Project description:Understanding the molecular basis of plant heat tolerance helps to predict the consequences of climate warming on plant performance, particularly in vulnerable environments. Our current understanding comes primarily from studies in Arabidopsis thaliana (Arabidopsis) and select crops exposed to short and intense periods of heat stress. In this study we sought to characterise the transcriptomic responses of Wahlenbergia ceracea (W. ceracea), an Australian alpine herb, to growth under sustained moderate warming. We compared responses of pre-defined tolerant and sensitive lines, based on measures of photosynthetic thermostability, to growth under cool (24/15 ºC day/night) or warm (30/20 ºC day/night) temperatures to identify the pathways involved in heat tolerance and acclimation. Under warmer growth temperatures, W. ceracea up-regulated genes involved in RNA metabolism, while down-regulating those involved in photosynthesis and pigment metabolism. In tolerant lines, genes related to photosystem II and light-dependent photosynthetic reactions were more strongly down-regulated. This suggests that the regulation of electron transport and its components may be involved in thermal acclimation. Our results also highlight the importance of hormonal gene networks, particularly those responsive to ethylene, during longer-term moderate warming. In conclusion, our results point to post-transcriptional processes and the stabilisation of the electron transport chain as candidate mechanisms for thermal acclimation in heat-tolerant W. ceracea. The study also revealed many temperature-responsive genes with no detectable ortholog in Arabidopsis, whose characterization may enhance our understanding of physiological acclimation and have relevance for the biotechnological improvement of threatened species and crops.

Project description:In recent decades, global warming has become an indisputable fact on the Tibetan Plateau. Alpine ecosystems are very sensitive to global warming, and the impact may depend on the degree of atmospheric nitrogen (N) deposition. The previous studies have paid more attention to year-round warming, but the effect of winter warming has been unstudied. In this study, a manipulative experiment was conducted, consisting of warming and N addition. It was carried out since 2010 in an alpine meadow, and three types of warming treatments were set up: no warming (NW), year-round (YW), and winter warming (WW). Warming significantly increased air and soil temperature, but decreased soil moisture. Under no N addition, YW showed significantly decreased ecosystem respiration (Reco) in 2012, and WW decreased Reco in 2014. Under N addition, neither YW nor WW had significant effects on Reco, indicating that N addition compensated the negative effect of warming on Reco. Annually, YW and WW decreased ecosystem carbon (C) emissions, and the extent of the reduction was even larger under WW. Under no N addition, both YW and WW significantly decreased aboveground biomass. Moreover, especially under no N, YW and WW significantly decreased soil inorganic N. WW also had negative effects on soil microbial biomass C. Structure equation modeling showed that soil moisture was the most important factors controlling Reco, and soil inorganic N content and microbial biomass C could explain 46.6% and 16.8% of the variation of Reco. The findings indicate that soil property changes under warming had substantial effects on ecosystem C efflux. The inhibitory effects of winter warming on ecosystem C efflux were mainly attributed to the decline of soil N and microbial biomass. Thus, the effects of winter warming on ecosystem C emissions in this semiarid alpine meadow are not as serious as expected and largely depend on N deposition.

Project description:Fire disturbances are becoming more common, more intense, and further-reaching across the globe, with consequences for ecosystem functioning. Importantly, fire can have strong effects on the soil microbiome, including community and functional changes after fire, but surprisingly little is known regarding the role of soil fire legacy in shaping responses to recent fire. To address this gap, we conducted a manipulative field experiment administering fire across 32 soils with varying fire legacies, including combinations of 1-7 historic fires and 1-33 years since most recent fire. We analyzed soil metatranscriptomes, determining for the first time how fire and fire legacy interactively affect metabolically-active soil taxa, the microbial regulation of important carbon (C), nitrogen (N) and phosphorus (P) cycling, expression of carbohydrate-cycling enzyme pathways, and functional gene co-expression networks. Experimental fire strongly downregulated fungal activity while upregulating many bacterial and archaeal phyla. Further, fire decreased soil capacity for microbial C and N cycling and P transport, and drastically rewired functional gene co-expression. Perhaps most importantly, we highlight a novel role of soil fire legacy in regulation of microbial C, N, and P responses to recent fire. We observed a greater number of functional genes responsive to the interactive effects of fire and fire legacy than those affected solely by recent fire, indicating that many functional genes respond to fire only under certain fire legacy contexts. Therefore, without incorporating fire legacy of soils, studies will miss important ways that fire shapes microbial roles in ecosystem functioning. Finally, we showed that fire caused significant downregulation of carbon metabolism and nutrient cycling genes in microbiomes under abnormal soil fire histories, producing a novel warning for the future: human manipulation of fire legacies, either indirectly through global change-induced fire intensification or directly through fire suppression, can negatively impact soil microbiome functional responses to new fires.

