Project description:The widespread commercial use of TiO2-NPs, such as in medical stents, has led to extensive investigations on their toxicity and biological impact but lacks a detailed study on their effects. Proteomics and transcriptomics are key methodologies to unravel cellular dynamics, macromolecular interactions and biological response to NP exposure through the protein or gene identification and quantification. We focus on integration of several Omics strategies that could offer a more complex understanding of the impact. In this study, our goal was to gain insight into the cellular response after the exposure to sublethal concentration of TiO2-ultra small nanoparticles (USNPs) and NPs by analyzing the modification on the genomic and proteomic expression in human microdermal endothelial cells (HMEC-1). We investigated the correlation between the NP size and the cellular response to the exposure. We evaluated if multiOmics approaches could thus reveal more extensive information on the potential cellular responses to NPs exposure.

Project description:The present study aimed to investigate the human serum proteome wide interaction of titanium dioxide nanoparticles (TiO2-NPs). Methodologically, the specific interaction of serum proteins with TiO2-NPs were explored through high resolution proteomic profiling. Here we employed LC-MS/MS based HRMS protocol to explore the list of interacted proteins. Further, novel human serum proteins interacted with TiO2-NPs and its associated molecular pathways have been illustrated.

Project description:Here we provide a set of 16 comparable transcriptome measures to monitor early changes in gene expression upon NP exposure. We evaluated A. thaliana response to 8 different types of NPs (metallic, carbonaceous and ranging from 10 to 80 nm) in comparison to biotic and abiotic stress inducers that represent most common environmental challenges for plants. Biotic stress was induced by infection with a necrotizing fungus (Alternaria brassicicola) or a hemibiotrophic bacterium (Pseudomonas syringae). Abiotic stresses induced by hypersaline conditions, drought and mechanical wounding were assayed in our plant growth model. The effect of abscisic acid (ABA), the most studied stress-responsive phytohormone which mediates stomatal closure and other reponses to drought and osmotic stress (ref), was also tested in the gene expression and phenotypes of NP-exposed plants. Project description:Transcriptional profiling of the digestive gland tissue of female mussel Mytilus galloprovincialis exposed to TCDD, n-TiO2 and their binary mixture Background: Exposure of marine organisms to pollutant mixtures may affect the pattern of contaminant uptake/bioaccumulation, as well as of gene expression in the tissues. Despite the growing concern over the potential biological impact of nanoparticles (NPs) in the aquatic environment, little is known about their interactions with other pollutants.We have recently shown that in the marine mussel Mytilus galloprovincialis exposure to n-TiO2, one of the most widespread type of NPs in use, in combination with 2,3,7,8-TCDD, chosen as model organic xenobiotic, can exert antagonistic or synergistic effects on different biomarkers from the molecular to the tissue level, depending on cell/tissue and type of measured response. An integrated approach involving immunhistochemical and transcriptomic analysis was employed to clarify the itteractive effects of n-TiO2 and TCDD in mussels digestive gland. In particular,TCDD bioaccumulation was evaluated utilizing specific anti-TCDD fluorescent antibodies. Moreover, immunohistochemical evaluation of antioxidant and cytoskeletal components was performed. To provide clues about how the molecular response to the investigated compounds is modulated, we used a cDNA microarray with1673 sequences. In animals exposed only to TiO2, functional genomics analysis of the microarray data (48 differentially expressed genes (DEGs)) highlighted three biological processes, largely dominated by the up-regulation of microtubule-based movement-related genes. Exposure to 2,3,7,8-TCDD yielded 49 DEGs exhibiting distinct patterns in terms of biological processes. Finally, exposure to the mixture rendered 62 GEGs characterized by the regulation of response to chemical stimulus, microtubule-based movement and intracellular signal transduction. Our data should be carefully considered in view of the biological effects of emerging pollutants, particularly in case of mixture chemicals. Project description:Titanium dioxide nanoparticles (TiO2 NPs) constitute the top five NPs in use today. A recent study determined that oral administration of TiO2 NPs increases plasma glucose in mice. In the current study, RNA sequencing (RNA-seq) technology was used to investigate genome-wide effects of TiO2 NPs. Clustering analysis of the RNA-seq data showed the most significantly enriched gene ontology terms and KEGG pathways related to the endoplasmic reticulum (ER) and ER stress. Molecular biology verification showed that TiO2 NPs activated a xenobiotic biodegradation response and increased expression of cytochrome P450 family genes in mouse livers, thus inducing ER stress in mice. ER stress activated MAPK and NF-B pathways and induced an inflammation response, resulting in phosphorylation of the insulin receptor substrate 1 and, consequently, insulin resistance. This was the main mechanism by which TiO2 NPs increased plasma glucose in mice. Meanwhile, ER stress disturbed the monooxygenase system, and thus generated reactive oxygen species (ROS). Relief of ER stress with 4-phenylbutyric acid inhibited all the above effects of TiO2 NPs, including the generation of ROS. Therefore, TiO2 NP-induced ER stress was a decisive factor with a central role in plasma glucose disturbance in mice.

Project description:In order to evaluate the identification of genes and pathways, the global gene expression profiles were assessed in response to UV, TiO2 and UV+TiO2 on nematode, Caenorhabditis elegans. We performed whole genome DNA microarray experiments using age synchronized young adult C. elegans population exposed to UV, TiO2 and UV+TiO2 for 24h. We used whole genome microarrays to screen for global changed in C. elegans transcription profiles and with subsequent quantitative analysis conducted on selected genes.
