Transcriptional profiling and comparative genomic analysis of an invasive and non-invasive strain of Candida albicans

Project description:Invasion of host tissue by the human fungal pathogen, Candida albicans is an important step during many forms of candidosis. However, not all C. albicans strains possess the same invasive and virulence properties. It is known for example that the two clinical isolates SC5314 and ATCC10231 differ in their ability to invade into host tissue and to cause infections. Strain SC5314 is invasive whereas strain ATCC10231 is non-invasive and strongly attenuated in virulence as compared to SC5314. In this study we compare the in vitro transcriptional profiles and the genotypic profiles of these two widely used laboratory strains in order to determine the principal biological and genetic properties which may govern the different potential for invasiveness and virulence. Project description:The emergence of insecticide resistance is a fast-paced example of the evolutionary process of natural selection. In our study, we investigated the molecular basis of resistance in the myiasis-causing fly Cochliomyia hominivorax (Diptera: Calliphoridae) to organophosphates (OP). By sequencing the RNA from surviving larvae treated with OP (resistant condition) and non-treated larvae (control condition), we identified genes displaying condition-specific polymorphisms, as well as those differentially expressed. Both analyses revealed that resistant individuals have altered expression and allele-specific expression of genes involved in proteolysis (specifically serine-endopeptidase), olfactory perception and cuticle metabolism, among others. We also confirmed that resistant individuals carry almost invariably the Trp251Ser mutation in the esterase E3 known to confer OP and Pyrethroid resistance. Interestingly, genes involved in metabolic and detoxifying processes (notably cytochrome P450s) were found under-expressed in resistant individuals. An exception to this were esterases, which were found up-regulated. These observations suggest that reduced penetration and aversion to OP contaminated food, may be important complementary strategies of resistant individuals. The specific genes and processes found are an important starting point for future functional studies. Their role in insecticide resistance merits consideration to better the current pest management strategies.

Project description:Understanding how chromatin organisation is duplicated on the two daughter strands is a central question in epigenetics. In mammals, following the passage of the replisome, nucleosomes lose their defined positioning and transcription contributes to their re-organisation. However, whether transcription plays a greater role in the organization of chromatin following DNA replication remains unclear. Here we have monitored protein re-association to newly replicated DNA upon inhibition of transcription using iPOND coupled to quantitative mass spectrometry. We show that RNAPII acts to promote the re-association of hundreds of proteins with newly replicated chromatin, including ATP-dependent remodellers, transcription factors, DNA repair factors, and histone methyltransferases.

Comparative genomic hybridization to study capsule switching between serogroup C and W of Neisseria meningitidis strains

Project description:Changing antigenic structure such as capsule polysaccharide is a common strategy for bacterial pathogen to evade host immune system. In this regard, the recent emergence of an invasive W:2a:P1.7-2,4 ST-11 strain in New Zealand, which is an uncommon pathogenic serogroup, was investigated for its genetic origins. Molecular typing of 103 meningococcal isolates with similar serotyping characteristics was undertaken to determine genetic relationships. Results indicated that the W:2a:P1.7-2,4 strain had emerged via capsule switching from an existing group C strain (C:2a:P1.7 2,4). Neither of the upstream and downstream sites of recombination could be elucidate but sequence analysis demonstrated that at least 45 kb of DNA was involved in the recombination event. This included the entire capsule gene cluster. Genomic DNA isolated from serogroup C strains (n=4) and serogroup W strains (n=4) were compared using Pan-neisseria DNA microarray.
