PBMC expression profiles of BCG vaccine protected infants and of BCG vaccinated infants who develop tuberculosis disease

Project description:Analysis of PBMC from 10 week old infants vaccinated with BCG at birth. During follow-up 26 infants developed tuberculosis (TB) (non-protected by BCG) and 20 infants did not develop TB despite documented exposure (protected by BCG). PBMC were stimulated with media only or reconstituted BCG for 4 and 12 hours. Results provide insight into the mechanisms behind the failure of BCG to protect against disease.

Transcriptome responses to Salmonella enterica serovar Reading in vaccinated and unvaccinated turkeys

Project description:The 2017-2019 foodborne outbreak of Salmonella enterica serovar Reading (S. Reading) in North America revealed the need for effective control of this serovar in turkey production. This study evaluated two live-attenuated Salmonella vaccines against an outbreak-associated strain of S. Reading in turkeys. At 1 day and 3 weeks of age, male turkey poults were either mock-vaccinated or given either an internally developed cross-protective vaccine and a commercially available vaccine. At 7 weeks of age, poults were challenged with S. Reading; one mock-vaccinated group was mock-challenged. Along with assessment of Salmonella colonization and dissemination, acute transcriptomic responses in the cecal tonsil were characterized at 2 days post inoculation and revealed decreased expression of genes encoding intestinal transporters and tight junction proteins. Vaccination with either vaccine mitigated most of the transcriptional changes in intestinal health-related genes induced by S. Reading in turkey cecal tonsil.

Transcriptional comparison of cynomolgus macaques (Macaca fascicularis) following infection with Lassa or Lujo viruses

Project description:Microarray analysis of PBMC from cynomolgus macaques collected longitudinally over the course of infection with Lassa-Josiah, Lassa-Z132, Lassa-SorombaR, or Lujo viruses (n=3 animals/infection condition). 3 macaques from each group were infected intramuscularly with 10^4 PFU of Lassa-Josiah, Lassa-Z132, Lassa-SorombaR, or Lujo viruses. PBMC were collected at days 1, 4, 7, 10, 13, and 29 (for surviving animals). We performed microarray analysis on PBMC samples using Agilent rhesus macaque arrays on all samples, as well as on PBMC from 3 uninfected animals for use as a control.

Transcriptomic profiles of porcine PBMC from nonimmune and ASFV-immune pigs upon in vitro stimulation

Project description:We describe the transcriptomic recall response to African swine fever virus (ASFV) upon in vitro stimulation in porcine peripheral blood mononuclear cells (PBMC) obtained three weeks after receiving an intranasal dose of 106 pfu of the live attenuated BA71ΔCD2 vaccine prototype. PBMC from 4 unvaccinated and 4 vaccinated pigs were stimulated with BA71ΔCD2 or left untreated for 10 hours, RNA was extracted, and bulk RNA-sequencing was performed. Major transcriptional changes were found in BA71ΔCD2-stimulated samples from vaccinated pigs. Indeed, compared to unvaccinated control pigs, cells from vaccinated animals showed an upregulation of genes corresponding to a robust Th1 response. Together with this adaptive immune response, we also distinguished an innate immune response characterized by the expression of macrophage-related inflammatory genes. Altogether, our results demonstrate that intranasal vaccination of pigs with BA71ΔCD2 induce a systemic Th1 memory response that is concomitant with a rapid enhancement of innate immunity upon in vitro activation.
