Project description:Baby leaves of lettuce (Lactuca sativa L.) are widely used to produce ready-to-eat salads all around the year. For this purpose, lettuce plants are cultivated under plastic tunnels and harvested at a very early growth stage, between 30-45 days after sowing depending on the season, cultivar and site of cultivation. Usually, the plastic film covering the cultivation tunnels transmits only a very small amount of the solar UVB radiation and partially attenuates UVA radiation. We used UV treatment post-harvest of fresh-cut lettuce baby leaves to compare the efficiency of two UV emission wavebands centred at 306 and 366 nm (achieved by employing narrow-band UV LEDs sources) to identify putative transcriptomic signatures. Project description:In the Central Andean region in South America, high-altitude ecosystems (3500-6000 masl) are distributed across Argentina, Chile, Bolivia and Peru, in which poly-extremophilic microbes thrive under extreme environmental conditions. In particular, in the Puna region, total solar irradiation and UV incidence are the highest on Earth, thus, restraining the physiology of individual microorganisms and the composition of microbial communities.
UV-resistance of microbial strains thriving in High-Altitude Andean Lakes was demonstrated and their mechanisms were partially characterized by genomic analysis, biochemical and physiological assays. Then, the existence of a network of physiological and molecular mechanisms triggered by ultraviolet light exposure was hypothesized and called "UV-resistome". It includes some or all of the following subsystems: (i) UV sensing and effective response regulators, (ii) UV-avoidance and shielding strategies, (iii) damage tolerance and oxidative stress response, (iv) energy management and metabolic resetting, and (v) DNA damage repair. Genes involved in the described UV-resistome were recently described in the genome of Nesterenkonia sp. Act20, an actinobacterium which showed survival to high UV-B doses as well as efficient photorepairing capability.
The aim of this work was to use a proteomic approach together with photoproduct measurements to help dissecting the molecular events involved in the adaptive response of a model High-Altitude Andean Lakes (HAAL) extremophilic actinobacterium, Nesterenkonia sp. Act20, under artificial UV-B radiation. Our results demonstrate that UV-B exposure induced over-abundance of a well-defined set of proteins while recovery treatments restored the proteomic profiles present before the UV-challenge. The proteins involved in this complex molecular network were categorized within the UV-resistome subsystems: damage tolerance and oxidative stress response, energy management and metabolic resetting, and DNA damage repair.

Project description:Purpose: UV-B radiation is a pivotal photomorphogenic signal and positively regulates plant growth and metabolite biosynthesis. In order to elucidate the transcriptional regulation mechanism underlying UV-B-induced artemisinin and flavonoid biosynthesis in Artemisia annua, the transcriptional response of A. annua leaves to UV-B radiation was analyzed using the Illumina transcriptome sequencing. Methods: For UV-B treatment, six-week-old A. annua seedlings growing under normal growth condition were supplemented with extra narrowband UV-B lamps (Philips TL20W/01RS; 1.5 μmol·m-2·s-1)(Yin et al. 2016). The most recently expanded leaf for each A. annua seedling was collected at 0, 2, 4 and 6 hours after exposure to UV-B radiation. Results: A total of 10706 differentially expressed genes including 533 transcription factors, were identified. Based on the expression trends of the differentially expressed factors as well as artemisinin and flavonoid biosynthesis genes, we speculated that transcription factors belonging to 6 clusters were most likely to be involved in the regulation of artemisinin and/or flavonoid biosynthesis. The regulatory relationship between transcription factors and artemisinin/flavonoid biosynthetic genes was further studied. Dual-LUC assays results showed that AaMYB6 is a positive regulator of AaLDOX, which belongs to flavonoid biosynthesis pathway. In addition, we identified a R2R3MYB transcription factor, AaMYB4 which positively mediated both artemisinin and flavonoid biosynthesis pathways by activating the expression of AaADS and AaDBR2 in artemisinin biosynthesis pathway and AaUFGT in flavonoid biosynthesis pathway. Conclusions: our findings provide fundamental knowledge for the further analysis of the parallel transcriptional regulation of artemisinin and flavonoid biosynthesis in A. annua L. under UV-B radiation.

Project description:DNA damage caused by UV radiation initiates cellular recovery mechanisms, which involve activation of DNA damage response pathways, cell cycle arrest and apoptosis. To assess cellular transcriptional responses to UVC-induced DNA damage we compared time course responses of human skin fibroblasts to low and high doses of UVC radiation known to induce a transient cellular replicative arrest or apoptosis, respectively. UVC radiation elicited >3-fold changes in 460 out of 12,000 transcripts and 89% of these represented downregulated transcripts. Only 5% of the regulated genes were common to both low and high doses of radiation. Cells inflicted with a low dose of UVC exhibited transcription profiles demonstrating transient regulation followed by recovery, whereas the responses were persistent after the high dose. A detailed clustering analysis and functional classification of the targets implied regulation of biologically divergent responses and suggested involvement of transcriptional and translational machinery, inflammatory, anti-proliferative and anti-angiogenic responses. The data support the notion that UVC radiation induces prominent, dose-dependent downregulation of transcription. However, the data strongly suggest that transcriptional repression is also target gene selective. Furthermore, the results demonstrate that dose-dependent induction of cell cycle arrest and apoptosis by UVC radiation are transcriptionally highly distinct responses. Keywords: other

Project description:UV-B radiation affects leaf growth in a wide range of species. In this work, we demonstrate that UV-B levels present in solar radiation inhibits maize leaf growth without causing any other visible stress symptoms, including accumulation of DNA damage. We conducted kinematic analyses of cell division and expansion to understand the impact of UV-B radiation on these cellular processes. Our results demonstrate that the decrease in leaf growth is a consequence of a reduction in cell production, and a shortened growth zone (GZ) in UV-B irradiated leaves. To determine the molecular pathways involved in UV-B inhibition of leaf growth, we performed RNA sequencing on isolated GZ tissues of control and UV-B exposed plants. Our results show a link between the observed leaf growth inhibition and the expression of specific cell cycle and developmental genes, including Growth Regulating Factors (GRFs) and transcripts for proteins participating in different hormone pathways.

Project description:Recently, we found a dioecious plant Populus cathayana males possess a greater tolerance to enhanced UV-B radiation than do females. To carry this work forward, comparative transcriptome analyses were carried out. Similar to previous studies, a set of conserved functions and pathways related to UV-B stress were detected in males and females, regardless of the sex. In addition, sex-specific responses via transcriptome remodeling were also detected as shown in the changes of sex-related gene expression occurred in some pathways. For example, a lot of differentially expressed genes (DEGs) involved in amino acid metabolism were mainly up-regulated in males, but down-regulated in females. Moreover, we found some DEGs expressed predominantly or exclusively in one sex, which may directly contribute to sex-related physiological responses. 4 samples examined: (i) males exposure to decreased solar UV-B radiation (MC); (ii) females exposure to decreased solar UV-B radiation (FC); (iii) males exposure to ambient solar UV-B radiation (MU); and (iv) females exposure to ambient solar UV-B radiation (FU). Nine plants of each sex were exposed to each treatment, and RNA samples from the 9 individuals were pooled with equal proportion.