Project description:Anthropogenic global warming, nitrogen addition, and overgrazing alter plant communities and threaten plant biodiversity, potentially impacting community productivity, especially in sensitive mountain grassland ecosystems. However, it still remains unknown whether the relationship between plant biodiversity and community productivity varies across different anthropogenic influences, and especially how changes in multiple biodiversity facets drive these impacts on productivity. Here, we measured different facets of biodiversity including functional and phylogenetic richness and evenness in mountain grasslands along an environmental gradient of elevation in Yulong Mountain, Yunnan, China. We combined biodiversity metrics in a series of linear mixed-effect models to determine the most parsimonious predictors for productivity, which was estimated by aboveground biomass in community. We examined how biodiversity-productivity relationships were affected by experimental warming, nitrogen addition, and livestock-grazing. Species richness, phylogenetic diversity, and single functional traits (leaf nitrogen content, mg/g) represented the most parsimonious combination in these scenarios, supporting a consensus that single-biodiversity metrics alone cannot fully explain ecosystem function. The biodiversity-productivity relationships were positive and strong, but the effects of treatment on biodiversity-productivity relationship were negligible. Our findings indicate that the strong biodiversity-productivity relationships are consistent in various anthropogenic drivers of environmental change.

Project description:Nitrogen (N) deposition might alleviate degradation of alpine grassland caused by N limitation on the Tibetan Plateau (TP). To determine such limitation and quantify the N-induced N retention in plant, a six-year fertilization experiment with six levels of N addition rates (0, 1, 2, 4, 8 and 16 g N m-2 yr-1) was conducted in the Namco alpine steppe and additional 89 experiments with multi-level N addition were also synthesized worldwide among which 27 sites were on the TP. In general, N addition promoted N retention in plants, and this increasing trend diminished at the critical N rate (Ncr). The maximum N retention capacity (MNRC) of plants at Ncr was strongly correlated with initial aboveground net primary productivity with a slope of 0.02, and the MNRC of grasslands globally ranged from 0.35 to 42.59 g N m-2 yr-1, approximately account for 39% of Ncr. Tibetan alpine grassland had a low average MNRC (2.24 g N m-2 yr-1) with distinct regional characteristic, which was much lower in the western TP (0.80 g N m-2 yr-1) than the eastern TP (4.10 g N m-2 yr-1). Our results inferred 0.33-1.21 Tg N yr-1 (0.22-0.79 g N m-2 yr-1) can be retained and 5.65-20.11 Tg C yr-1 (3.67-13.06 g C m-2 yr-1) can be gained by Tibetan alpine grasslands under current N deposition level. With the aggravation of N deposition, the alpine steppe ecosystem might continuously absorb N and C until N deposition reaches Ncr.

Project description:Because of severe abiotic limitations, Antarctic soils represent simplified ecosystems, where microorganisms are the principle drivers of nutrient cycling. This relative simplicity makes these ecosystems particularly vulnerable to perturbations, like global warming, and the Antarctic Peninsula is among the most rapidly warming regions on the planet. However, the consequences of the ongoing warming of Antarctica on microorganisms and the processes they mediate are unknown. Here, using 16S rRNA gene pyrosequencing and qPCR, we report a number of highly consistent changes in microbial community structure and abundance across very disparate sub-Antarctic and Antarctic environments following three years of experimental field warming (+ 0.5-2°C). Specifically, we found significant increases in the abundance of fungi and bacteria and in the Alphaproteobacteria-to-Acidobacteria ratio. These alterations were linked to a significant increase in soil respiration. Furthermore, the shifts toward generalist or opportunistic bacterial communities following warming weakened the linkage between bacterial diversity and functional diversity. Warming also increased the abundance of some organisms related to the N-cycle, detected as an increase in the relative abundance of nitrogenase genes via GeoChip microarray analyses. Our results demonstrate that soil microorganisms across a range of sub-Antarctic and Antarctic environments can respond consistently and rapidly to increasing temperatures, thereby potentially disrupting soil functioning.
