Metagenomic analysis of rumen and fecal microbiome of goats fed with enzyme pretreated feeds

ABSTRACT: This study aims to investigate the impact of enzyme cellulase and xylanase pretreated dietary feed on goat Capra hircus gut microbiome. The primary focus is to understand how these enzyme pretreated feeds affect rumen and fecal microbial communities. Utilizing a 16S metagenomic approach we analyse how different pretreated feedstuff influence the structure of rumen and fecal microbial communities and their association with various physiological parameters of goat.

Project description:One important obstacle in using softwood in biorefineries is its recalcitrance to enzymatic saccharification. In the present work, Thermothielavioides terrestris LPH172 was cultivated on three different steam-pretreated spruce materials and on non-treated spruce, harboring different hemicellulose content and structural characteristics. The aim of the work was to map the substrate-induced changes in the secretome of T. terrestris when grown on differently treated spruce materials and evaluate the hydrolytic efficiency of the secretome as supplement of a commercial enzyme mixture. The cultivation of T. terrestris was monitored by enzyme activity measurements of endo-cellulase, endo-xylanase, endo-mannanase, laccase and peroxidase. Proteomic analysis was performed on the secretomes induced by these materials to map the differences in enzyme expression.

Nutrigenomics and nutritional analysis of juvenile Asian seabass (Lates Calcarifer) fed five different commercial feeds

Project description:In this study, the relative effect of consuming five different commercial feeds (Feed B-F) and and frozen fish (control; Feed A) for two months on our selected juvenile Asian seabass (77.3g ± 22.4g) were investigated. The growth performance, the biochemical analysis of their flesh, the histology of their midgut and the transcriptome of their midgut and liver were compared to each other and controls. Our customised agilent microarray platform (GPL17855), were used to investigate the liver transcriptomic expression of the different Groups fed with different Feeds. No differential expression were detected between commerical Feeds (B, C, E and F). A total of 397 differentially expressed transcripts (Foldchange cutoff: -1.5≤ or ≥1.5, Pvalue with FDR (<0.05) were detected between Feed D and control Feed A.

Exosomal microRNA sequencing in Tongxinluo pretreated and non-treated MSC-derived exosomes

Project description:In order to investigate the specific mechanisms underlying the cardioprotective effects of Tongxinluo pretreated MSC-derived exosomes (MSCTXL-Exo) in cardiac repair, we performed microRNA sequencing on exosomes secreted from Tongxinluo pretreated MSCs and non-treated MSCs to identify differentially expressed miRNA. We found that 18 miRNAs were identified to be upregulated and 25 miRNAs downregulated (over 2-fold change) in MSCTXL-Exo compared to MSC-Exo.
