RNA sequencing (RNA-seq) of LAIR-1 overexpression in SKOV-3 ovarian cancer cell lines

Project description:Leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1), is widely expressed by solid tumor cells and functions as a tumor suppressor. Our previous studies have found that LAIR-1 is expressed in ovarian cancer tumor tissues and in a variety of epithelial ovarian cancer cell lines and that LAIR-1 could inhibit ovarian cancer cell growth by suppressing the PI3K-AKT-mTOR pathway. The aim of the present study was to obtain a comprehensive understanding of the molecular mechanism of LAIR-1 in regulating the biological behavior of ovarian cancer cells. The approach taken was to perform RNA-seq sequencing on SKOV3 cells overexpressing LAIR-1, and to conduct further validation by combinatorial integration of bioinformatics analyses to assess the genetic alterations and using biological experimental findings.

Project description:Type-2 innate lymphoid cells (ILC2s) are central players in type-2 asthma. They initiate eosinophil infiltration and airway hyperreactivity (AHR) through cytokine secretion. Leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1) is an inhibitory receptor considered as immune checkpoint in different inflammatory diseases. Here we assessed the role of LAIR-1 in murine ILC2s. WT and LAIR-1 KO mice were intranasally challenged with IL-33 and pulmonary ILC2s were sorted to perform RNA sequencing. Our results unravel a novel regulatory axis in ILC2s with the capacity to reduce their activation.

Project description:Recent studies reported the inhibitory properties of LAIR-1 in different inflammatory diseases, including allergic and viral airway inflammation. However, the implication of LAIR-1 in ALI/ARDS remains unknown. Poly(I:C), a synthetic analog of (ds)RNA was administrated intranasally to mimic acute inflammation in viral infections and therefore induce experimental ALI/ARDS. RNA sequencing approach was used to explore and compare lung macrophage transcriptomic profiles in wild-type and LAIR-1 knock-out mice.

Remodeling of the tumor microenvironment via blockade of LAIR-1 and TGF-β signaling [bulk RNA-seq]

Project description:Collagens in the extracellular matrix (ECM) provide a physical barrier to tumor immune infiltration, while also acting as a ligand for immune inhibitory receptors. Transforming growth factor-β (TGF-β) is a key contributor to shaping the ECM by stimulating the production and remodeling of collagens. TGF-β-activation signatures and collagen-rich environments have both been associated with T-cell exclusion and lack of responses to immunotherapy. Here we describe the effect of targeting collagens that signal through the LAIR-1 inhibitory receptor in combination with blockade of TGF-β and programmed cell death ligand 1 (PD-L1). This approach remodeled the tumor collagenous matrix, enhanced tumor infiltration and activation of CD8+ T cells, and repolarized suppressive macrophage populations resulting in high cure rates and long-term tumor-specific protection across murine models of colon and mammary carcinoma. The results highlight the advantage of direct targeting of ECM components in combination with immune checkpoint blockade therapy.
