The protective effect of hydrocinnamic acid on Crohn’s disease.

Project description:Polyphenols are promising therapeutics for Crohn’s disease (CD), yet their bioavailability is largely dependent on the gut microbiome. The bioactive metabolites, metabolic pathways, and anti-inflammatory mechanisms underlying their effects remain poorly defined. Through integrated multi-omics and dietary analysis, we identified hippuric acid as a reduced metabolite in CD patients and a biomarker of polyphenol–microbiome interactions. We further demonstrate that hydrocinnamic acid, derived from microbiome metabolism of dietary trans-cinnamic acid and subsequently converted to hippuric acid by the host, ameliorates colitis in mice via the PPAR-γ pathway. Notably, we identified Bifidobacterium breve Bbr60 and Escherichia coli FAH as novel hydrocinnamic acid producers, complementing the established pathway in Clostridium sporogenes and suggesting additional microbial contributors. Collectively, our findings establish hydrocinnamic acid as a diet-dependent, colon-produced, mild PPAR-γ agonist with the potential to minimize adverse effects associated with traditional PPAR-γ agonists. This work establishes a foundation for developing microbiota-targeted, polyphenol-based therapeutic strategies for CD patients.

Plasma exosomes miRNAs high-throughput sequencing in patients after STEMI 3-6 months

Project description:ST-segment elevated myocardial infarction (STEMI) is one of the most severe forms of cardiovascular heart diseases. In the present study, we aim to describe differential expressed plasma exosome-miRNAs in patients with post-STEMI 3-6 months. Methods: Consecutive patients with ages from 40 to 80 years and 25 males among them 3-6 months after STEMI (n=11) were compared to sex-matched healthy control (n=10). The mean age of the patient was 64.71±10.40 years and 89.2% were male. Compared to the healthy control group, the plasma exosome-miRNAs were assessed by using microarray assay (IIIumnia Hiseq 2500). Profile of plasma exosome-miRNAs related to heart diseases was established both in the patient and control groups. The specificity of the lowest expressed exosome-miRNAs for all subjects was evaluated by using a quantitative real-time polymerase chain (qPCR). plasma exosomes miRNAs were obtained from STEMI patients after 3-6 months and healthy subjects

Characterization of the platelet transcriptome by RNA sequencing in patients with acute myocardial infarction.

Project description:Recent technological advances have made transcriptome sequencing (RNA-seq) possible in cells with low RNA copy number including platelets. Resulting studies have used RNA-seq in platelets isolated from healthy individuals to characterize the platelet transcriptome. However, platelets, possibly through gene expression changes, contribute to the etiology of and response to cardiovascular disease and events. To address this, we performed the largest human platelet RNA-seq analysis to date in 34 platelet samples: 16 ST-segment elevation myocardial infarction (STEMI), 16 non-STEMI (NSTEMI), and 2 controls.
