Spinetoram sublethal exposure stimulates Dengue virus replication in Aedes aegypti by dysregulating oxidative phosphorylation

Project description:Vector-borne diseases are closely linked to the environment by the ecology of the vectors and their hosts. Elucidating these causal relationships is one of the most pressing challenges faced by researchers and public health scientists as increasing anthropogenic alternations in the environment can drive shifts in vector-borne disease dynamics. However, as one of the major environmental stressors, the impact of sublethal exposure of insecticides on vector behavior remains poorly investigated. Here, we analyzed how sublethal exposure of the promising vector-control bioinsecticide spinetoram on Aedes aegypti larvae, the latest generation of the spinosyns that have been suggested as a promising new bioinsecticide for vector control, at the larval stage of Ae. aegypti alters adult performance and susceptibility to dengue virus (DENV) infection. While spinetoram sublethal exposure leads to extended immature development time, these survivors were significantly smaller and exhibited weaker blood-feeding capacity than normal females. More importantly, surviving females presented higher DENV susceptibility than the control group after spinetoram sublethal exposure on larvae. Mechanistically, the transcriptomic analysis showed that inhibition of oxidative phosphorylation (OXPHOS) may function in stimulating DENV production in adult Ae. aegypti. In vitro study revealed that spinetoram induces apoptosis via mitochondrial reactive oxygen species (mtROS)-regulated OXPHOS dysregulation in Aag2 cells, and thereby may stimulate DENV production through the metabolic switch between OXPHOS and glycolysis. Altogether, our data demonstrate that sublethal spinetoram exposure could act as a crucial factor on life traits and vector competence in Ae. aegypti. Given that other insecticides are known to induce mitochondrial dysfunction in mosquito vectors, this research may have wider implications.

Evaluation of chronic sublethal effects effects of the organophosphate flame retardant Tris(2-butoxyethyl)phosphate (TBEP) using Daphni magna transcriptomic response

Project description:The aim of this study was to measure chronic sublethal effects of the flame retardant Tris(2-butoxyethyl)phosphate (TBEP) on gene expression, associated protein activity and life-history endpoints (reproduction, development) in the freshwater crustacean Daphnia magna. We exposed <24h old D. magna neonates for 21 days to culture medium (control) and to 3 sublethal doses of TBEP: 14.7, 147 and 1470 M-BM-5g/L. Exposure doses were chosen from 48h acute toxicity testing and represent 1/10000, 1/1000 and 1/100 of the LC50, respectively. Each condition (control and the 3 different doses) were performed on 3 independant biological replicates, with 10 individuals each. We used a custom 15000 sequences D. magna microarray to measure transcriptional effect of each of the TBEP doses in comparison to unexposed controls. We exposed <24h old Daphnia magna to culture medium (unexposed controls) and 3 sublethal doses of TBEP (14.7M-BM-5g/L, 147M-BM-5g/L and 1470M-BM-5g/L) for 21 days. We used 3 biological replicates of 10 individuals for each condition.

Transcriptome responses to sublethal concentration of abamectin in Tetranychus truncatus analyzed by deep-sequencing

Methods: We sequenced the transcriptome of Tetranychus truncatus treated with sublethal concentration of abamectin and control. Results: we provide a comprehensive transcriptome resource for Tetranychus truncatus derived from treating with sublethal concentration abamectin and control. Conclusions: The new genomic resources and data that we present in this study for Tetranychus truncatus will substantially facilitate molecular studies of underlying mechanisms involved in abamectin resistance.

Sublethal concentrations of antibiotics cause shift to anaerobic metabolism in Listeria monocytogenes and induce phenotypes linked to antibiotic tolerance

Project description:The human pathogenic bacterium Listeria monocytogenes was exposed to antibiotics both during clinical treatment and as a saprophyte. As one of the keys to successful treatment is continued antibiotic sensitivity, the purpose of this study was to determine if exposure to sublethal antibiotic concentrations would affect the bacterial physiology and potentially induce tolerance to antibiotics. Transcriptomic analyses demonstrated that each of four antibiotics caused a compound-specific gene expression pattern related to (the) mode-of-action of the particular antibiotic. All four antibiotics caused the same changes in expression of several metabolic genes indicating a shift from aerobic to anaerobic metabolism driven by the induction of lmo1634 and the repression of alsA and lmo1992. This shift in metabolism could be a survival strategy in response to antibiotics and is further supported by the observation that a Îlmo1634 mutant was more sensitive to bactericidal antibiotics. The monocin locus encoding a cryptic prophage was induced by co-trimoxazole and repressed by ampicillin and gentamicin. This expression pattern correlated with the observed antibiotic-dependent biofilm formation, indicating a role of monocin in antibiotic-induced biofilm formation and a ÎlmaDCBA mutant confirmed this correlation. Thus, sublethal concentrations of antibiotics caused metabolic and physiological changes indicating that the organism is preparing to withstand lethal concentrations of antibiotics. Investigation of mRNA and sRNA expression profiles of L. monocytogenes EGD cells exposed to sublethal concentrations of four different antibiotics i.e. ampicillin, tetracycline, gentamicin and co-trimoxazole for 3h.

Differentially expressed genes in radioresistant nasopharyngeal cancer cells

Project description:The NPC parental cell lines were seeded at a density of 1 x 105/T25 flask (30% to 40% confluency) in complete medium. After 24 hours of cell seeding, various doses from 6 to 14 Gy were administered to determine the sublethal dose for each cell line. The radiation was delivered at room temperature at 300 cGy/min with a linear accelerator (2100EX, Varian, VA, USA). To establish RR subclone, the NPC cells were received the sublethal dose of irradiation. After treatment, the surviving cells were selected and cultured to produce first generation of the subline cells. Again, the subline cells were received sublethal dose of irradiation and the surviving cells were cultured to produce next generation of the subline cells.
