the temporal pattern of female and male matings, and the relationship between female age and the age and dominance of their mates.ResultsThe peak of yearling female matings was four days later than the peak for older females. Younger females, and especially yearlings, mated with younger and lower-ranking males than older females. Similarly, young males and lower-ranking males mated with younger females than older males and higher-ranking males. Furthermore, the timing of matings by young males coincided with the peak of yearling female matings, whereas the timing of older male matings (irrespective of rank) coincided with the peak of older female matings.ConclusionsAssortative mating, through a combination of indirect and/or direct female mate choice, can help explain the persistence of genetic variation for male traits associated with reproductive success.

Project description:BackgroundAnimals living in social groups can benefit from conducting the same behaviour as other group members. If this synchronisation is achieved by copying the behaviour of other individuals, we would expect synchrony to be more likely when pairs of individuals are close together.ResultsBy comparing the behaviour of a focal individual with its nearest, second nearest and third nearest neighbour and a control individual, we show that pairings of fallow deer Dama dama are more likely to be active or inactive at the same moment in time if they are closer together. We also demonstrate that synchronisation in the group happens more often than would be expected by chance.ConclusionsOur findings suggest that there is a relationship between the synchronisation of behaviour and the spatial proximity of individuals. Spatial proximity is likely to be an important influence on how likely individuals are to be synchronised, although care needs to be taken to separate social and environmental influences on individual behaviour.

Project description:Inter-individual differences are necessary for selection to act, while plasticity (intra-individual variation) may buffer against selection. Sleep is a critical self-maintenance behaviour but, unlike most behaviours, the causes and consequences of its inter- and intra-individual variation in wild animals is poorly understood, particularly in neonates where sleep plays a key role in development. We have shown previously that free-ranging neonate fallow deer (Dama dama) differ in sleep during the first few weeks of life. Here, we test whether individual variability in sleep is organised systematically across the population, and whether these individual differences are associated with chronic stress measured using hair cortisol, or the timing of birth. Four dimensions of sleep behaviour (total sleep time, sleep fragmentation, sleep quality, and sleep distribution over 24-h) were quantified using state-of-the-art triaxial accelerometers. We then used a multivariate mixed-effects model in a Bayesian framework to evaluate covariation between multiple dimensions of sleep behaviour, and quantify the relative importance of chronic stress and the timing of birth, while accounting for the confounding effects of environmental conditions and age. We found that the timing of birth and chronic stress were not associated with changes in sleep between individuals. While both total sleep time and the number of bouts per day declined with age, their rate of development covaried, but no other sleep dimensions covaried. Our results represent an in-depth analysis of natural variation in sleep, and show that individual differences in four aspects of sleep architecture in free-living fallow deer fawns are strong but independent of one another and unrelated to chronic stress or the timing of birth. We suggest that covariation between sleep dimensions might emerge later in life and effects of cortisol and birth timing might be very short and transient.

Project description:(1) Background: Wild cervids play an important role in transmission cycles of tick-borne pathogens; however, investigations of tick-borne pathogens in sika deer in Germany are lacking. (2) Methods: Spleen tissue of 74 sympatric wild cervids (30 roe deer, 7 fallow deer, 22 sika deer, 15 red deer) and of 27 red deer from a farm from southeastern Germany were analyzed by molecular methods for the presence of Anaplasma phagocytophilum and Babesia species. (3) Results: Anaplasma phagocytophilum and Babesia DNA was demonstrated in 90.5% and 47.3% of the 74 combined wild cervids and 14.8% and 18.5% of the farmed deer, respectively. Twelve 16S rRNA variants of A. phagocytophilum were delineated. While the infection rate for A. phagocytophilum among the four cervid species was similar (71.4% to 100%), it varied significantly for Babesia between roe deer (73.3%), fallow deer (14.3%), sika deer (27.3%) and red deer (40.0%). Deer ≤2 years of age tested significantly more often positive than the older deer for both A. phagocytophilum and Babesia species. (4) Conclusions: This study confirms the widespread occurrence of A. phagocytophilum and Babesia species in wild cervids and farmed red deer in Germany and documents the co-occurrence of the two tick-borne pathogens in free-ranging sika deer.
