Drosophila WDFY3/ Bchs overexpression impairs neural function

Project description:Pathogenic variants in WDFY3, a gene encoding for an autophagy adaptor termed ALFY, are linked to neurodevelopmental delay and altered brain size in human probands. While the role of WDFY3 loss-of-function is extensively studied in neurons, little is known about the effects of WDFY3 upregulation in different cell types of the central nervous system (CNS). We show that overexpression of the Drosophila melanogaster WDFY3 ortholog, Bchs, in either glia or neurons impaired autophagy and locomotion. Bchs glial overexpression also increased VNC size and glial nuclei number significantly, whereas neuronal Bchs overexpression affected wing and thorax morphology. We identified 79 genes that were differentially expressed and overlapped in flies that overexpress Bchs in glial and neuronal cells, respectively. Additionally, upon neuronal Bchs overexpression differentially expressed genes clustered in gene ontology categories associated with autophagy and mitochondrial function. Our data indicate that glial as well as neuronal Bchs upregulation can have detrimental outcomes on neural function.

Neuroligin 2 Governs Synaptic Morphology and Function Through RACK1-Cofilin Signaling in Drosophila

Project description:Neuroligins are transmembrane cell adhesion proteins well-known for their genetic links to autism spectrum disorders. Neuroligins can function by regulating the actin cytoskeleton, however the factors and mechanisms involved are still largely unknown. Here, using the Drosophila neuromuscular junction as a model, we reveal that F-Actin assembly at the Drosophila NMJ is controlled through Cofilin signaling mediated by an interaction between DNlg2 and RACK1, factors not previously known to work together. The deletion of DNlg2 displays disrupted RACK1-Cofilin signaling pathway with diminished actin cytoskeleton proteo-stasis at the terminal of the NMJ, aberrant NMJ structure, reduced synaptic transmission, and abnormal locomotion at the third-instar larval stage. Overexpression of wildtype and activated Cofilin in muscles are sufficient to rescue the morphological and physiological defects in dnlg2 mutants, while inactivated Cofilin is not. Since the DNlg2 paralog DNlg1 is known to regulate F-actin assembly mainly via a specific interaction with WAVE complex, our present work suggests that the orchestration of F-actin by Neuroligins is a highly dynamic and complex process critical for neural connectivity.

Active forgetting requires Sickie function in a dedicated dopamine circuit in Drosophila

Project description:Forgetting is an essential component of the brain memory management system, providing a balance to memory formation processes by removing unused or unwanted memories, or by suppressing their expression. However, the molecular, cellular and circuit mechanisms underlying forgetting are poorly understood. Here we show that the memory suppressor gene, sickie, functions in a single dopamine neuron (DAn) by supporting the process of active forgetting in Drosophila. RNAi knockdown (KD) of sickie impairs forgetting by reducing the Ca2+ influx and DA release from the DAn that promotes forgetting. Co-immunoprecipitation/mass spectrometry analyses identified cytoskeletal and presynaptic active zone (AZ) proteins as candidates that physically interact with Sickie, and a focused RNAi screen of the candidates showed that Bruchpilot (Brp), a presynaptic AZ protein that regulates calcium channel clustering and neurotransmitter release, impairs active forgetting like sickie KD. In addition, overexpression of brp rescued the impaired forgetting of sickie KD, providing evidence that they function in the same process. Moreover, we show that sickie KD in the DAn reduces the abundance and size of AZ markers but increases their number, suggesting that Sickie controls DAn activity for forgetting by modulating the presynaptic AZ structure. Our results identify a new molecular and circuit mechanism for normal levels of active forgetting and reveal a surprising role of Sickie in maintaining presynaptic AZ structure for neurotransmitter release.

Folic acid regulates neural mRNA m6A epitranscriptome via one-carbon metabolism in Drosophila and mammals

Project description:Folic acid is one of the B vitamins and is involved into neural function and brain development. Increasing evidence showed that folic acid can regulate neural gene function through DNA modification, while regulatory roles of folic acid in RNA modification remain largely unknown. In this study, we investigated the role of folic acid in regulating neural mRNA m6A epitranscriptome in Drosophila and mammals. We found that the folate treatment induced a significant increase in mRNA m6A levels compared to the control group in Drosophila and S2 cells. MeRIP-seq analysis indicated that carbon metabolism pathway and neural-related pathways were mainly affected. Subsequently, we migrated our validation experiments to human cells, our results showed that folate also affected mRNA m6A modification through one-carbon metabolic pathway in human cells, especially in neuronal cells. We also validated the effect of folic acid on mRNA methylation in mice, the results showed that folic acid treatment can significantly increase the expression of m6A-related proteins (Mettl3, Mettl14, FTO) and neural mRNA m6A methylation levels in mice brain. Moreover, we found that folic-acid producing Lactobacillus plantarum can significantly affects the m6A modification of mRNA in the host. In conclusion, we demonstrated that folic acid can participate in mRNA m6A modification through one-carbon metabolic pathway in Drosophila and mammals, and found that it has a significant effect on neural-related genes and pathways.
