Proteomics and metabolomics of different delivery modes in human pregnancy

Project description:This project investigates the impact of different delivery modes and labor on maternal and neonatal health by analyzing 40 participants, including 16 spontaneous vaginal deliveries (VD), 16 prelabor cesarean deliveries (CS), and 8 intrapartum cesarean sections (Intra_CS). Using label-free proteomics and untargeted metabolomics, both amniotic fluid and cord blood samples were analyzed respectively to identify variations in protein and metabolite profiles associated with the mode of delivery. The study aims to uncover biological pathways influenced by labor and delivery mode, providing insights into how these factors shape maternal and neonatal outcomes, with implications for improving perinatal care and long-term health strategies.

MicroRNAs in adipocyte-derived extracellular vesicles in maternal and cord blood are related to neonatal adiposity

Project description:Maternal body size, nutrition, and hyperglycemia contribute to neonatal body size and composition. There is little information on maternal-fetal transmission of messages which influence fetal growth. We analyzed adipocyte-derived small extracellular vesicular (ADsEV) microRNAs in maternal and cord blood to explore their adipogenic potential. Differential expression (DE) of ADsEV miRNAs in adipose vs. lean neonates was studied before and after adjustment for maternal gestational diabetes mellitus (GDM), adiposity, and vitamin B12-folate status.

INSIGHT Microbiome Study

Project description:<p>This is a study of the oral and gut microbiome of 226 mother-child dyads enrolled in the INSIGHT (Intervention Nurses Start Infants Growing on Healthy Trajectories) study. INSIGHT is a randomized, controlled trial comparing a responsive parenting intervention designed for the primary prevention of childhood obesity against a control. </p> <p>The microbiome portion of the study was designed to investigate the relationship between a cross-sectional view of the child's microbiome (at two years of age) and the patterns of growth between birth and 2 years. These first-born children were deeply studied in this time period with data collected on a wide variety of variables including mode of delivery, sex, weight and height (collected at 7 time points), medication usage, diet information, and maternal health information (gestational weight gain, gestational diabetes, smoking during pregnancy). Microbiome samples from the child (buccal swab and stool sample) and their mother (buccal swab) were collected at the child's 2-year clinical research visit. </p>

Umbilical cord blood metabolome differs in relation to delivery mode, birth order and sex, maternal diet and possibly future allergy development in rural children

Project description:Allergy is one of the most common diseases among young children yet all factors that affect development of allergy remain unclear. In a small cohort of 65 children living in the same rural area of south-west Sweden, we have previously found that maternal factors, including prenatal diet, affect childhood allergy risk, suggesting that in utero conditions may be important for allergy development. Here, we studied if metabolites in the umbilical cord blood of newborns may be related to development of childhood allergy, accounting for key perinatal factors such as mode of delivery, birth order and sex. Umbilical cord blood plasma samples from 44 of the participants were analysed using gas chromatography-mass spectrometry metabolomics; allergy was diagnosed by specialised paediatricians at ages 18 months, 36 months and 8 years and included eczema, asthma, food allergy and allergic rhinoconjunctivitis. Nineteen cord blood metabolites were related to future allergy diagnosis though was no clear pattern of up- or downregulation of metabolic pathways. In contrast, perinatal factors birth order, sex and mode of delivery affected several energy and biosynthetic pathways, including glutamate and aspartic acid - histidine metabolism (p=0.004) and the tricarboxylic acid cycle (p=0.006) for birth order; branched chain amino acid metabolism (p=0.0009) and vitamin B6 metabolism (p=0.01) for sex; and glyoxylate and dicarboxylic acid metabolism (p=0.005) for mode of delivery. In conclusion, the cord blood metabolome includes individual metabolites that reflect lifestyle, microbial and other factors that may also be associated with future allergy diagnosis, and also reflects temporally close events/factors. Larger studies are required to confirm these associations, and perinatal factors such as birth order or siblings must be considered in future cord-blood metabolome studies.

Fetal metabolic programing increases the susceptibility of cardiovascular diseases in the offspring due to maternal high-fat-diet during diabetic pregnancy

Project description:Increasing trends of obesity in childbearing women with a concurrent gestational diabetes are often associated with adverse fetal metabolic cues. Not only the carbohydrates but also fats are considered to contribute additional dietary fuels to the fetal hearts. To examine the hitherto unknown epigenetic mechanisms on the offspring’s cardiometabolic health due to maternal high-fat (HF) diet (40% kcal) or streptozotocin (STZ)-induced diabetic pregnancy, or both, we carried out an epigenetic characterization of neonatal heart tissue using chromatin immunoprecipitation (ChIP) sequening and a previously validated rat model. Chromatin landscape of offspring’s heart tissue revealed differential peaks distribution on various promoter regions mapped to the rat genome due to histone (H3) modifications by acetylation (H3Ac) or trimethylations of lysine 4 and 27 (H3K4me3 and H3K27me3, respectively). Ongoing evaluations include gene ontology and disease ontology analyses. Together, it is expected that the findings will show that maternal HF-diet with or without gestational diabetes changes the cardiac histone signature in rat offspring, potentially leading to development of novel disease prevention strategies.

Project description:Epidemiological studies have associated maternal metabolic conditions such as obesity and gestational diabetes with poor health outcomes in the offspring. Epigenetic mechanisms may help explain the intrauterine influence that mothers have on their offspring during pregnancy. Here, using Illumina's MethylationEPIC array technology, we have longitudinally profiled the blood methylomes of children born to mothers with obesity and obesity with gestational diabetes, and healthy controls, during the first year of life (measurements at 0, cord blood; 6 and 12 months, peripheral blood).

High-fat diet during mouse pregnancy impairs fetal heart development

Project description:Maternal obesity is associated with detrimental outcomes in offspring. However, the specific effects of gestational exposure to a high-fat diet (HFD) on fetal heart development remain poorly understood. In the study, we investigated that maternal overnutrition accounted for defective development in offspring, and cell proliferation was restricted severely in neonatal organ. Starting with this clue, we found decreased cell cycle and similar cross area of cardiomyocytes in offspring exposed to a maternal HFD. Moreover, RNA sequencing analysis detected that HFD diminished fatty acid metabolism, enhanced inflammation response and up-regulated substantially the pathway TP53 regulates transcription of cell cycle genes in postnatal day 0 (P0) cardiomyocytes. Further, normal lactation after delivery not only recovered from growth retardation but also maintained normal cardiac function in offspring. These data demonstrated a potential TP53 pathway-dependent mechanism through which maternal HFD contributed to offspring cardiac defects.
