Project description:Green tea catechins are known as antioxidants beneficial to human health, but they are also defensive compounds produced by tea plants to protect them from herbivores. However, some herbivores can cope with green tea catechins and parasitize tea plants. We investigated the adaptation mechanisms of the spider mite Tetranychus kanzawai, an important arthropod pest of tea plants, to the defensive functions of green tea catechins. The degree of adaptation of spider mites to tea plants was correlated with the intra- and interspecific differences in their behavioral and xenobiotic responses to green tea catechins.

Project description:Metabolic disorders, such as obesity and type 2 diabetes, are major public health concerns worldwide. Dietary interventions, such as tea consumption, have been suggested as an effective strategy to prevent and treat metabolic disorders. White adipose tissue, as the main energy storage organ in mammals, plays a critical role in the regulation of whole-body metabolism. Recent studies have shown that the microenvironmental cell composition and metabolic network of white adipose tissue can be modulated by dietary factors, including tea consumption. However, the underlying mechanisms and the effects of tea consumption on white adipose tissue in the context of high-fat diet-induced metabolic disorders are not fully understood. Therefore, this study aimed to investigate the effects of tea consumption on the microenvironmental cell composition and metabolic network of white adipose tissue in high-fat diet-fed mice.

Project description:The effects of increasing addition of green tea in dietary changes the bacterial populations in broiler ileum were evaluated. Four hundreds of AA broilers were randomly assigned to four groups with green tea addition of 0, 0.5, 1 and 2 percent in the diet. The body weight showed no difference but a digital increase positively correlated with addition of green tea. The content of green tea had a linear effect of lengthening the ileum villi. The barcoded DNA pyrosequencing method was used to reveal 15 phyla, 1157phylotypes and 3098 16S operational taxonomic units (OTUs). The most predominant bacterial phyla were Firmicutes (56.89%), Actinobacteria (30.58%), Proteobacteria (8.61%) and Bacteroidetes (2.72%). As the proportion of additional green tea increased, the abundance of phylum Actinobacteria (p=0.003) and Proteobacteria (p=0.049) almost linearly increased, while the proportion of Firmicutes (p=0.027) linearly decreased. Only 2 OTUs were significantly affected by the increased additive, Corynebacteriaceae (p=0.011) and Staphylococcaceae (p= 0.006). Triplot analysis suggested that the dominant phyla of Verrucomicrobia, TM7 and Actinobacteria were clearly related to the addition of green tea. Moreover, green tea addition influenced the construction of microbiota, and lengthened the villus in ileum by Monte Carlo permutation test. These findings provide a new understanding of the ileal microbial ecology, which may be useful in modulating the gut microbiome, and also the proper usage of powdered green tea.

Project description:Down syndrome is the main genetic cause of intellectual disability and is due to triplication of human chromosome 21 (HSA21). Green tea extracts containing epigallocatechin-3-gallate (green tea) improve cognition both in mouse models and individuals with Down syndrome. We here analyzed the proteome and phosphoproteome alterations in a Down syndrome mouse model, the partial trisomic Ts65Dn mice, and the effect produced by the green tea extract and environmental enrichment (EE). Trisomic hippocampi presented a dysregulated proteome, especially when looking at the phosphorylation level in cognitive-related categories (synaptic proteins, neuronal projection, neuron development, microtubule), and GTPases/kinase activity and chromatin related categories. Green tea, EE, and their phospholipids in the plasma membrane and regulates signal transduction pathways, transcription factors, DNA methylation, mitochondrial function and phosphorylation, and autophagy to exert many of its beneficial biological actions Of interest for DS, it inhibits the activity of the Dual Specificity Tyrosine-Phosphorylation-Regulated Kinase 1A (DYRK1A), a DS candidate gene located in the 21q22.2 human chromosome region4,5. Previous work from our group showed that EGCG partially rescues the effects of overexpression of a DS candidate gene, DYRK1A, on the proteome and phosphoproteome of the hippocampus of TgDyrk1A mice6. However, the extent to which these mechanisms apply to a trisomy scenario is unknown. To get insight in these mechanisms we analyzed changes in protein abundances and phosphorylation in Ts65Dn mice, and their disomic counterparts in baseline conditions and upon three treatments known to improve cognition in Ts65Dn: i) green tea extract containing EGCG, ii) environmental enrichment (EE), and iii) their combination.

Project description:Purpose: Microarray technologies provide a unique opportunity to deeply investigate bacterial molecular responses to treatments. Pseudomonas syringae pv. actinidiae (Psa) is the causal agent of the bacterial canker of kiwifruit causing severe economic losses worldwide. At present, integrated control strategies include chemical treatments with copper-based products and preventive measures but the high virulence and fast spreading of the bacterium are hardly controlled by such measures, and especially copper use is questioned because of the possible appearance of copper resistant bacterial strains. The present project aims at the identification of Psa responses to green tea treatment (Gunpowder variety) at sub-lethal concentration (0.4 mg/ml). Methods: Psa cells were cultured in liquid KB (controls) or in KB supplemented with Gunpowder tea (Gunpowder-trateted) at 0.4 mg/ml EGCG for 24 h at 28°C. The microarray experiments on Gunpowder treated or untreated samples in biological triplicate resulted in 6 samples to be analyzed. Conclusions: This work identified important molecular mechanisms involved in Psa responses upon Gunpowder green tea treatment.

In this study, broccoli-supplemented diet induced changes in the gut microbiome and its role in host and bacterial gene expression were examined in mice following consumption of the Total Western Diet (TWD), which is based on NHANES data and represents the composition of a typical American diet. C57BL/6 male mice were fed a TWD for six weeks followed by a supplementation of 0, 0.5, 1 or 2.5% broccoli powder for three weeks. Microbial communities from cecal contents were taxonomically profiled using 16S and metagenomics sequencing, and metatranscriptomics was used to assess functionality of bacterial species. Cecum tissues were also analyzed for host transcriptomics.

Project description:We found that western diet consumption resulted in decrease in the percentage of normal Paneth cell population in wild type mice, indicating that western diet could negatively affect Paneth cell function. Subsequent generations of western diet consumption further reduced percentages of normal Paneth cell population. We performed fecal microbiota composition profiling. Male mice were used at 4-5 weeks of age. Fecal samples were collected for microbiome analysis.
