Project description:During myogenic differentiation the architecture and proteome of muscle stem cells undergo extensive remodelling. These processes are only partially understood and display impairments in age and disease. Using mass spectrometry to analyse protein dynamics during myogenic differentiation, we identified the actin nucleator Leiomodin 1 (LMOD1) increasing in abundance in early phases of myogenic differentiation. Knockdown of LMOD1 in primary myoblasts severely affected myogenic differentiation and fusion of myotubes, while overexpression had the opposite effect. Mechanistically, we show that LMOD1 interacts with Sirtuin1 (SIRT1) and that both follow similar changes in nuclear/cytoplasmic localization during differentiation. We demonstrate that LMOD1 influences SIRT1 localization and the expression of a subset of its target genes. Consistently, depletion or pharmacological inhibition of SIRT1 partially rescues the effects of LMOD1 knockdown. Our work identifies a novel regulator of myogenic differentiation that might be a potential target to improve muscle regeneration in aging and disease.

Project description:While cancer-associated fibroblasts (CAFs) have been historically considered pro-tumorigenic, several studies found that myofibroblast-like CAFs (myCAFs) may have tumor-restraining properties. In our previous study (PMID: 37863888), we performed multi-regional bulk transcriptomics profiling of 8 ER+/PR+/HER2- breast tumors and matched peri-tumoral tissues. We identified an adipose-enriched peri-tumoral subtype which was significantly associated with poorer overall survival in breast cancer patients, in contrast to a myofibroblast-enriched subtype. Here, we performed single-nucleus RNA-sequencing of 2 ER+/PR+/HER2- invasive breast carcinomas and their matched peri-tumoral tissues (1-7 cm from tumor margins), as well as 1 reduction mammoplasty (RM) sample. We identified tumor-induced changes in the peri-tumoral adipocytes compared to RM adipocytes, as well as a high abundance of myCAFs in the larger tumor, whereas a distinct myofibroblast-like subpopulation was found in the peri-tumoral tissues of the smaller tumor. Together with deconvolution analyses of TCGA data and 3D co-culture experiments, we show that that myofibroblasts have both tumor-promoting or -restraining roles depending on their location within or surrounding the tumor.

The main goal is to identify the similarities of zMNC myogenic differentiation with that of mammalian myoblast differentiation. Critical time points were used to identify a switch from the activity of cell proliferation genes to myogenic structural genes. 15-20 adult zebrafish dorsal skeletal muscles were isolated at each of 6 distinct time points (day 0, day 1, day 4, day 7, day 10, day 14) in replicates.

SNAIL is a key regulator of rhabdomyosarcoma progression and myogenic differentiation

Project description:Rhabdomyosarcoma (RMS) is a frequent non-epithelial tumor of soft tissue that originates from a myogenic differentiation defect. Expression of SNAIL transcription factor is elevated in the alveolar subtype of RMS, characterized by a low myogenic differentiation status and high aggressiveness. SNAIL affects RMS metastasis by reorganization of actin cytoskeleton, regulation of ezrin expression and chemotaxis to HGF and SDF-1. The differentiation of human RMS diminishes SNAIL level. SNAIL silencing completely abolishes the growth of human RMS xenotransplants. SNAIL inhibits myogenic differentiation of RMS by binding to the MYF5 promoter, suppressing its expression, displacing MYOD from canonical to alternative E-box sequences and regulating myomiRs expression. SNAIL silencing allows the re-expression of MYF5 and canonical MYOD binding, promoting RMS cell myogenic differentiation. These novel results open potential avenues for the development of innovative therapeutic strategies based on SNAIL silencing.
