Project description:The goals of this project are to use a liquid chromatography-tandem mass spectrometer (LC-MS/MS) to detect bacterial proteomic profiles of Acinetobacter baylyi ADP1 and their quantitative protein responses under exposure to 30 mg/L of artificial sweeteners (saccharine, sucralose, aspartame, and acesulfame potassium). The control group without artificial sweeteners exposure was also set up for comparison. Each treatment was conducted in biological triplicate. By comparing the proteomic profiles of artificial sweeteners-treated groups and control group, the effects of artificial sweeteners on bacterial translational levels can be revealed.

Project description:The goals of this study are to use Next-generation sequencing (NGS) to detect bacterial mRNA profiles of wild-type Acinetobacter baylyi ADP1 and Pwh1266 plasmid, and their mRNA response under the exposure of four artificial sweeteners, including saccharin, sucralose, aspartame, and acesulfame potassium. 3 mg/L of each sweetener was used to treat the mixture of bacteria cell and plasmid. The group without dosing any artificial sweeteners was the control group. Each sample was conducted in triplicate. By comparing the mRNA profiles of experimental groups and control group, the effects of these four artificial sweeteners on the transcriptional levels can be revealed. Illumina HiSeq 2500 was applied. The NGS QC toolkit (version 2.3.3) was used to treat the raw sequence reads to trim the 3’-end residual adaptors and primers, and the ambiguous characters in the reads were removed. Then, the sequence reads consisting of at least 85% bases were progressively trimmed at the 3’-ends until a quality value ≥ 20 were kept. Downstream analyses were performed using the generated clean reads of no shorter than 75 bp. The clean reads of each sample were aligned to the Acinetobacter baylyi ADP1 reference genome (NC_005966) using SeqAlto (version 0.5). Cufflinks (version 2.2.1) was used to calculate the strand-specific coverage for each gene, and to analyze the differential expression in triplicate bacterial cell culture. The statistical analyses and visualization were conducted using CummeRbund package in R (http://compbio.mit.edu/cummeRbund/). Gene expression was calculated as fragments per kilobase of a gene per million mapped reads (FPKM, a normalized value generated from the frequency of detection and the length of a given gene.

Project description:We systematically investigated the responses of five human gut microbiomes to 21 common sweeteners, using an approach combining high-throughput ex vivo microbiome culturing and metaproteomics to quantify functional changes in different taxa. Hierarchical clustering based on metaproteomic responses of individual microbiomes resulted in two clusters. The first cluster was composed of non-caloric artificial sweeteners (NAS) and two sugar alcohols with shorter carbon backbones (4-5 carbon atoms), and the second cluster was composed of sugar alcohols with longer carbon backbones. The metaproteomic functional responses of the second cluster were similar to the prebiotic fructooligosaccharides and kestose, indicating that these sugar alcohol-type sweeteners have potential prebiotic functions.

Project description:BackgroundRecent epidemiological cohort studies have suggested that consumption of artificial sweeteners (AS) is associated with adverse cardiovascular events and mortality. However, these population association studies cannot establish a causal relationship. In this study we investigated the effect of long-term (1-year) consumption of AS (Equal and Splenda, two commonly used AS) on cardiovascular health and survival in rats.MethodsAdult Sprague-Dawley rats (both sexes, 4-5 months old) were randomized into the following 3 groups: control (n = 21), AS Equal (n = 21) and Splenda (n = 18). In the AS groups, Equal or Splenda was added to the drinking water (2-packets/250 ml), while drinking water alone was used in the control rats. The treatment was administered for 12 months. Cardiovascular function and survival were monitored in all animals.ResultsIt was found that rats in the AS groups consistently consumed more sweetened water than those in the control group. AS did not affect body weight, non-fasting blood cholesterol, triglycerides, blood pressure or pulse wave velocity. There were no significant differences in left ventricular wall thicknesses, chamber dimension, cardiac function or survival. AS did not affect heart rate or atrial effective refractory period. However, rats in both Equal and Splenda groups had prolonged PR intervals (63 ± 5ms in Equal, 68 ± 6 ms in Splenda, vs 56 ± 8 ms in control, p < 0.05) and a tendency of increased atrial fibrillation inducibility.ConclusionLong-term consumption of AS does not affect cardiovascular structure, function or survival but may cause some electrophysiological abnormalities with prolonged PR intervals and a tendency of increased atrial fibrillation inducibility in rats.

Project description:The prevalence of obesity is increasing, and the origins of obesity and metabolic dysfunction may be traced back to fetal life. Currently, overweight pregnant women are advised to substitute sugar-sweetened beverages with diet drinks containing artificial sweeteners. Recent evidence suggests that the consumption of artificial sweeteners during pregnancy increases the risk of obesity in the child, but the mechanism is unknown. We hypothesized the transportation of artificial sweeteners across the placenta into the fetal circulation and the amniotic fluid. We included 19 pregnant women who were given an oral dose of acesulfame, cyclamate, saccharin, and sucralose immediately before a planned caesarean section. Nine women were included as controls, and they refrained from an intake of artificial sweeteners. The maternal and fetal blood and amniotic fluid were collected during the caesarean section, and concentrations of artificial sweeteners were measured using mass spectrometry. We found a linear relationship between the fetal plasma concentrations of artificial sweeteners and the maternal plasma concentrations, with adjusted coefficients of 0.49 (95% CI: 0.28-0.70) for acesulfame, 0.72 (95% CI: 0.48-0.95) for cyclamate, 0.51 (95% CI: 0.38-0.67) for saccharin, and 0.44 (95% CI: 0.33-0.55) for sucralose. We found no linear relationship between amniotic fluid and fetal plasma concentrations, but there were positive ratios for all four sweeteners. In conclusion, the four sweeteners investigated all crossed the placenta and were present in the fetal circulation and amniotic fluid.

Project description:Background/objectivesArtificial sweeteners are commonly used food additives that provide sweetness without calories. Once considered harmless due to their lack of metabolism, recent studies suggest that they may have unintended effects, potentially stimulating appetite and increasing food intake, leading to weight gain. This study aimed to assess consumer perceptions of artificial sweeteners in food, examine consumption frequencies of products containing them, and explore their potential influence on body mass index.MethodsA cross-sectional study was conducted using two voluntary and anonymous surveys administered via Google Forms.ResultsThe study included 649 participants: 324 parents of preschool and school-aged children and 325 university and secondary school students. A substantial proportion of parents (59.3%) recognized artificial sweeteners as common sugar substitutes in beverages like juices, soft drinks, and protein drinks. Awareness was notably higher among students (88.9%). While most participants held a negative attitude toward artificial sweeteners, their awareness and engagement with food label reading were low. Multivariate linear regression identified significant associations: Male gender (β = 1.17, p < 0.001) and older age (β = 0.42, p < 0.001) were associated with higher BMI. Additionally, participants who rarely or never consumed carbonated soft drinks had a lower BMI (β = -1.48, p = 0.039), while those who occasionally consumed snacks had a higher BMI (β = 0.51, p = 0.039).ConclusionsThis research underscores the urgent need for public health initiatives addressing misconceptions, raising food label reading practices, while encouraging healthier consumption habits through educational campaigns. Additionally, the study's insights will help assess the potential cumulative health impacts of artificial sweetener intake.
