Project description:Differentiation into plasma cells (PC) enables secretion of ~10,000 antibody molecules per second. This extraordinary capacity requires the upregulation of PC transcriptional determinants that increase immunoglobulin mRNA synthesis, coordinate alternative 3’end processing of the heavy chain transcript from the distal to proximal poly-adenylation site (PAS), and remodel the secretory pathway. Based on the difficulty of prospectively identifying secretory cells, we developed a dual-fluorescent protein reporter mouse to study the post-transcriptional-level transition from membrane-anchored to secretory immunoglobulin M (µM PAS and µS PAS, respectively), in single cells. We observed (i) dynamic acquisition of graded µS PAS usage during PC differentiation (ii) IRF4 and Blimp-1 functioned hierarchically to increase µ abundance as well as µS PAS usage and (iii) graded µS populations did or did not express Blimp-1. Interestingly, the low and high µS and Blimp-1-expressing populations arose from distinct developmental intermediates that exhibited dissimilar endoplasmic reticulum features revealing unpredicted complexity of PC differentiation. The novel cell and µS PAS fate trajectories may have implications for discrete specializations of the secretory pathway.

Project description:Mycobacterium neoaurum (Mn), an efficient sterol consumer, has been modified to transform sterols to produce valuable steroid intermediates, which can be widely used as precursors in the synthesis of steroid hormones.To deepen our understand of the underlying mechanisms of Mn to the in vitro and in vivo environment during growth on a critical carbon source of sterol and accumulation of valuable steroid intermediates, transcriptome studies were performed to characterize the differences between wide-type Mn and various mutant Mn strains for steroid accumulation. Totally, five Mn strains were investigated, including wide-type Mn ATCC25795 cultured without (Mn-C) and with sterol addition (Mn-CC), mutant Mn ATCC25795 strains producing 9OHAD (Mn-9OHAD), ADD (Mn-ADD), and 1,4-HBC (Mn-HBC), respectively.

Project description:The mammalian gut is inhabited by a large and complex microbial community that lives in a mutualistic relationship with its host. Innate and adaptive mucosal defense mechanisms ensure a homeostatic relationship with this commensal microbiota. Secretory antibodies are generated from the active polymeric Ig receptor (pIgR)-mediated transport of IgA and IgM antibodies to the gut lumen and form the first line of adaptive immune defense of the intestinal mucosa. We probed mucosal homeostasis in pIgR knockout (KO) mice, which lack secretory antibodies. We found that in pIgR KO mice, colonic epithelial cells, the cell type most closely in contact with intestinal microbes, differentially expressed (>2-fold change) more than 200 genes compared with wild type mice, and upregulated the expression of anti-microbial peptides in a commensal-dependent manner. Detailed profiling of microbial communities based on 16S rRNA genes revealed differences in the commensal microbiota between pIgR KO and wild type mice. Furthermore, we found that pIgR KO mice showed increased susceptibility to dextran sulfate sodium (DSS)-induced colitis, and that this was driven by their conventional intestinal microbiota. In conclusion, secretory antibodies or the pIgR itself are required to maintain a stable commensal microbiota. In the absence of these humoral effector components, gut homeostasis is disturbed and the outcome of colitis significantly worsened. 4 groups: wild type mice treated with antibiotic (5 replicates), wild type mice left untreated (5 replicates), pIgR KO mice treated with antibiotic (6 replicates), and pIgR KO mice left untreated (6 replicates).

Project description:Waldenström macroglobulinemia (WM), is a rare non-Hodgkin lymphoma preceded by a clinically asymptomatic benign monoclonal gammopathy of undetermined significance (IgM-MGUS). The factors underlying the malignant progression between these two IgM-monoclonal gammopathies remain poorly understood. The non-coding genome is increasingly recognized as an important driver of disease pathogenesis, and there remains a lack of exploration of the influence of non-coding RNAs within the IgM-monoclonal gammopathy disease spectrum. The present study explores the role of microRNA (miRNA) and long non-coded RNA (lncRNA) in IgM-gammopathies and specifically elucidates potentially regulated protein-coding genes and pathways. A comprehensive analysis of miRNA, lncRNA and protein-coding coding genes was conducted on 28 subjects, 17 patients with WM, 6 patients with IgM-MGUS, and 5 normal controls. Differential expression analysis revealed a number of dysregulated miRNA and lncRNA between IgM-gammopathies compared to normal controls and specifically between IgM-MGUS and WM. Pathway analysis was conducted utilizing differentially expressed mRNA with correlated biological expression of targeting miRNA. Here, a number of pathways were implicated influencing a gamut of cellular functions, including cell signaling, metabolism, replication, and immune regulation in both WM compared to IgM-MGUS and IgM-gammopathies compared to controls. This report is additionally one of the first published analyses of lncRNAs in WM, where we demonstrate a number of lncRNA associated with transcription and apoptosis regulation genes in WM compared to IgM-MGUS. This study highlights the role of non-coding RNA in IgM-gammopathies and the potential to recognize novel targets which may halt or slow the malignant progression between these two diseases.

Here we examine the complement activation profiles, binding kinetics, and conformational changes in IgM in different antigen-bound states. Only in the complex with a surface-displayed antigen with multiple Fab arms engaged was the IgM fully capable of driving complement activation. The structural changes within IgM upon antigen binding were monitored by Hydrogen/Deuterium Exchange reveal the predominant structural changes within the Fc core domains upon activation. Collectively, this work establishes key structural and functional qualities that define the complement-active form of IgM.
