Project description:The Illumina Infinium 27k Human DNA methylation Beadchip v1.2 was used to obtain DNA methylation profiles across approximately 27,000 CpGs in white blood cells from women with and without BRCA1 mutation. Whole blood samples were drawn from 30 BRCA1 mutation carriers (15 women with breast cancer and 15 healthy women with mean age 57.2) and 30 females without BRCA1 mutation (15 breast cancers and 15 healthy with mean age 57.1). All samples were collected between 2001 and 2008. The samples were drawn from women attending the General Faculty Hospital in Prague and the complete coding sequence, intron-exon junctions and large rearrangements for BRCA1 and BRCA2 genes were tested.

Project description:Genetic factors contribute to the development of ischemic stroke but their identity remains largely unknown. We tested the association with ischemic stroke of 210 single nucleotide polymorphisms (SNPs) associated with pathways functionally related to stroke. We observed an association between the rs7956957 SNP in LRP1 and next performed microarrays analysis in healthy individuals to investigate possible associations of LRP genotypes with the expression of other genes. Twelve blood samples were obtained from twelve different healthy subjects carrying different genotypes for the rs7956957 SNP of the LRP1 gene (GG, CG or CC).

Project description:In a randomized cross-over study, thirty-two healthy, young, slightly overweight women and men went through a two times six-day iso- and normocaloric diet intervention. Liquid meal replacement smoothies were designed composed of 65:15:20 (AHC) and 27:30:43 (BMC) energy percent (E %) of carbohydrates, proteins and fats respectively (Note: AHC - Diet A, High carbohydrate (65:15:20 energy percent (E %) of carbohydrates, proteins and fats, respectively. BMC - Diet B, Moderate carbohydrate (27:30:43E % of carbohydrates, proteins and fats, respectively)). Fasting blood samples for transcriptome analyses were collected before and after each diet period.
