Project description:Enzyme-mediated decomposition of soil organic matter (SOM) is controlled, amongst other factors, by organic matter properties and by the microbial decomposer community present. Since microbial community composition and SOM properties are often interrelated and both change with soil depth, the drivers of enzymatic decomposition are hard to dissect. We investigated soils from three regions in the Siberian Arctic, where carbon rich topsoil material has been incorporated into the subsoil (cryoturbation). We took advantage of this subduction to test if SOM properties shape microbial community composition, and to identify controls of both on enzyme activities. We found that microbial community composition (estimated by phospholipid fatty acid analysis), was similar in cryoturbated material and in surrounding subsoil, although carbon and nitrogen contents were similar in cryoturbated material and topsoils. This suggests that the microbial community in cryoturbated material was not well adapted to SOM properties. We also measured three potential enzyme activities (cellobiohydrolase, leucine-amino-peptidase and phenoloxidase) and used structural equation models (SEMs) to identify direct and indirect drivers of the three enzyme activities. The models included microbial community composition, carbon and nitrogen contents, clay content, water content, and pH. Models for regular horizons, excluding cryoturbated material, showed that all enzyme activities were mainly controlled by carbon or nitrogen. Microbial community composition had no effect. In contrast, models for cryoturbated material showed that enzyme activities were also related to microbial community composition. The additional control of microbial community composition could have restrained enzyme activities and furthermore decomposition in general. The functional decoupling of SOM properties and microbial community composition might thus be one of the reasons for low decomposition rates and the persistence of 400 Gt carbon stored in cryoturbated material.

Project description:Wildfires produce large amounts of pyrogenic carbon (PyC), including charcoal, known for its chemical recalcitrance and sorption affinity for organic molecules. Wildfire-derived PyC can be transported to fluvial networks. Here it may alter the dissolved organic matter (DOM) concentration and composition as well as microbial biofilm functioning. Effects of PyC on carbon cycling in freshwater ecosystems remain poorly investigated. Employing in-stream flumes with a control versus treatment design (PyC pulse addition), we present evidence that field-aged PyC inputs to rivers can increase the dissolved organic carbon (DOC) concentration and alter the DOM composition. DOM fluorescence components were not affected by PyC. The in-stream DOM composition was altered due to leaching of pyrogenic DOM from PyC and possibly concurrent sorption of riverine DOM to PyC. Decreased DOM aromaticity indicated by a lower SUVA245 (-0.31 unit) and a higher pH (0.25 unit) was associated with changes in enzymatic activities in benthic biofilms, including a lower recalcitrance index (β-glucosidase/phenol oxidase), suggesting preferential usage of recalcitrant over readily available DOM by biofilms. The deposition of particulate PyC onto biofilms may further modulate the impacts of PyC due to direct contact with the biofilm matrix. This study highlights the importance of PyC for in-stream biogeochemical organic matter cycling in fire-affected watersheds.

Project description:This article contains data related to the research article entitled "An Investigation of Organic Matter Quality and Quantity in Acid Soils as Influenced by Soil Type and Land Use" (Shen et al., 2018) [1]. The data was collected using a chemical fractionation scheme of soil organic matter (OM). This involved the separation of organic carbon (OC) fractions based on their solubility in (i) cold and hot water, (ii) 0.1 M sodium pyrophosphate (pH ~ 10), and (iii) 2% HF solution, and the residue remaining after the HF extraction. The OM in this residue, after treatment with 2% HF solution, was characterised using pyrolysis (Py)-GC/MS. This technique involves thermal decomposition of OM into various pyrolysis products, which are then chromatographically separated and determined by mass spectroscopy. This technique has been used to semi-quantify individual soil OM constituents so that in-depth information on soil OM molecular fingerprints is provided. This article presents a detailed dataset of physical-chemical characterization, OC fractions and OM molecular fingerprints of 62 soil samples for a range of soil orders (i.e., Allophanic, Brown, Gley, Pallic and Recent) and land uses (i.e., permanently grazed pasture, ungrazed/unmanaged grasslands, annual cropping) across New Zealand. Principal component analysis was used to investigate the relationships of different soil properties with OC fractions and OM chemistry so that the underlying mechanisms responsible for the differences encountered in OM quantity and quality between soil orders and land uses are understood.

Project description:Soil microbial activity (SMA) is vital concerning carbon cycling, and its functioning is recognized as the primary factor in modifying soil carbon storage potential. The composition of the microbial community (MC) is significant in sustaining environmental services because the structure and activity of MC also influence nutrient turnover, distribution, and the breakdown rate of soil organic matter. SMA is an essential predictor of soil quality alterations, and microbiome responsiveness is imperative in addressing the escalating sustainability concerns in the Himalayan ecosystem. This study was conducted to evaluate the response of soil microbial and enzyme activities to land conversions in the Northwestern Himalayas (NWH), India. Soil samples were collected from five land use systems (LUSs), including forest, pasture, apple, saffron, and paddy-oilseed, up to a depth of 90 cm. The results revealed a significant difference (p < 0.05) in terms of dehydrogenase (9.97-11.83 TPF µg g-1 day-1), acid phosphatase (22.40-48.43 µg P-NP g-1 h-1), alkaline phosphatase (43.50-61.35 µg P-NP g-1 h-1), arylsulphatase (36.33-48.12 µg P-NP g-1 h-1), fluorescein diacetate hydrolase (12.18-21.59 µg g-1 h-1), bacterial count (67.67-123.33 CFU × 106 g-1), fungal count (19.33-67.00 CFU × 105 g-1), and actinomycetes count (12.00-42.33 CFU × 104 g-1), with the highest and lowest levels in forest soils and paddy-oilseed soils, respectively. Soil enzyme activities and microbial counts followed a pattern: forest > pasture > apple > saffron > paddy-oilseed at all three depths. Paddy-oilseed soils exhibited up to 35% lower enzyme activities than forest soils, implying that land conversion facilitates the depletion of microbiome diversity from surface soils. Additionally, reductions of 49.80% and 62.91% were observed in enzyme activity and microbial counts, respectively, with soil depth (from 0-30 to 60-90 cm). Moreover, the relationship analysis (principal component analysis and correlation) revealed a high and significant (p = 0.05) association between soil microbial and enzyme activities and physicochemical attributes. These results suggest that land conversions need to be restricted to prevent microbiome depletion, reduce the deterioration of natural resources, and ensure the sustainability of soil health.
